In order to study on the effects of antibiotics upon the riboflavin metabolism, the change of riboflavin level in blood caused by the administration of antibiotics was observed in patients of liver and kidney deseases and in normal subjects. A significant decrease of the amount of total and esterified riboflavin and the ester ratio in blood was observed in the patients. In all cases, a temporary decrease of riboflavin content of blood occurred by the administration of antibiotics. The effect of antibiotics was more remarkable on patients than on normal subjects. Even if antibiotics was administered, clinical signs of riboflavin deficiency were scarcely detected. However, there may be some conditions, being susceptible to the deficiency in the patients.

Effects of administration of ATP or co-administration with cocarboxylase and α-lipoic acid were examined on 104 patients who showed abnormal ST・T and arrythmia on electrocardiogram. Remarkable effects were exerted on coronary insuffciency or premature beats observed on electrocardiogram in patients of arteriosclerosis, essential hypertension, renal and endocrinological disease. However, no effective influence was observed on patients of myocardial infarction, auricular fibrillation, paroxysmal tachycardia and cardiac valuvlar diseases. It was found that a continuous administration was more effective than a single dosage of these cofactors and co-administration exerted more remarkable effects on these symptoms than a single use of ATP.

The experiments were made on the adrenal hypertrophy in thiamine deficiency. In thiamine deficient rats, a significant increase of weight of the adrenal gland due to the hypertrophy of the adrenal cortex was observed. The symptom was restrained by administration of cortisone, while it was not affected by DOCA. ACTH, however, showed a promoting effect on the symptom. In histopathological and histochemical studies, a hyperfunction at the fasciculated zone of the adrenal cortex was observed at the early phase of thiamine deficiency, while at the later phase a functional depression was observed. These findings indicate that a stress caused by the deficiency exerts a certain effect on the pituitary-adrenocortical system.

Riboflavin-5'-monosulfate injected to animal can be analyzed by the method using both lumiflavin fluorescence method and paper chromatography. For the separation of riboflavin-5'-monosulfate, FAD, FMN and riboflavin by paper chromatography, the mixture of n-butanol・acetone・glacial acetic acid・water (5 : 2 : 1 : 3,v/v) was recommended as a suitable mobile phase.

Thirty minutes after the injection of 500μg of riboflavin-5'-monosulfate to a rat, riboflavin-5'-monosulfate was proved to be emigrated into the liver, heart, kidney and intestine. However, riboflavin-5'-monosulfate could not be easily exchanged for physiological flavin compounds, i. e., FAD, FMN or riboflavin. Injected riboflavin-5'-monosulfate was rapidly excreted into urine.

The effect of FMN and FAD on the cramp of rat caused by the injection of phenol was studied by the measurement of time of duration of the cramp. The results, examined by statistical treatment, indicated that either FMN or FAD acts so as to diminish the cramp.

Estimation of α-lipoic acid in urine and serum of patients of liver diseases, diabetes mellitus and polyneuritis was carried out The α-lipoic acid levels were low as compared with those of the normal subject, especially in patients of liver cirrhosis. This result indicates that there may be α-lipoic acid depletion in patients of liver cirrhosis.

Studies were made, using rats, on the urinary excretion of α-lipoic acid and its content of the liver in such conditions as acute or chronic CCl_4 poisoning, alloxan diabetes and thiamine deficiency. Chronic CCl_4 poisoning and thiamine deficiency caused a remarkable decrease of the urinary excretion of α-lipoic acid, while in alloxan diabetes the excretion rather increased. The hepatic content of α-lipoic acid in rats in all experimental conditions was lower than that in the normal rats. However, the decrease of the hepatic content caused by acute CCl_4,poisoning was not so much remarkable.

The rate of increase of pyruvate and α-ketoglutarate in urine excreted for the first one hour and the next one hour after an intravenous administration of 20ml of 20% fructose solution was higher in patients of liver disease, diabetes mellitus and polyneuritis than in the normal subjects. The acetylating activity after an oral administration of 500 mg p-aminobenzoic acid was lower in these patients than in the normal subjects. An administration of 10 mg α-lipoic acid improved the distubance of the α-keto acid metabolism and acetylation in these patients.

Estimation of thiamine and riboflavin contents of the whole body of the sucking mouse was carried out after birth. Increase of riboflavin in the whole body was proportional to the gain in body weight, while thiamine increased rapidly up to a definite level and afterwards the increase was very slow.

Young and adult rats of about 40g and 150g, respectively were fed on a riboflavin deficient diet and weight gain, urinary excretion of the vitamin, thiamine and riboflavin contents of blood and organs and tissue respiration of the organs were estimated for 7 weeks. It was found from these experiments that young rats was more susceptible to the deficiency in comparison with adult animals. In young rats the decrease of riboflavin content caused by the deficiency was most remarkable in muscle followed by the liver, kidney, heart and brain in the order, while the corresponding order was the liver, muscle, kidney, heart and brain in adult rats. The amount of riboflavin in lung was found in both groups to be not affected by the deficiency. The amount of riboflavin excreted in urine decreased remarkably at the beginning of the deficient feeding. After several days, its amount reached to an extremely low level. Although the tissue respiration indicated a sharp decrease, in riboflavin deficiency, the amount of thiamine in organs did not so much change.

Rats and mice of about 80g and 12g respectively were fed on a diet containing various amounts of riboflavin for 50 days, and weight gain, urinary excretion and the amounts of the vitamin in organs and tissue respiration of the organs were estimated. When 15 μg of riboflavin was administered daily to rats, riboflavin-deficiency occurred. The maximum riboflavin content of the whole body was observed at the 30th day of a daily administration of more than 400 μg of the vitamin. Even though much more amount of riboflavin was given, the length of time needed for the maximum level could not be shortened. By the administration of riboflavin, the amount of the vitamin in the liver, kidney and heart greatly increased, while in the lung only a slight increase was observed.

Thiamine content of blood was determined and urinary excretion after an administration of the vitamin was estimated in 77 cases of dermatitis, 89 cases of acute and subacute eczema, and 62 cases of chronic eczema. At the same time, blood pyruvic acid levels were also determined as adjunct to the studies on thiamine metabolism. Thiamine metabolism was seldom disturbed in the comparatively simple eczema with localized lesions, as in the dermatitis. On the contrary, a large proportion of the cases of widely spread eczema with a strong tendency to involve the exposed area of the skin, was associated with disturbed thiamine metabolism. It was worthy to note that there was disturbed metabolism of various kinds, such as macrocytic anemia, functional disturbance of the liver and achylia gastrica, along with thiamine deficiency, in these cases.

In view of the fact that the disturbed thiamine metabolism by some types of eczema and dermatitis was often associated with macrocytic anemia, functional disturbance of the liver, and achylia gastrica, studies were carried out on these syndromes. Among 103 cases, mainly of dermatitis and eczema, 16 cases were shown to have macrocytic anemia and 9 cases normocytic anemia. Ten cases of macrocytic anemia, namely 62.5% of the 16 cases showed functional disturbance of the liver, while in 12 cases achylia gastrica was observed. Rabbits developed anemia in the advanced stage of thiamine deficiency, though the symptom was not manifested during the period when the deficiency was in progress. However it was supposed by the determination of mean corpuscular volume and mean corpuscular hemoglobin concentration that the erythrocytes had a tendency to be macrocytic in thiamine deficiency.
