Studies of the chromatographic behavior of vitamin B_<12> on columns of ion exchange resin led the authors to establish a simple method of determination of vitamin B_<12> in vitamin mixtures by using a strongly basic anion exchange resin, IRA-410. The results obtained by this method showed a good accordance with those of microbiological assay. We also confirmed that vitamin B_<12> was not decomposed during the chromatographic procedures of this method.

Riboflavin content was estimated by the lumiflavin method with urine specimens collected from 8 a.m. to noon for successive two days. The subjects examined were divided into two groups : In group 1,40 boys from 8 to 10 years of age with clinical signs of nutritional dystrophy were used. Group 2 consisted of 40 boys with the same range of age without nutritional dystrophy. Riboflavin concentration in the forenoon urine was 2.8±0.7μg% and 8.0±2.0μg% in groups 1 and 2 respectively. The absolute amount of riboflavin in the two forenoon urine was 11.4±2.5μg and 32.0±8.1μg in groups 1 and 2 respectively. It may be concluded that urinary output of riboflavin was lower in the boys with nutritional dystrophy than in those without it.

In order to confirm the mechanism of the reaction of ascorbic acid with benzene diazonium salts, aniline and 13 kinds of its derivatives were examined. It was assumed that the presence of nitro radical in ortho or para position of aniline would be essential for the production of an intensely colored reaction medium, and also that methoxy group in the residual para or ortho position would serve as auxochrome group, while hydroxy and amino groups would not. It was found that p-nitroaniline gave a higher color reaction with a small amount of ascorbic acid (0.5-0.05mg) than that value with 4-methoxy-2-nitroaniline, and a linear relationship between the color intensity and the amount of ascorbic acid could be ascertained within the above meationed range of ascorbic acid, while this method and also 4-methoxy-2-nitroaniline method were interfered by the presence of glutathione, cysteine, glucose, xylose, and tannine in the test solutions.

Active glycerol dichlorohydrine was used and observations were made with a fluorescence microscope. (1) In the retina, especially in the pigment epithelium, vitamin A appeared 3 hours after its administration, but the quantity was not always proportional to the administered doses, no positive result was detected by the glycerol dichlorohydrine method. (2) The amount of vitamin A required for regeneration of rhodopsin, is very small, but vitamin A plays an important role in the maitenance of function recovery of rhodopsin in the pigment epithelia and visual cells. (3) The delay of rhodopsin regeneration of vitamin A-deficient rats is due to functional insufficiency of the pigment erithelia and visual cells, which recover in a few days after administration of vitamin A, rather than to the shortage of vitamin A for direct use in the process of rhodopsin regeneration.

Histochemically active glycerol dichlorohydrine was used and it was observed by a fluorescence-microscope. (1) The intestinal absorption was more ready with tunny liver oil and distilled cod liver oil than with synthetic vitamin A oil, shark liver- and cod liver oil. (2) In cells, some granules of vitamin A were found together, and the others not together, with lipoid. In the former the granules were fine in rats adminstered with natural vitamin A oil, and coarse and various in size in rats with synthetic oil. In the latter, all the granules were minute in size. They were less in number in rats with synthetic vitamin A oil than in those with natural oil. (3) In the adrenal cortex, vitamin A was found in a little larger amount in rats administered with natural vitamin A oil than those with the synthetic one, but no such difference could be seen in the retina. (4) Fat is oxidized and destroyed in the lung. The vegetative oils are more difficult to be utilized than the animal fats. (5) The discharge of vitamin A by the kidney is somewhat more rapid with natural vitamin A oil than with synthetic one.

It has been reported that the ability of excrement for destroying riboflavin seems to be different with the species of animals. This paper deals with the decomposition of riboflavin by feces of rats fed with various kinds of diets. The results obtained suggest that the ability of the feces of rats, fed with animal diets, for destroying riboflavin seems to be stronger than those of rats fed with vegetable diets.

This paper deals with the synthesis of riboflavin by culturing without shaking Escherichia coli communis, Alcaligenes faecalis and Proteus vulgaris at 37℃ for one week in bouillon medium or Anderson's synthetic medium. It was found that Alcaligenes faecalis and Proteus vulgaris cultured in bouillon medium could not synthesize riboflavin at early period, but when cultured in Anderson's medium riboflavin was synthesized at the same period. Synthesis of riboflavin reached its maximum within a few days, but the period of time required varied between the two media.

When 3μg of vitamin B_<12> was injected every other day, its excretion in urine scarcely increased for the first 45 hours, but a marked increase in the 8 hours urine after injection was observed on the 13th day. It seemed that vitamin B_<12> in the body was saturated to a certain degree at the latter stage. In case of intramuscular injection of vitamin B_<12>, an increase in its excretion in urine for the 8 hours after the injection was more marked with a larger dosis (over 20μg than with a smaller dosis (under 20μg). Vitamin B_<12> content in the liver and kidneys increased markedly, by its injection in 8 hours, especially in the liver. It seemed that there was a certain limit in vitamin B_<12> content in the liver and that, when 10μg of the vitamin was administered, the liver was just saturated with the vitamin and the surplus vitamin administered was excreted in urine rapidly in free form.

When streptomycin (10mg) was injected every other day, urinary excretion of vitamin B_<12> for 45 to 46 hours after injection increased considerably for about two weeks, but the administration of chloramphenicol caused a prompt decrease. In the case of injection of both streptomycin (10mg) and vitamin B_<12> (1μg) every other day, the excretion of vitamin B_<12> in the 45 hours urine increased markedly on the 12th-14th day and decreased again on and after the 18th day. When streptomycin was injected intramuscularly every other day for three weeks, a marked increase of vitamin B_<12> content in liver tissue and blood was observed. The results of these experiments suggest changes in the balance of intestinal bacterial flora by the antibiotics.
