Lipoic acid-dependent and CoA- and NAD-linked oxidative decarboxylation of pyruvate and 2-oxoglutarate is the main pathway in mammals and proceeds via decarboxylation, acyl generation, acyl transfer and electron transfer.reaction sequences by the participation of several enzymes. In addition to CoA and NAD, TDP, a bivalent metal ion (Mg^<2+> or Ca^<2+>), protein-bound lipoic acid, and FAD are essential. Enzyme systems that catalyze this reaction have been isolated from pig heart Keilin-Hartree preparation as soluble and well-organized multienzyme complexes with molecular weights in the millions. These two complexes have been dissociated into their constituent enzymes, α-keto acid dehydrogenase, lipoate acyltransferase and lipoamide dehydrogenase, and have been reconstituted, both functionally and structurally, from their isolated component enzymes. The basic geometry of these complexes has been investigated from correlated biochemical and electron microscopic studies. Regulatory properties of both complexes are also discussed. The comparative analysis of fatty acid composition of red blood cell membranes obtained from young (6 months) and old (18 months) albino rats showed that the decrease of palmitic acid and the increase of stearic and polyunsaturated fatty acids (carbon number> 20) were demonstrated with aging. In order to separate the blood cells into fresh and aged ones, the centrifugal fractionation was applied upon the blood obtained from individual animal. It was found that the membrane lipids of aged cells on the fatty acid pattern in comparison with young cells showed the reduced palmitic acid and increased stearic and polyunsaturated fatty acids as shown in the blood cells of aged animals. Comparative studies of fatty acid pattern on the same aged cell fractions between young and old animals showed that the age-dependent changes on the cell level were more intensified in old animals. It suggests that the fatty acid composition of red blood cell membrane of old animals is different from that of young animals at the time when new-born cells are produced. The requirement for succinate in a thiamine-1ess mutant strain of E. coli, 70-23,was substituted by both lysine and methionine, although there was a considerable lag before growth occurred with these substances. About one tenth as much amount of thiamine was formed in the mutant cells grown anaerobically with succinate as compared with the wild-type cells grown under the same conditions. This might account for the fact that suspensions of the organism grown on succinate oxidized pyruvate very slowly but did so quickly upon the addition of thiamine. Quantitative studies indicated that succinate-dependent growth was markedly stimulated by thiazole moiety of thiamine. Furthermore, thiazole moiety alone was able to support very slow growth of the mutant under anaerobic conditions, while aerobically the growth did not occur even in the presence of pyrimidine moiety. It was already pointed out that a large amount of pyrimidine was accumulated in the culture when the organism was grown with a limited amount of thiamine. Therefore, it followed that in the mutant strain thiamine might not be formed until the considerable accumulation of intracellular metabolites including succinate, pyrimidine and thiazole moieties had occurred. The genus has been identified both with fumarate production, and rhizoid formation. Tachibana (1961) found that many strains of the genus Rhizopus, especially such high-acid-yield strains as R. nigricans and R. javanicus produced riboflavin cyclic 2', 5'-monophosphate (RCP) with GTP-dependent enzyme. The RCP forming activities of R. oligosporus and R. chinensis were so lower as a few percents of the activity of R. javaleicus. The formation of rhizoid of R. oligosporus during slide culture was observed 5 days later than that of R. javanicus. The growth of R. oligosporus and R. chinensis on the synthetic media containing 1% CaCO_3 which are favorable both to the growth and to the acid production for R. javanicus, was very poor. It was suggested that some relationship between the function of RCP and the formation of rhizoid, besides the acid production, would exist. (Received November 4,1970)

The influence of nicotinic acid (NiA) deficiency was investigated upon gastric acidity, peptic activity, intrinsic factor (IF) activity and vitamin B_<12> binding capacity of gastric juice and gastric mucosa of rats on NiA deficient diet. After forty days on the deficient diet, the amount of gastric acid in the gastric juice was higher than that of the control rats. Matching to this phenomenon, both pH values of the gastric juice and pH values on the gastric wall were lower. IF activity and vitamin B_<12> binding capacity of the gastric mucosa were lower and, on the contrary, both the activity and capacity were higher in the gastric juice, in the NiA deficient rats than those in the control rats. Histologic findings showed damage, atrophy, pycnosis and necrosis in parietal cells and chief cells in the NiA deficient rats. These facts suggest that the membrane of the secreting cells of the stomach was so fragile owing to NiA deficiency to have had damaged or broken during the collection of gastric juice and the contents of these cells leaked into the gastric juice. Thiamine, when incubated with Lactobacillus fermenti hsarvested from thiamine-poor broth, was quickly uptaken by cells; time course of increase of both the total and free thiamine in cells were similar and phosphorylated thiamine was found to be predominant over free thiamine. Thiamine-uptake by Lactobacilli cells, when pyrithiamine or Amprolium was added, was more or less inhibited; pyrithiamine showed not only strong inhibition of thiamine-uptake but also decrease of phosphorylated thiamine, while Amprolium had weak inhibition of thiamine-uptake with negligible effect to phosphorylation of thiamine. Lactobacilli cells, however, when preincubated with pyrithiamine or Amprolium, were demonstrated to uptake thiamine on incubation with thiamine to the same extent as cells without preincubation with antithiamine compounds; therefore competitive inhibition of thiamine-uptake by cells was proved only when thiamine and pyrithiamine (or Amprolium) were added to the uptake process. Competitive uptake of thiamine and pyrithiamine (or Amprolium) by cells on incubation was also demonstrated by chemical determination of thiamine and antithiamine compounds. (Received Nov. 24,1970)

Thiamine diphosphate(TDP), when incubated with Lactobacillus fermenti harvested from thiamine-poor broth, was more effectively uptaken by cells than thiamine. Practically no free thiamine was found in cells during the process of thiamine-uptake. Glucose is essential for thiamine-uptake which was proved to be active transport, because typical inhibition by DNP or NaF but no inhibition by ouabain had been shown. Pyrithiamine or Amprolium showed stronger inhibition against uptake of TDP by cells than that of thiamine. When ^<32>P-TDP was incubated with Lactobacilli cells, uptaken ^<32>P was found to coincide with uptaken thiamine. As such phosphatase inhibitors as molybdate and phosphate had no inhibition to uptake of TDP by cells. TDP seems to be uptaken by Lactobacilli as TDP de novo, not as free thiamine after hydrolysis by phosphatase. (Received Nov. 24,1970)

It has been reported that some vitamins besides their function as coenzyme regulate the enzyme activity by induction (coenzyme induction) or repression (coenzyme repression) of the enzyme proteins. In this paper, the activity changes of pyruvate and 2-oxoglutarate dehydrogenases with thiamine contents were examined by using Escherichia coli. When a Hfr strain (KG 33) derived from E. coli K 12,auxotrophic for thiazole was grown overnight in Davis medium containing minimal concentration (10-^8M) of thiamine for growth, bacterial thiamine content of the mutants was about one fifth of that of the parent and its α-keto acid dehydrogenase activities were fairly low as compared with that of the parent. The addition of thiamine diphosphate to a reaction mixture of the enzyme extracted from the mutant restored the dehydrogenase activity to the extent of the enzyme extracted from the parent or the mutant grown with excess thiamine (10-^6M). Similar results were obtained with a mutant (KG 674) of E. coli K 12,auxotrophic for thiamine. Elevation of thiamine contents in E. coli K 12 by adding thiamine (10-^6M) or phenylalanine (10-^2M) in the growth medium caused no increase of the dehydrogenase activity as compared with the bacteria grown without adding the compounds. These results suggested that the synthesis of apoenzymes was neither impaired by thiamine depletion nor induced by excess thiamine in the cells. Extensive dialysis of the enzyme extract from the mutant grown on thiamine minimum caused to decrease the dehydrogenase activity without change of thiamine content, while the same treatment of the enzyme extract from the mutant grown on excess thiamine had no effect on enzyme activity but decrease on thiamine content in the extract.
