ジャーナル フリーThe studies on the reactivity of thiazolium ylide of thiamine were carried out and the following new informations were obtained. 1. The nucleophilic reaction of thiazolium ylide of thiamine with aldehydes afforded not only the known HET type products but also the unknown new keto type products under the hydrophobic conditions. It was also investigated that the reactions of thiazolium ylide of thiamine with α-ketoaldehydes, dialkylacylphosphonates, dialkylhydrogenphosphonates, isocyanates and isothiocyanates produced the various type of adducts most of which provided the interesting reaction behaviours respectively. 2. The electrophilic reaction of thiazole-C_2-carbon of thiamine with cyclic secondary amines afforded 1 : 1 adduct. 3. Reaction of thiamine sodium salt with carbondioxide or the treatment of thiamine chloride salt with organic bases afforded thiamine free base as stable crystals which reacted with some electrophiles and ethyl chloroformate to give interesting unusual products respectively. 4. It became evident that there were marked differences of pyrimidine-C_4-substituted thiamine analogues, in the reaction with diethylbenzoylphosphonate, but we could not obtain the definite conclusion about the role of pyrimidine-C_4-NH_2 group of thiamine on the coenzymatic activity of thiamine from our experimental data mentioned above.抄録全体を表示PDF形式でダウンロード (845K) -

ジャーナル フリーA nekoflavin-like substance was isolated from cat liver. The substance was not distinguished from nekoflavin by thin layer chromatography, gel chromatography, ultraviolet and visible absorption spectra, fluorescence spectrum, pH-fluorescence curve, and periodic acid oxidation. It was not decomposed by treatments with 2N HCl (100℃, 2h), 0.05N NaOH (room temperature, overnight), and alkaline phosphatase. However, it was photodecomposed to lumiflavin (in alkaline) and lumichrome (in acidic medium) which were identified by thin layer chromatography, visible absorption spectrum, and pH-fluorescence curve. ジャーナル フリーNineteen foods were examined for loss of folic acid during boiling. Leaching out into the water was also examined. Free folate contents diminished to 10-50% after boiling for 5 minutes. Fifteen-50% was present in the boiling water and so 20-70% of free folate was destroyed in 5 minutes. Free folate remained after 15 minutes was only 5-10% and that remaining in the boiling water was 1-10%. 20-90% of free folate was destroyed in 15 minutes. Total folate contents diminished to the level of 20-90% by boiling for 5 minutes. Considering the folate present in the water, the loss of folate by destruction in 5 minutes was about 30%. By boiling for 15 minutes, 20-40% of folate remained and 40-60% of total folate was destroyed. Pteroylglutamic acid was not destroyed by boiling for 15 minutes. ジャーナル フリーThe biosynthesis of 6-methylmercaptopurinylcobamide (MPC) and its coenzyme from using the resting cells of P. arabinosum, their physicochemical properties and biological as well as biochemical activities are dealt with. MPC obtained by incubation of Factor B and 6-methylmercaptopurine with the resting cells of P. arabinosum was 13% of the total corrinoids formed. The UV-visible absorption spectra of MPC and MPC coenzyme showed absorption maxima at 295,361,515 and 545 mμ and at 261,295,375 and 458 mμ, respectively. Microbiological activities of MPC were 53% and 7% of those of cyanocobalamin for Escherichia coli 215 and Ochromonas malhamensis, respectively. It is also interesting that MPC coenzyme has about one twentieth activity of cobalamin coenzyme in enzymatic 1,2-propanediol dehydration.抄録全体を表示PDF形式でダウンロード (520K) -

ジャーナル フリーThiamine monophosphate pyrophosphorylase (EC 2.5.1.3) has been purified approximately 320-fold from baker's yeast, and the kinetic studies were carried out in the presence of several organic phosphate compounds and metabolites. High-energy phosphate compounds including ATP, ADP acetyl phosphate, phosphoenolpyruvate inhibited the enzyme activity at the physiological concentrations in yeast. The effectors were noncompetitive with respect to both substrates and caused no change in the apparent Km for both substrates with a decrease in V_<max>. A shift of pH optimum to the alkaline side was observed in the presence of the effectors and the enzyme tended to be desensitized to the effectors at pH 9.0,with partly loss of enzyme activity. ジャーナル フリーWhen DL-penicillamine was added to basal diet, immediate weight loss occured. The DL-penicillamine administered rats were frequently followed by appearance of running fits and acrodynia-like symptoms. However, the growth inhibition and these symptoms could be reversed by injection of pyridoxine. When D-penicillamine was added to the basal diet, no significant growth inhibition and symoptoms occured. Phosphatidylcholine in liver were more significantly found in microsome fraction of liver of DL-penicillamine treated rats.
