ジャーナル フリーSporolactobacillus inulinus has been known to require essentially valine, leucine, biotin and calcium pantothenate and secondarily glutamic acid. But, this organism can not grow in the medium containing glucose and minerals in addition to the above mentioned nutrients. In an attempt to prepare the minimum growth medium, effects of amino acids and vitamins on the growth of the bacteria were investigated in details. A minute quantity of thiamine, 2.5 ng/ml, was known to be essential. The reason of misunderstanding in the previous paper is considered to result from contamination from the natural medium (GYP). Supplementary effect of folic acid or p-aminobenzoic acid (PABA) was also found. Thus the substantial composition of the basal minimum medium was determined as : glucose, sodium acetate, minerals, valine, leucine, glutamic acid, biotin, calcium pantothenate, PABA and thiamine.抄録全体を表示PDF形式でダウンロード (567K) -

ジャーナル フリーDuring the course of study to prepare a minimum growth medium for Sporolactobacillus inulinus, necessity of minute amount of thiamine and stimulatory effect of p-aminobenzoic acid (PABA) were demonstrated. According to the result of this study a basal minimum medium (BMM) was proposed essential requirement of minute amount thiamines stimulated by PABA in the preceding paper of this series. Curiously, however, it was found that when a natural medium (GYP) was used for preculture instead of the synthetic medium, the tested microorganism could not grow in the BMM. This shows that the BMM is not always satisfactory. Hence, the growth-promoting effects of various amino acids were reexamined. The results showed that cysteine was indispensable and, on the contrary, PABA became unnecessary for growth of this organism. Thus the minimum medium for Sporolactobacillus inulinus was finally determined as follows : glucose, sodium acetate, minerals, biotin, calcium pantothenate, thiamine, valine, leucine, glutamic acid and cysteine.抄録全体を表示PDF形式でダウンロード (403K) -

ジャーナル フリーA Method for the determination of ubiquinone homologues (UQ) in serum and liver is described. Serum or liver homogenate was saponified with methanolic alkaline solution after addition of 2,3,6-trimethyl-5-nonaprenyl-1,4-benzoquinone (TQ-9) as an internal standard and then extracted with n-hexane. The n-hexane extract was evaporated under N_2 gas at 30℃, and the residue was dissolved in dioxane. The sample solution was applied to the high-speed liquid chromatography (HSLC) using a Permaphase ODS column and ethanol・water (80 : 20) as a mobile phase. The peaks of UQ and TQ-9 were not disturbed by other fat-soluble substances when an on-line high-sensitive multiple wavelength detector set at 275 nm (absorption maximum of UQ) was used as a detector. The proposed HSLC method was found to be more simple and specific than the other conventional determination methods.抄録全体を表示PDF形式でダウンロード (578K) -

ジャーナル フリーThe reaction of ubiquinone (UQ) with ethylcyanoacetate (ECA) to obtain a fluorescent substance and the application of the reaction to the quantitative determination of UQ in serum by high-speed liquid chromatography (HSLC) were studied. The most sensitive and quantitative fluorescence was obtained when UQ was reacted with ECA at 50℃ for 20 hours. Since the fluorescent substance was unstable to exposure of light, the reaction should be performed in a brown tube. Serum was extracted with n-hexane in the presence of ethanol and the extract was evaporated under N_2 gas at 30℃. The residue was reacted with ECA reagent and then applied to the HSLC using Permaphase ODS column and ethanol・water (75 : 25) as a mobile phase. The peak was detected with a spectrofluorometer (Ex. 430 nm, Em. 530 nm). The proposed method was not affected by other fat-soluble substances and more specific and simple than the other conventional determination methods.抄録全体を表示PDF形式でダウンロード (593K) -
