From the ethyl acetate soluble fraction of alcoholic extract of Pteridium aquilinum, two flavonoid pigments, astragalin (kaempferol-3-glucoside) and isoquercitrin (quercetin-3-glucoside), were isolated in the form of the yellow and needle crystals, respectively. Moreover the presence of a small quantity of rutin (quercetin-3-rhamnoglucoside) and two unknown flavonoid pigments was proved by the method of the paper chromatography. For studying the thiamine-decomposing activity of the pigments, 10μg of thiamine was incubated with 10μg of several flavonoid pigments at 70° for one hour at pH 7.0 in a total volume of 10ml and the activity was expressed by the quantity of decomposed thiamine. From the results of this experiment, it was proved that the pigments possessing free orthodihydroxyl group in the side phenyl radical such as quercetin and its glycosides had a marked activity. It is therefore presumed that at least a portion of the so-called "thermostable antithiamine factor" of bracken is isoquercitrin and rutin therein.

The thiamine-decomposing activity of a series of flavonoids and phenols was investigated and the flavonoids containing o-diphenol in the side chain as well as pyrocatechol and its derivatives were found generally to have a very potent activity of thiamine decomposition. The glycoside formation at 3-hydroxy group showed varying influences according to the various derivatives.

The thiamine-decomposing thermostable factor (SF) of ferns was isolated in a crystalline form. The absorption spectrum in the ultraviolet region agrees quite well with that of astragalin, but the infrared absorption spectrum showed minor differences in spite of general resemblance. The melting points, elementary analysis and Rf values of paper chromatography were found to be somewhat different. It is therefore concluded that the major part of bracken SF is a flavnnol, akin to astragalin. Further, it was proved by paper chromatography quercetin and isoquercitrin, the compounds which have already been reported by the author to have very potent SF activity.

Changes in the contents of ascorbic, dehydroascorbic and diketogulonic acids in 4 kinds of leafy vegetables, 3 kinds of root vegetables and two parts of a fruit during the storage at room temperature for 10-21 days were successively pursued. The decrease in ascorbic acid was more considerable in comparison with the fall or the rise of dehydroascorbic acid and diketogulonic acid.

Day old chick was raised 30 days long on the basal diet with or without terramycin in amount of 30 mg per kg of the diet. When the basal diet containing skimmed milk powder was used, the growth rate was increased by the addition of terramycin. But the number of caecum microorganisms, the concentration of serum calcium, and the contents of vitamin A, B_1,B_2,C of the liver and muscle were not affected noticeably. When a basal diet containing soybean oil meal was used, no marked change was produced by the addition of terramycin, contrary to our previous reports.

Further studies were made on the problem offered by us why the availability of B vitamins differs between animal and vegetable diets. Experiments were performed on adult subjects and rats, and it was confirmed that dietary components, especially cellulose and protein, played an important role in this problem. 1) Thiamine in urine and feces increased when cellulose was added to the diet, suggesting that thiamine synthesis by intestinal flora was accelerated. 2) On the other hand, the urinary excretion of riboflavin increased only by adding both cellulose and protein. 3) Vitamin B_<12> or methionine and tryptophan were effective remarkably.

Observations were made on 350 men over ten months. It was found that vitamin B_2 deficiency occurred more readily under some unusual environmental conditions, such as at low ambient temperatures (-10 to 10℃) with high humidity (85%) or in sunless atmosphere. The subjects were put on a standard meal of 2900 cal. containing 95 g of protein, 18 g of fat, 190μg of thiamine, 150μg of riboflavin, 14 mg of ascorbic acid and an ample amount of vitamin A, riboflavin only lacking entirely. A considerable reduction in the visual power and dark adaptation occurred after seven months which could completely be restored by administration of riboflavin.

Investigations on the seasonal variation of vitamin B_<12> content in activated sludge showed that there seemed to be an optimum temperature for the growth of the microorganisms participating in the production of vitamin B_<12> in activated sludge, since the vitamin B_<12> content was highest in the activated sludge sampled in autumn followed by the spring sample and lowest in the sample obtained in winter and summer.

Estimation of vitamin B_<12> and total solids in the eluates obtained by the treatment of carbon cake on which the vitamin was adsorbed, with various concentrations of acetone showed that vitamin B_<12> was not eluted with acetone of concentrations less than 20 % while a considerable amount of impurities was removed by acetone of the same concentrations. Consequently, washing of the carbon cake with 15-20 % acetone and elution with 50-60 % acetone afforded to purify the original solution obtained from activated sludge approximately 20-25 fold.

Riboflavin in blood was extracted with water by heating the mixture at 80℃ for 15 minutes. The solution was mixed with trichloroacetic acid aqueous solution, and riboflavin in its supernatant was determined by the lumiflavin fluorescence methed. (cf. Vitamin, 6,528).

The determination of riboflavin content in human urine by the lumiflavin method not only gives no correct value, but also is sometimes difficult, owing to the fluorescent substances which inhibit the determination. Investigations on the fluorescent substances inhibiting the determination indicated that anthranilic acid and its conjugate disturbed the estimation.
