A pure deficiency of thiamine was produced in rats, and their neurological and cardiovascular symptoms, and body temperature were observed by means of electromyograph, electrocardiograph, and thermocouple respectively. These results were contracted with histological finding and changes in various blood components. Similar experiments were carried out with the control group of underfed rats for the purpose of checking the influence of inanition, which always came into question in the experimental thiamine deficiency. Basic general observations were first made on normal, thiamine-deficient, and control underfed rats. In the thiamine-deficient group anorexia and body weight decrease developed from the end of the second week, paralytic and gastrointestinal symptoms (diarrhea) appeared from the fourth week, and the animals died in the fifth week. Oral administration of thiamine in a daily dose of 5 μg in the end stage of thiamine deficiency caused increase in appetite and body weight on the next day, and cured the paralytic and gastrointestinal symptoms in several days. The effect of single injection of thiamine one mg lasted for two weeks. In the control underfed group the change in body weight was similar to that of thiamine-deficient group, but appetite and vitality were good till end. Paralytic and gastrointestinal symptoms were not observed, and almost all animals died in the sixth week.

In thiamine deficient group neuroparalytic symptoms appeared in the fourth week, resulting in abasia in the fifth week. Ataxia was also observed in this period. Oral administration could be cured by for several days. A marked paralysis was observed at the end stage. Neuroparalytic symptoms were not observed in the control underfed group. Electromyographical observations on the thiamine-deficient group showed the appearance of fibrillation and fasciculation in the fourth to fifth week, but not in the control group. Histological examination of the sciatic nerve revealed slight degeneration of axis cilinder on the 27th and 30th day, while no alternation was found in the control group. Histological examination of the gastrocuremic muscle revealed increase of fat and degeneration in the thiamine-deficient group, but no change except disappearance of fat and glycogen in the control group.

Bradycardia increased during the course of thiamine deficiency, leading to heart rate of about 300 in the fifth week. Arrhythmia, increase in the right axis deviation, and changes in ST (elevation or depression) and T (increase or decrease) were found. Thiamine injection to the thiamine-deficient group produced prompt improvement of these electrocardiographical changes, but the injection of vitacamphor or noradrenaline did not. Neostigmine injection to thiamine-deficient group increased bradycardia and arrhythmia, while atropine injection improved arrhythmia. Injection of pyruvic acid to thiamine-deficient group caused elevation of ST, depression of T and slight decrease in heart rate, while injection of lipoic acid following pyruvic acid improved negative T, and increased heart rate. Thiamine-deficient group showed marked increase in hemoglobin and hematocrit, marked decrease in circulatory plasma volume, but no change in serum protein, except slight decrease of albumin in the fourth and fifth week. Constant relation was not found between the change in serum and cardiac muscle electrolytes, and electrocardiographic changes. In thiamine-deficient group marked increase in the heart weight and dilatation of right heart, and increase of fat and degeneration in histological findings were observed, while no significant changes were detected in the control group.

The temperatures of the gastrocunemic muscle, rectum, and skin were determined by means of thermocouple. In control underfed group no changes were detected until the fifth week. In thiamine-deficient group fall of body temperature appeared from the third week, and gradually increased toward the end stage. Fluctuation of skin- and muscle-temperature was observed from the end of the second to the beginning of the fourth week. In the case with marked hypothermia in the end stage of thiamine deficiency, oral administration of thiamine in a daily dose of 5 μg elevated the body temperature from the next day, leading to the normal temperature of the muscle, rectum, and skin in 3 to 5 days. However, fluctuation was observed in 3 to 10 days in the muscle temperature, and in 5 to 10 days in the skin temperature. TTG and DNP injection in thiamine-deficient group caused prominent rise of the body temperature in cases of the 23th day of deficiency. Reduction of glycogen and degeneration was observed histologically in the liver and muscle. The adrenal glands showed maximum increase in weight on 23 to 30th day of thiamine deficiency, and the most marked change of hyperfunction on the 24th day. In 20 out of the 23 hospitalized cases with beriberi a slight fever was found, which was improved by the treatment in 11 cases, but not in 8 cases.

Vitamin B_<12> content of 11 samples of mash of "Takuan-zuke" and 20 samples of "Nukamiso" (fermented paste of rice bran) was determined by Euglena gracilis var. bacillaris. The vitamin B_<12> content of the former was from 32 to 182 mμg per 100 g, and the mean value was 74 mμg per 100 g. The vitamin B_<12> content of the latter varied from 30 to 5040 mμg per 100 g, and the mean value was 700 mμg per 100 g. From vitamin B_<12>-rich "Nukamiso" various strains of yeast, acid forming bacteria and aerobic bacteria were isolated, and their vitamin B_<12> producing ability was examined. It was confirmed that yeasts had slightly vitamin B_<12> producing ability, as 12-28 mμg/dl of broth, however some strains of aerobic bacteria proved to have a strong vitamin B_<12> producing ability, its maximum value being 5.2 μg/dl of broth. On the contrary, lactic bacteria and other acid forming bacteria had no vitamin B_<12> producing ability but rather seemed to consume vitamin B_<12>.

Effects of antibiotics on the growth of lactic bacteria, Lactobacillus arabinosus 17-5,L.casei E, L.fermenti 36 and Streptococcus faecalis R, in a complete or controlling synthetic medium were investigated. Aureomycin inhibited the growth of Streptococcus faecalis R at a concentration of 10μg/ml in the controlling medium, but not at the same concentration in the complete medium. Chloramphenicol stimulated the growth of L.arabinosus 17-5 and L.fermenti 36 at a concentration of 0.5-1.5μg/ml in the controlling medium. Other antibiotics tested did not show such an effect.
