The content of coenzyme A in the liver of rats was determined by the method of Kaplan and Lipmann. The content of coenzyme A in the liver of rats to which desoxypyridoxine and hydroxymethylpyrimidine were administered with vitamin B_6 was markedly less than that of the control ones. The content of coenzyme A in the liver of rats to which desoxypyridoxine and hydroxymethylpyrimidine were administered without vitamin B_6 was less than that of the control ones. However, when isonicotinic acid hydrazide was administered to rats, the content of coenzyme A in the liver was more than that of the control ones both in the vitamin B_6-supplemented group and the deficient group.

In order to learn the relation between hematopoietic function in the bone marrow and xanthopterin metabolism, the authors determined the urinary excretion of xanthopterin in normal persons and patients with various blood diseases, and furthermore examined the change in the urinary excretion of xanthopterin in the course of these diseases. The results obtained showed that the daily urinary excretion of xanthopterin averaged 4.4 mg in 8 normal persons, and there were no differences between both sexes. The urinary excretion of the substance of pregnant women was increased. In 20 patients with various diseases, the daily urinary excretion of xanthopterin in 4 of 6 patients with aplastic anemia and each patient with chronic myelogenous leukemia, lymphatic aleukemia, chronic lymphatic leukemia and Kala-a〓ar, respectively, increased remarkably as compared with that of the control. On the contrary, in a young woman with aplastic anemia accompanied by the tendency of bleeding and a patient with anemia due to ancylostomiasis in the recovery stage of anemia the excretion of xanthopterin decreased. In either a patient with hypochromic anemia or anemia due to ancylostomiasis, the daily excretion of xanthopterin decreased gradually in the course of treatment.

The purpose of this study is to learn the relationship between the hematopoietic function of bone marrow and xanthopterin metabolism. The xanthopterin content was measured by the method of Koschara, modified by the authors in the peripheral blood and organs, especially the bone marrow and urine, of normal rabbits, and of the animals at the recovery stage of bleeding anemia or suffering from either benzene poisoning or X-ray irradiation. In the rabbits at the recovery stage of bleeding anemia, the xanthopterin content of the bone marrow was increased remarkably. The urinary excretion of xanthopterin showed no remarkable change. When the hematopoietic function of the bone marrow in rabbits was injured by benzene poisoning or X-ray irradiation, the xanthopterin content of the bone marrow showed no remarkable change in the former, but showed a remarkable decrease in the latter, while the urinary excretion of xanthopterin was decreased in both cases. It might be concluded that there is a close relation between the hematopoietic function and xanthopterin content in the bone marrow of rabbits, and that xanthopterin content runs parallel with riboflavin content.

Young rats were kept on a riboflavin deficiency diet, and riboflavin content in several organs of these animals was examined weekly. The riboflavin content of the liver, heart and kidney was decreased to about 1/4-1/5 as much as that of the control 4 to 5 weeks after the onset of feeding, while that of spleen and adrenal glands was not decreased. When these riboflavin deficient rats were given either 40μg of riboflavin with irregular intermittence or given orally 80μg or 40μg of riboflavin daily, the riboflavin content of these organs was rapidly increased. The most remarkable increase was observed in the rats given 80μg of riboflavin daily.

The effects of 2-methyl-4-amino-5-aldehyde and 5-carboxyl-pyrimidine, which were oxidized on the 5th position of toxopyrimidine, on mice were observed. The administration of the former caused death without convulsion, and that of the latter showed neither toxicity nor protective effect for the convulsion caused by toxopyrimidine.

Vitamin A and carotene added to "Miso" were decomposed considerably when kept in the thermostat at 33℃. The loss of vitamin A after 24 hours was 0-30%, varying with the kinds of "Miso". An attempt was made to apply thiobarbituric acid test (TBA) for fats in "Miso". "Miso" showing high TBA values were found to have less retention of vitamin A. No lipoxidase-activity was observed in "Miso".

Comparisons of the amounts of various inorganic salts excreted in the urine and faeces of rabbits and rats were made between the healthy and experimentally induced rachitic animals, especially when vitamin D_2 was injected. The injection of the vitamin caused a decrease of excretion of Ca, P, Fe and Cu in both the healthy and rachitic animals, and in far greater degree in the rachitic animals. On the contrary, the excretion of potassium decreased in the healthy animals, whereas it increased remarkably in the rachitic ones.

EFFECT OF RIBOFLAVIN, FLAVIN ADENINE MONONUCLEOTIDE OR FLAVIN ADENINE DINUCLEOTIDE UPON RIBOFLAVIN LEVELS OF BLOOD AND SALIVARY Na/K RATION IN CHILDREN WITH NUTRITIONAL DYSTROPHY Twenty nine boys with nutritional dystrophy were divided into 4 groups, and 1 mg of riboflavin, FMN or FAD was given subcutaneously for 4-5 successive days. The amounts of free and esterified riboflavin were measured fluorometrically prior to, and 4 days after the administration of the vitamin, and 10 days after the last injection of the vitamin. The rise of the concentration of both free and esterified riboflavin in blood was highest in FAD-group of these 4. Temporary decrease in the salivary Na/K ratio was found in free riboflavin-group and FMN-group, but not in FAD-group. This finding suggests that hyperactivity of the adrenal cortex might be required for the formation of FAD from free riboflavin or FMN.

When 10mg of riboflavin dissolved in 100ml of physiological saline was given into the stomach of dogs, the amount of the vitamin excreted into the bile was markedly smaller in malnourished dogs. Hepatic change, which was observed histologically in the malnourished dogs, was considered to be one of the factors relating to the failure of excreting riboflavin into the bile in the malnourished dogs.

Dogs were fed for 30-111 days on a deficient diet, which was low in protein and riboflavin, and high in carbohydrate. The composition of the deficient diet was similar to that which has customarily been taken by the inhabitants in Hirosaki area where nutritional dystrophy (so-called Shibi-Gatchaki) is prevalent. The riboflavin concentration in portal blood was estimated prior to, one hour, three hours, and five hours after the injection of riboflavin into the stomach of dogs. An increase in riboflavin, especially the esterified form, in the portal blood was markedly smaller in the malnourished dogs than in the control. The result showed that the intestinal mucosa of malnourished dogs was inferior to the control in the phosphorylating activity for the phosphorylation of riboflavin.

1) DL-Norleucine, L-histidine, L-phenylalanine or human red cell inhibits the growth of an assay organism, E. coli #215,while these inhibitions are relieved by the addition to the medium of methionine-free casamino acids to yields 0.25 per cent of a final concentration. Casamino acids enables the medium to protect against the growth inhibitory effect which occur during the autoclaving of the medium.2) Among 29 microorganisms tested, the greatest amount of vitamin B_<12> was found in cells of a strain of Pseudomonas aerginosa.

It was found that folic acid biosynthesis took place in two steps : first, p-aminobenzoyl-glutamate formation from PABA and L-glutamate, and second, folic acid formation from the formed p-aminobenzoylglutamate and xanthopterin. From experiments using an enzyme treated with ion-exchange resin (IRA-400) it was found that for the peptide formation from PABA and L-glutamate ATP, CoA and Mg ion were required as co-factors, and that by this reaction the equal moles of pyrophosphate and AMP were formed. From the fact that the p-aminobenzoylhydroxamate formation did not decrease in amount by the use of IRA-400 treated enzyme, and also from some other evidences., it was concluded that p-aminobenzoyl-AMP and p-aminobenzoyl-CoA acted as active intermediates in this reaction. The folic acid biosynthesis from p-aminobenzoyl-glutamate and xanthopterin was measured quantitatively with the method of IR-4B fractionation. This enzyme could also be separatd into active apo-enzyme and co-enzyme by the IRA-400 treatment.
