The relation between the chemical structure and the antioxidant effect of phenolic antioxidants was studies. The results on the whole were consistent with those reported by previous investigators. But as for polynuclear phenolic antioxidants, We have found an interesting result on the influence of the nucleus and the side-chain on the antioxidant effect.

Thiamine and riboflavin contents of 3 kinds of leafy vegetables, 3 kinds of root vegetables and 3-4 kinds of fruits placed at room temperature were estimated for ten days in succession. Losses of vitamins varying from 15 to 57 per cent were found. The losses being different in different foodstuffs, no general relationship of the progress of loss could be found.

One of the unknown riboflavin compounds which had previously been found by us in the cultural solution of the synthetic medium with the mutant of Aspergillus oryzae, was proved to be identical with riboflavinylglucoside isolated by Whitby with rat liver. In the present experiment, it was found that this mold easily synthesized not only riboflavinylglucoside, but also several sugar compounds of riboflavin in the reaction mixture containing riboflavin and maltose. Factors of the synthesis of riboflavinylglucoside were investigated, and soluble starch, salicin, lactose, melibiose, sucrose and glucose were found to be able to replace maltose as glucose donor, while riboflavin monophosphate could not replace riboflavin when phosphatase-free enzyme was used. It was suggested that the biochemical significance of these sugar compounds would be related to polysaccharide synthesis by Aspergillus oryzae.

It was found that the mutant of Aspergillus oryrae produced a new riboflavinphosphate. This new phosphate (Rf 0.17 by butanol, acetic acid and water) was isolated from cultural solution of this mold, and it was compared with other flavins on the characteristics of paper chromatogram or absorption spectrum. The chemical composition of this phosphate was found to be corresponding to riboflavin diphosphate, and its structure was suggested to be a cyclic form, since this phosphate could not reveal coenzyme activity of malic dehydrogenase of this mold.

The active principle in Saccharomyces cerevisiae which is powerful as an antioxidant is soluble in 70% alcohol and stable to 100℃. The concentrate of the active substance was obtained from the yeast by the following procedures : The yeast was extracted with 70% alcohol. The extract was adsorbed to active charcoal after the removal of impurities by acid clay. The active cake was eluted with ammonia-acetone, then precipitated with phosphotungstic acid. The addition to liver oil of 1% of this concentrate could practically keep vitamin A contained in liver oil stable.

The investigation indicated that the sulphur in the essential oil of Eutrema wasabi, a pungent component of it, was not utilized as a source of sulphur in the biosynthesis of thiamine with Staphylococcus aureus.

Thiamine content and its phosphorylation in several tissues of normal and alloxan diabetic rats were determined. They were considerably less in alloxan diabetic rats than in normal ones. After intravenous injection of thiamine the increase of the content of this vitamin was less in every tissue of diabetic animals than in that of normal. Its phosphorylation was also suppressed in the former. But, if insulin was administered prior to the injection of thiamine to the rats the content and the phosphorylation became the same as in the normal rats.

The absorption rate of thiamine by excised intestinal loops of diabetic rats was compared with that of normal rats. There was no significant difference between the amount of thiamine absorbed by the intestine of normal rats and that of insulin administered rats. On the other hand, the absorption by the intestine of totally pancreatectomized or alloxan diabetic rats was coosiderably less than in the case of the normal rats, and esterification of the absorbed thiamine was not seen at all in the former. When ATP was added into the intestinal lumen with thiamine, the absorption increased markedly in alloxan diabetic rats and the absorbed thiamine was phosphorylated to some extent, while in normal rats such effect of ATP on the absorption of thiamine was very slight.

The administration of pituitrin did not cause any significant change in the thiamine content of several tissues of normal rats. The ratio of bound thiamine to free one was not affected by this hormone. But in alloxan diabetic rats the thiamine content in liver and kidney increased after pituitrin and its phosphory1ation rate was enhanced. When pituitrin was administered together with thiamine to diabetic rats, the increase of thiamine content was much more considerable than when thiamine alone was used and its phosphorylation was also promoted. In normal rats these effects of pituitrin were only slight.

P^<32>-labeled FMN was prepared from the liver of rabbit, loaded with P^<32>-phosphate by using an ion exchange resin, IRA-400. The content of the produced FAD was determined by employing P^<32>-labeled FMN and ATP as substrates and homogenized liver of rat as enzyme, observing for 30 minutes at pH 7.4 and 35℃. The content of P^<32> per one mol was the same in FMN and FAD. Therefore, FAD is proved to have been synthesized from FMN in the following formula : FMN-P^<32> + ATP→FAD-P^<32> + Pyro. P

Emmerie-Engel's method of colorimetric determination of α-tocopherol was reexamined. The extinction value at 520mμ of colored solution followed Lambert-Beer's law in the limits of 0.01-0.5mg α-tocopherol. The stability of reagents and of colored solution was examined. Oxidized products of α-tocopherol separated by the aluminachromatography, and phenolic antioxidants presented color by addition of reagents as was the case with α-tocopherol. The method of removing them was described.

Furter-Meyer's method generally applied to the determination of α-tocopherol was discussed. The absorption spectrum of colored solution was characterized by a peak at 466mμ, and the extinction value at the absorption maximum increased in proportion to the concentration of α-tocopherol in the limits of 0.4-4.0mg. The conditions of coloring, the stability of colored solution and the method of removing other colorable substances by nitric acid treatment were studied. Satisfactory results were obtained by modified methods of Emmerie-Engel's and of Furter-Meyer's.

The effects of sugar compounds (riboflavinylglucoside, riboflavinylgalactoside) were compared with those of phosphoras compounds (riboflavin monophosphate, riboflavin diphosphate) of riboflavin upon the growth of riboflavin-less mutant of Aspergillus oryzae. It was found that positive values of difference (%) of growth efficiency (mycelium/sugar consumed) were obtained in presence of flavin glycosides, while negative values were obtained with flavin phosphates. Moreover the mycelium obtained in presence of flavin glycosides was found to contain a larger amount of glycogen and glucan. This seems to suggest that flavinylglycosides accelerates transglycosidation.

It was found that Clostridium acetobutyricum performed the synthesis of oligosaccharides from maltose, lactose and sucrose. The bacterium also synthesized enzymatically several riboflavinylglycosides, having the same Rf values with those isolated by us with As pergillus oryzas. The mechanism of oligosaccharide formation seems to proceed in the following steps, as was already proposed by us in Aspergillus oryzas : B_2-G+G→B_2-GG→B_2+CG B_2-GG+G→B_2-GGG→B_2+GGG

The author synthesized hydroxythiamine triphosphate by phosphorylating hydroxythiamine with metaphosphate according to Viscontini et al., and Koike. The author found that the synthetic hydroxythiamine triphosphate inhibited competitively the action of thiamine triphosphate in the carboxylase system isolated from bakers' yeast according to Green et al. This result agreed with that of Velluz et al.

Studying the isolation of crystalline vitamin B_<12> from activated sludge in industrial scale, a plan for pilot plant was worked out as follows : (1) Dried raw material is extracted with 5 parts of water at 90-100℃ for an hour.(2) Vitamin B_<12> in the filtrate is adsorbed on 0.3% of active carbon.(3) Adsorbent is washed with 15% acetone to remove impurities and then eluted with 50% acetone.(4) Concentrating the acetone eluate under reduced pressure, the vitamin is extracted by shaking the concentrate with phenol-butanol mixture.(5) The solvent layer is stirred with ether and water to transfer vitamin B_<12> to aqueous layer.(6) The aqueous layer obtained is rum through the column of Amberlite IRC-50 to adsorb the vitamin followed by the elution with 0.1N HCl in 75% acetone.(7) To remove hydrochloric acid, the eluate is run through the column of IR-4B. The acetone eluate of pink color is concentrated to a small amount. Finally, vitamin B_<12> is crystallized from acetone. One gramme of vitamin B_<12> may be obtained from one ton of dried activated sludge. Details of each process will be described in other papers.

Several conditions for the extraction of vitamin B_<12> from activated sludge and for the adsorption of the vitamin on active carbon were investigated. The results obtained from the experiments indicate that the satisfactory extraction of vitamin B_<12> from activated sludge is attained by heating at 90-100℃ for 30-60 minutes with 5 parts of water to the dried raw material, and that the addition of 0.2-0.5% (weight/volume) of active carbon to the extract makes an adsorption of 70-90% of the vitamin possible.

A kind of disease with the main symptom of motor paralysis of extremities was found among chickens. The disease happens chiefly in the spring and summer. The recidivation can often be seen. Symptomatically and pathologic-histologically the disease is similar to the experimental B-avitaminosis. Moreover Bacillus aneurinolyticns Kimura et Aoyama was found in the feces and intestinal tracts including duodenum of the chickens. Therefore the bacillus must be considered as a cause of the disease. The disease of slight symptoms can be got well spontaneously. Subcutaneous injection of thiamine or peroral administration of streptomycin is effective for the disease.

Riboflavin photolysis in an alkaline medium as in the case of the determination of riboflavin with lumiflavin method, is seriously inhibited by the addition of apple juice. The juices of other fruits such as pear, peach, grape also inhibit the photolysis, but is not as affective as apple juice. After the investigation on the components of apple juice, we found that fructose is the inhibitor of the photolysis and that sorbose has the same property.

The melanine tissue of frog skin contains riboflavin and several pterines. A violet fluorescent substance having the Rf value of 0.41 was recognized on the paper chromatograms (developed with n-butanol-acetic acid-water) with the extract of this tissue obtained by the treatment with 5% ammonia solution at 80℃ for 15minutes. The above treatment of pure riboflavin solution gave also the identical spot with that of the tissue extract. This substance is a new one, quite different from riboflavin or its known derivatives.

The quality of vitamin oil for export has so far been evaluated only by personal senses. A simple and rapid method for evaluation of vitamin oil applying color test wish p-quinone was devised. The principle of the method is as follows : 0.1% p-quinone solution in alcohol is added to acid-extract of vitamin oil. The color appearing after 5 minutes is compared with that of standard solution. The results obtained by this method with many samples of vitamin oils were in good agreement with those by the sensual method.

Phillips' rubber scale is often used for calculation of Rf-values in paper chromatography. One may, however, feel insecure as its reliability depends upon the uniformity of elongation of the rubber plate. We have therefore devised a scale made of celluloid or transparent plastic plate in the form of right-angled triangle with graduated lines drawn parallel to its base. The scale is so placed on the paper that the apex of the triangle is located at the original line of the paper and its base line at the end line of solvent. The graduated line passing through the center of each spot indicates its Rf-value. This scale can show exact values and is advantageous as it can be used in the dark room if substance is fluorescent.
