1. BCG Wax D
4 antigen does not react with leprosy or tuberculosis serum without the addition of cardiolipin and choresterol.
2. The antigenic components in this experiments were as follows; 0.05cc. of 0.1% BCG D
4, each 0.2cc. of 0.2% cardiolipin, 1% choresterol, and 0.55cc. of absolute alcohol to make a total of 1cc.

4, Mycobacterium tuberculosis (Aoyama B strain) Wax D
220, and tuberculin polysaccharide Lot
4 fraction, BCG Wax D
4 showed the highest titer of th maximum hemolysis at x128, while the other two showed the titer at x64.
4. Using this amount of antigen, reactions were observed with the serum of leprosy or tuberculosis, but not of pregnacy. Teh reaction was also positvie with the serum of syphilis in about 10%, which was considered as a nonspecific reaction because of the addition of cardiolipin In the antigen.
5. The chemcal structure and nature are are clarified in this antigen, and its antigenic activity is stable.
6. The problem of cross reaction between leprosy and tuberculosis with the use of this antigen is to be eliminated by changing the amount of choresterol and/or by using cephalin in the future.
7. Among 100 cases of leprosy, 84% of lepromatous, 62.5% of macular, and 72.4% of neural leprosy showed positive reaction with the use of this antigen.
