Vitamin B_<12> activities of fifty-seven kinds of Japanese foods were determined by the microbiological assay with Lactobacillus leichmannii 4797,and compared with the data reported in literature. It was recognized that small fish and shell-fish products which are edible together with viscera are a good source, especially in Japan, of vitamin B_<12> beside the intestines of animals.

The method of microbiological assay of vitamin B_<12> using Ochromonas malhamensis which responds specifically to cyanocobalamin was investigated. The organisms were cultured in the aluminium-capped test tubes placed slantly in incubator for 90 hours at 29℃, and then the growth was determined. It was recognized that the available range of determination of vitamin B_<12> per tube was 0.02-0.2mμg. When more than 4 mg or 2 mg of salts was included in the tubes containing 0.05mμg or 0.2mμg of vitamin B_<12> per tube respectively, the growth of the organism was considerably inhibited. Less than 0.5mg of DL-methionine per tube was not effective to the growth of the organisms. Vitamin B_<12> values of animal foods found by this method were almost the same with the values by the method using L. leichmannii #4797. However, vitamin B_<12> values of weeds by the method with O. malhamensis were less than 80% of those by the method with L. leichmannii.

It was confirmed that the crude thiaminase II of Bacillus aneurinolyticus could catalyze the hydrolysis of thiothiamine into 2-methyl-4-amino-5-hydroxymethylpyrimidine and 2-mercapto-4-methyl-5-β-hydroxyethylthiazole moieties by the methods of paper partition chromatography and ultraviolet spectrophotometry.

OMP, a toxic decomposition product of thiamine can be clearly separated from other related compounds by paper partition chromatography using butanol・acetic acid・water (4 : 1 : 5) as developer and can be found located definitely under ultraviolet light (wave-length of 254mμ) as a dark image. Although semiquantitative experiment reveals that contamination of OMP or other impurities such as SB, TDS, B_1-thiazolone, Thiochrome, AMP or Th may not exceed 1% for ordinary crystalline thiamine salt, at least two unidentified substances having smaller or larger Rf value than thiamine appear on the cbromatogram. These substances show thiamine-like properties towards chemical reactions.

OMP, a toxic decomposition product in thiamine injection was fixed chromatographically according to the method presented in the previous paper. From examination of 51 injections with varying shelf life ranging from 1 to 10 years, it was presumed that older preparations may contain OMP to a considerable amount. OMP which was fixed on filter paper from 12 representative injections was eluted with the aid of dilute hydrochloric acid (PH 1.0) and then determined spectrophotometrically. It's absolute content in injection extended from 0.02 to 1.27mg per 1ml of solution and these figures were equivalent to 0.2-3.8mg of OMP per 100mg of thiamine or decomposition of 0.5-9.2% of originally prescribed thiamine. A certain parallelism was found between decrease in thiamine and increase in OMP.

The rate of increase in urinary pyruvate and α-keto-glutarate in the first and the second hour following the intravenous administration of 20ml of 20% fructose in comparison with urinary excretion of these acids in 1 hour before fructose administration was measured. In patients with various liver diseases the rate of increase in urinary α-keto-acids was more marked than that in normal subjects. This data suggests that there may be disturbance in the metabolism of a-keto-acids in these patients. The acetylating activity following oral administration of 500mg p-aminobenzoic acid was decreased in patients with various liver diseases than in normal subjects. The decrease of acetylating activity was most marked in those with acute hepatitis.

Biotin L-sulfoxide has been isolated from Aspergillus niger culture filtrate by Wright et al. This compound has been shown to be as active as biotin for Neurospora but have a negligible activity for lactic acid bacteria and yeast. In Japanese fermentation products, in which Aspergillus oryzae plays an important role in the manufacturing process, it may be expected that biotin L-sulfoxide is formed by the mold and its presence might cause a discrepancy of the assay values of biotin between Neurospora method and yeast or lactic acid bacteria method. In this paper the authors performed a comparative assay of biotin activity in various Japanese fermentation products using Neurospora crassa and Saccharomyces carlsbergensis, and found a considerable difference between the results obtained by the two methods, as expected. The biotin active substances in the samples were found to consist of biocytin, biotin D-and L-sulfoxides and biotin.

The bacterial cell fractions obtained from the supernatant liquor of digesters for human excreta and distillers' wastes are noteworthy vitamin B_<12> resources. Bioautographic identification and characterization of vitamin B_<12> and its related factors were investigated in the bacterial cell fractions above mentioned for the purpose of utilization for industrial raw materials of the vitamin. Water-saturated sec-butanol containing a trace of cyanide with or without acetic acid or potassium perchlorate was used as a developing agent. Since some of vitamin B_<12> factors have similar Rf values it seems to be difficult to separate them completely each other and identify them bioautographically. However, it was found that the bacterial cell fractions examined contained other vitamin B_<12> factors than cyanocobalamin, and that for complete separation and identification of vitamin B_<12> factors, other appropriate techniques were necessary than paper chromatography with bioautography.

Intravenous administration of lysine caused decreased urinary excretion of biotin in some patients with liver diseases, diabetes mellitus and polyneuritis. Increase of urinary biotin after biotin loading, was less in these patients when lysine was administered previously. Biotin administration caused increase in hepatic content of bound biotin in normal rat. Less marked increase was found in rats with acute CCl_4 poisoning or alloxan diabetes. Simultaneous administration of lysine caused more marked increase in hepatic content of bound biotin in normal rats. The similar effect of lysine was not demonstrated in rats with CCl_4 poisoning or alloxan diabetes. The biosynthesis of bound biotin seemed to be disturbed in these conditions.

The thiamine content of pressed barley from the market in Aichi Prefecture was averaged 0.91μg/g (standard deviation 0.42μg/g). This value was fairly lower than that obtained by the authors two years ago. Therefore, it might be necessary to enrich the pressed barley with thiamine.

Salmonella enteritidis S 64 was orally administered to normal and folic acid deficient rats. No death was recognized in the both groups in a low dose (4×10^8 cells) of the bacillus, while the animals deficient in folic acid died 10 to 12 days after the ingestion of the bacillus as much as 1×10^9 cells. In the dead animals, the bacillus was detected in the heart blood, the liver, the spleen and the intestinal tracts, but not in those of the normal animals. These results seem to show that a folic acid deficiency lowers a preventive power from the infection with Salmonella enteritidis in digestive tracts.

The relationship between the autonomic nervous center and stress in thiamine metabolism in the organs were observed. 1) In rabbits the stimulation of sympathetic center in the hypothalamus caused a considerable decrease of total thiamine concentration in the liver (-61%) and kidney (-68%) after one hour and recovery after 24 hours. The cold stimulation caused a decrease of total thiamine in the liver after one hour (-38%) but a considerable decrease after 24 hours (-54%). Likewise the cold stimulation caused a decrease in the kidney after one hour (-27%) but a considerable decrease after 24 hours (-53%). 2) Simultaneous administration of Chlorpromazine suppressed the decrease due to stimulation. 3) Subcutaneous injection of Chlorpromazine did not show any influence in thiamine concentration, except for a slight decrease in the liver and kidney after one hour.

Changes in thiamine concentration in the liver and kidneys were observed when the left ganglion caeliacum was stimulated by two different methods. 1) In rabbits a sham operation caused a decrease of total thiamine concentration in the liver (25-30%) and in the kidneys (27-36%). 2) The pharmacological stimulation by lead acetate and electric stimulation similarly caused a considerable decrease of total thiamine concentration in liver and kidneys after 1-12 hours (35-46%) . 3) Subcutaneous injection of Chlorpromazine suppressed the decrease due to sham operation and sympathetic stimulation. 4) The participation of the autonomic nervous system and pituitary adrenal system in thiamine metabolism was suggested by the above results.

In dogs, the absorption of thiamine and cocarboxylase in the blind canal which were made by partial ligation of the small intestines were observed. 1) Thiamine was well absorbed in a short period of time, but there was no significant partial difference of the intestine in regard to the blood levels and the intestinal absorption of thiamine. 2) Highest blood levels of cocarboxylase was attained within 30 minutes, persisted much longer than with thiamine and increased in esterified form. 3) The intestinal absorption of thiamine was suppressed when monoiodacetic acid and several antibiotics were administerd with thiamine.

In healthy male adults and patients the urinary and fecal excretion of thiamine were observed. 1) In 4 healthy male adults the values of urinary excretion of thiamine were between 150 and 215μg/day after 890μg/day of thiamine intake. 2) In patients the urinary excretion of thiamine was less than in cases of healthy adults. 3) There was no correlation between the fecal excretion of thiamine and its urinary excretion.
