    The diets added with 30,60,150 and 300 mg of carnithine per kg were given for six weeks, to male White Leghorn chicks of seven days old. The basal diet consisted of ground corn 35-38%, soybean oil meal 6-10%, fish meal 7-10%, minerals, liver oil and vitamin B mixture. During six weeks trials, carnithine improved the growth of chicks and the body weight increases over control without the addition of carnithine, were 4,16,13 and 12-23% in respective added carnithine amount. On the end of the trials, all chicks were killed and the compositions of the leg muscle and liver were compared, but no difference was found between the chicks with or without the addition of carnithine.

    A new medium containing no sugar was invented for the mixed culture of Clostridium thiaminolyticum and Lactobacillus bifidus. Clostridium and Lactobacillus grew well without morphorogical change in this medium, as in their suitable medium, on the separated cultures. On mixed culture of the both bacteria in the new medium, Clostridium grew well as in the separated culture, while the growth of Lactbacillus was suppressed a little. On the separated culture, final pHs of the culture of Clostridium and Lactobacillus were 7.2 and 4.8,respectively. In the case of mixed culture of the two, almost median value of pH 7.2 and 4.8 was maintained during the cultivation. Thiaminase activity of Clostridium in the mixed culture decreased about to one half of that in separated culture, while the cell number did not differ in mixed or separated culture within 4 to 5 days. Thus it seems that the growth of Clostridium thiaminolyticum, as well as Bacillus thiaminolyticus or B. aneurinolyticus, is not suppressed by the presence of Lactobacillus bifidus.

    Already, many investigations on the thiamine decomposing bacteria have been made, but except several points, the elucidation of their raison detre in living body is not completely known. Consequently, as one of the important research subjects on the thiamine decomposing bacteria, the symbiotic tests by mixed culture and animal experiments with other intestinal microorganisms were undertaken. The results of the experiments, using Lactobacillus bifidus or Lactobacillus acidophilus on one hand, and Bacillus thiaminolyticus or Bacillus aneurinolyticus on the other, indicate that Lactobacilli have almost no influence on the growth of aerobic thiaminase bacteria in vitro and in vivo as well as on the thiaminase production of the B. thiaminolyticus.

    It has been found that thiamine-protein complexes and unbound disulfide-type thiamines are produced by the reaction of thiamine or O-benzoyl thiamine (OBT) with iodine oxidized egg albumin at pH 7.3 or 9.0,37℃ for 60 minutes. The complexes were not separated into their components by means of zone-electrophoresis, dialysis and metaphosphoric acid deproteinization, but free thiamine was liberated by the reduction with sodium thiosulfate or cysteine. From these results, the complexes were comfirmed to be protein-thiamine mixed disulfides which were produced by the reaction of thiol-type thiamine with S-S group of the oxidized protein. By the reaction at pH 9.0 larger amounts of the combined thiamines were produced than those at pH 7.3. The result was considered due to a higher concentration of the thiol-type thiamines at pH 9.0. OBT showed a higher reactivity with the oxidized protein than that of thiamine. An optimal amout of iodine for the oxidation of protein was about 5 molar-equivalents of the protein.

    In a continued study on the interaction of thiol-type thiamines and oxidized egg albumin, the relationship between reaction pHs and produced amounts of the protein combined thiamines has been investigated. In the case of thiamine, the combined thiamine was obesrved at pH about 7.0,and rapidly increased at pHs above 8.2,showing maximum value at pHs above 10.0. Since this pH curve is correlated with the relation between pHs and the equilibrium concentration of the thiol-type thiamine, it can be deduced that the produced amounts of the combined thiamine are proportional to the concentration of the thiol-type thiamine in the reaction mixture. In the case of O-benzoylthiamine (OBT), the combination occurred even at pHs about 6.0,and higher amounts of combined thiamines were always observed at each pH than those in case of thiamine. The equilibrium concentrations of the thiol-type of thiamine and OBT in weakly alkaline medium were compared measuring anodic wave heights of polarography. The concentration of the thiol-type was found much higher in OBT than in thiamine.

    In a continued study on the interaction of the thiol-type of thiamine or O-benzoylthiamine and iodine-oxidized egg albumin, effects of denaturation on protein combined thiamines have been investigated. The largest amounts of the combined thiamines were produced when the oxidized protein prepared by the oxidation of native egg albumin was used in the reaction with the thiol-type thiamines. On the other hand, urea or heat denaturation before the oxidation caused a marked decrease of the produced amounts of the combined thiamines, however the denaturation after the oxidation had almost no effect. From these results, it was suggested that the denaturation before the oxidation had inhibitory action on the formation of protein intramolecular-S-S-linkages which took part in the reaction with the thiol-type thiamines.

    The existence of two forms of GOT was first shown enzymatically by Katunuma et al. Partially purification and dissimilar properties of mitochondrial GOT (GOT_M) and soluble fraction GOT (GOT_S) have been reported by the previous paper from our laboratory. The present paper describes the procedure of further purification and distinct characteristics of the enzymes. The use of (NH_4)_2SO_4 fractionation, heat treatment and chromatography on DEAE-cellulose effected both about 100-fold purification of GOT_M and GOT_S. Purified preparations obtained were homogeneous column chromatographycally, electrophoretically and ultracentrifugal analytically. Compared to GOT_S, 5.8S, GOT_M had a smaller S_<20>W 3.8S. The purified enzymes also differd in other physicochemical natures. GOT was retained and GOT unretained by DEAE-cellulose, whereas the reverse seemed to be true of the IRC-50. On zone electrophoresis, GOT_M moved to anode (-0.4 cm/hr) and GOT_S to cathode (+0.05 cm/hr) at pH 8 in veronal buffer. GOT_M (GOT_S) gave the following Michaelis constant : for aspartic acid, 1.8×10^<-3> (8.0×10^<-3>), for α-ketoglutaric acid, 5.8×10^<-3> (4.3×10^<-3>). Absorption spectra of the enzyme were recorded and showed typical spectra of vitamin B_6-enzyme without a trace of soret band. Spectrophotometric studies of the enzymes, especially of GOT_M, provided the information that the addition of L-aspartic acid, to pyridoxal phosphate or GOT at pH 5.4 decreased the absorbancy at 430mμ and increased at 340 mμ, showing the possibility of interconversion of the enzyme between pyridoxal phosphate form and pyridoxamine phosphate form even at low pH. Upon addition of L-alanine or L-ornithine, such a shift was not observed.

    Ornithine-δ-transaminase has been purified from rat liver mitochondria about 400 times active more than original mitochondria suspension using the technique of sonication, ammonium sulfate fractionation. heat treatment, DEAE-cellulose column chromatography and zone electrophoresis. The purified enzyme thus obtained is one component on ultracentrifugal pattern and electrophoresis. No activity of GOT and GPT was recognized. Supernatant fraction has not any activity. Sedimentation constant (S_<20>W) was 7.1×10^<-13> and molecular weight was approximately 115,000. It has been concluded that the enzyme is pyridoxal phosphate enzyme from the following data : i.e. the enzyme was strongly inhibited by vitamin B_6-enzyme inhibitors, such as INAH, thiosemicarbazide and hydroxylamine, and the resolved apoenzyme is able to recover its full activity by an addition of pyridoxal phosphate. The properties of the enzyme were as followes : (1) Km for ornithine 6.0×10^<-4>M, for α-ketoglutarate 8.45×10^<-4>M, and for pyridoxal phosphate 1.13×10^<-5>M. (2) Turnover number for α-ketoglutarate 63.66,for glyoxalate 12.02,for pyruvate 1.84,and for oxalacetate 0.46. (3) When oxalacetate was added to α-ketoglutarate-ornithine system, competitive inhibition between α-ketoglutarate and oxalacetate was recognized exactly.
