It is generally recognized that INAH (isonicotinic acid hydrazide) competes with vitamin B_6. We investigated on this problem using the activities of several enzymes, such as GOT (glutamic oxaloacetic transaminase) and asmine oxidases, which contain pyridoxal phosphate as cofactor, as objects to be studied. INAH blocks liver GOT activity both in vivo and in vitro, and simultaneously it is presumed that INAH damages liver cells and lets GOT flow out of liver into blood stream. Amine oxidase activities in liver and in brain were not influenced by the addition of INAH in vitro. Ribonucleic acid in liver increases after the administration of pyridoxine and decreases after the administration of INAH. Furthermore, pyridoxine reduces the toxicity of INAH against animals.

A congenital disorder in folic acid metabolism on a female child, aged 8 months, was presented. Clinical characteristics were round face, obesity, retardation of mental and physical development, and a slight tendency to hypersegmentation of neutrophils in the peripheral blood. Biochemically, an abnormally higher level of serum folic acid activity and an excessive excretion of formiminoglutamic acid into urine following histidine loading were confirmed. These abnormalities were considered to be due to a marked decrease in formiminotransferase activity which was demonstrated in the liver, biopsied from the patient. This defect in formiminotransferase in the liver may be of hereditary origin. A term "hyperfolic-acidemia with formiminoglutamic-aciduria following histidine loading" may be suggested for this entity of inborn error of folic acid metabolism.

At the microbioassay of vitamin B group (vitamin B_<12>, pantothenic acid, folic acid and vitamin B_6) in formula feed, the influence of feed additives (arsanilic acid, furazolidone, sulfaquinoxaline and arsenobenzene) on the assay values were studied. As the results, it was observed that those influences on each assay microorganism varied according to the kinds of additives. But, it was found that there was no influence of additives on the routine microbioassay of vitamin B group in formula feed.

Data presented in the previous paper have demonstrated that in oral administration O, S-bis (ethoxycarbonyl) thiamine (DCET) showed higher blood thiamine levels for a longer time than thiamine hydrochloride. The present investigation was undertaken to seek some compounds possessing a thiamine activity corresponding or more excellent to DCET. Nine S-mono- and O, S-bis-substituted thioalkoxycarbonyl- or thioalkoxythiocarbonyl thiamine derivatives (I〜IX), S-allyloxycarbonyl-O-tetrahydropyranyl-and S-thioallyloxycarbonyl-O-tetrahydropyranyl thiamine (X, XI) were synthesised by the methods analogous to those described previously. It was shown previously that in S-thioalkoxycarbonyl- or S-thioalkoxythiocarbonyl thiamines S→O rearrangement took place either in alkaline or in acidic medium, similarly as in the case of S-alkoxycarbonylthiamines. Based on this method, any substituted O, S-bis-substituted thiamine derivatives could be obtained easily. Some of these compounds synthesised when administered orally showed fairly high blood thiamine levels for a long time, corresponding to that of DCET.

The method described for the estimation of squalene involves chromatographic separation of the dehydrogenated reaction products of squalene with N-bromosuccinimide into several parts of DS 1〜6 (adsorbent : lime, developer of chromatograms : 10〜20% acetone in hexane), followed by spectrophotometric determination of DS 4 at 395 mμ. This method would be effective particularly for estimating squalene of natural source in the presence of other unsaturated polyenes. The configurations of DS l, 2 and 3 were assigned tentatively to be all-trans-, 10 or 14-monocis-hexamethyltetracosaundecaene and all-trans-hexamethyltetracosanonaene respectively.

CoQ, which is one of cafactors of the terminal oxidation system in liver cells, was measured by spectrophotometric method of Morton et al., using column chromatography. Acute CCl_4 poisoning caused a significant decrease of CoQ content of fresh liver in rats, whereas such decrease could not be found in chronic CCl_4 poisoning. In the intracellular distribution of CoQ, it is localized mainly in the mitochondrial fraction. The amounts of CoQ in the mitochondria of acute and chronic CCl_4 poisoned livers remarkably decreased compared with normal liver. It was found that rat liver contains mainly CoQ_9,although small amount of CoQ_7,CoQ_8 and CoQ_<10> are detected by paperchromatography in the mitochondrial fraction. Hepatic content of CoQ in cadavers had no difference between sex, young and old nor with and without liver diseases.

The enzyme obtained from a cell-free extract of baker's yeast is able to form thiamine diphosphate (TDP) from thiamine monophosphate (TMP) with ATP and Mg^<2+> as well as from thiamine. The enzyme which synthesize TDP from TMP was purified with heat treatment. ammonium sulfate fractionation and DEAE-cellulose column chromatography. The obtained enzyme was partially purified 18 fold in a specific activity of TDP formation from TMP, accompanying the thiaminokinase activity which was not able to separate and also containing a little TMPase activity. In order to exclude the participation of TMPase in TDP formation from TMP, a chemically synthesized TMP-^<32>P was used as a substrate and TDP-^<32>P formed enzymatically was isolated from the reaction mixture. The consistency of specific radio-activities of the TMP-^<32>P used and the formed TDP-^<32>P suggests the direct phosphorylation of TMP to form TDP, that is an exsistence of TMP-phosphokinase.

Schlenk et al. reported a convinient production method of S-adenosylmethionine by incubation of yeast in a glucose-salt medium containtng methionine. In this paper the authors studied on the possibility of the formation of DBCC by incubation of yeast in a glucose-salt medium containing hydroxocobalamin with or without methionine, expecting the incorporation of deoxyadenosyl group to the corrin ring in the same manner as in the S-adenosylmethionine formation. Against the expectation the formation of DBCC and the change of the amount of S-adenosylmethionine produced were not observed in the presence of hydroxocobalamin. However unidentified light- and CN^--labile vitamin B_<12>-group compounds were isolated from the incubation supernatant by DEAE-cellulose chromatography. The absorption spectra of these compounds were similar to those of cobamide coenzymes, but they did not exhibit coenzyme activity in Abeles ・ Lee's system. In paperionophoresis one of them showed ananologous behaviors to those of DBCC and the another behaved similarly as vitamin B_<12> coenzyme M of Takeyama and Buchanan. Based on the result of our other experiment in which the methyl analogue of cobamide coenzyme was synthesized from S-adenosylmethionine or S-me-thylmethionine and hydroxocobalamin after reduction with NaBH_4,the formation mechanism of the unidentified compounds, possible relationship to vitamin B_<12> coenzyme M and the hypothetical role in the methionine synthesis were discussed.
