The metabolites of 2-(4-chlorobenzoylamino)-3-[2(1H)-quinolinon-4-yl]propionic acid (OPC-12759) (1), which has a potent antiulcer activity towards acetic acid-induced gastric ulcer, were synthesized to confirm their structures and to examine their antiulcer activity. The structures of the major metabolites (2-4) in the rat were identified by means of comparisons with the synthetic compounds. The antiulcer activity of the metabolites (2-4) was found to be lower than that of 1.

The ginseng dammarane saponin, ginsenoside-Rg
1, and the oleanolic acid saponins, chiku-setsusaponin-IV and pseudo-ginsenoside-RT
1, were isolated in yields of 0.02, 0.15 and 0.04%, respectively, from roots and small rhizomes of Panax pseudo-ginseng WALL. subsp. pseudo-ginseng HARA collected at Nielamu, Tibet. A saponin formulated as 3-O-α-L-arabinofuranosyl(1→4)-β-D-glucuronide of oleanolic acid was also isolated in a partially purified state. The chemotaxonomical significance of the results is disccussed.

Color reaction systems among throium(IV) {Th(IV)}, lanthanum(III) {La(III)} and xanthene derivative such as gallein (gall), in the coexistence of amphoteric and nonionic surfactants were studied, and spectrophotometric determinations of these metal ions (0-50μg of Th(IV) or La(III)/10ml) at 620nm by using gall-Th(IV)-La(III) ternary complex in the coexistence of sodium N-lauroylsarcosine (LS) as an amphoteric surfactant and Brij-35 as a nonionic surfactant were developed. The influences of foreign ions in this proposed method were greatly reduced (to 1/2-1/10) in comparison with the method in the presence of cationic surfactant alone.

The binding of flavin-adenine dinucleotide (FAD) in DT-diaphorase was studies. Incubation with 4M guanidine-HCl at 0°C rapidly dissociated FAD from the enzyme, and incubation of the apo-enzyme with FAD at 37°C restored maximally about 50% (for menadione-reducing activity), or 330% (for ferricyanide-reducing activity) of the initial activity. The half-time required to reconstitute the apo-enzyme with FAD was 2min for menadione-reducing activity and the K
m value of the apo-enzyme for FAD was 0.23μM. These results show that DT-diaphorase contains non-covalently bound FAD as a prosthetic group.

To determine whether or not the high-molecular-weight (>2×10
6) hyaluronic acid in rabbit pericardial fluid contributes to the changes in the viscosity of the pericardial fluid, we compared the viscosity of rabbit pericardial fluid with that of various glycosaminoglycan solutions by using a cone and plate viscometer at 37°C. Rabbit pericardial fluid showed non-Newtonian flow behavior and had an apparent viscosity of about 1.03-1.04cP at a shear rate of 750s
-1 as 37°C. On the basis of the data on the viscosity of pericardial fluid and of high-molecular-weight hyaluronic acid solutions, the high-molecular-weight hyaluronic acid (about 0.01% (w/v)) could contribute to the increase in the viscosity and the rheological characteristics of the pericardial fluid. These results suggest that the high-molecular-weight hyaluronic acid in the pericardial fluid could provide the heart tissue with resiliency and compliance when the tissue is subjected to stretching or compressive forces.

When deoxyribonucleic acid (DNA) was treated with ascorbic acid (AsA) in the presence of Cu
2+ under aerobic conditions, significant releases of cytosine, thymine, guanine and adenine from the DNA molecule occurred. Addition of catalase or chemical scavengers of hydroxyl radical to the reaction mixture prevented the release of the bases. Monoesterified phosphate groups were found in the reaction mixture, indicating that the cleavage of DNA had occurred. The total amount of the bases released was almost equivalent to the amount of monoesterified phosphate groups formed by the reaction of DNA with AsA and Cu
2+. These results indicate that the release of the bases from DNA molecule was easily caused by activated oxygen, possibly by hydroxyl radicals and/or hydrogen peroxide, formed by the autooxidation of AsA in the presence of Cu
2+.

In order to confirm the pathophysiological isgnificance of the dissociation between the plasma levels of aminopeptidases A and B in spontaneously hypertensive rats, we tested the effects of several enzyme inhibitors including inhibitors of aminopeptidase B on the course of hypertension in these rats. In addition to an ihibitor of angiotensin-converting enzyme, inhibitors of aminopeptidase B (arphamenine B and bestatin) significantly suppressed the development of hypertension in these animals. This finding may represent an important clue to the pathogenesis of essential hypertension.

Two crystal forms of the new dihydropyridine derivative, methyl (E)-3-phenyl-2-propen-1-yl 1, 4-dihydro-2, 6-dimethyl-4-(3-nitrophenyl)pyridine-3, 5-dicarboxylate (FRC-8411), were obtained by recrystallization from methanol. These crystal forms were identified by using powder X-ray diffractometry, infrared spectroscopy, differential scanning calorimetry (DSC) and thermogravimetry. By means of DSC, the melting points of forms I and II were found to be 140 and 121°C, respectively.Forms I and II, having a similar particle size distribution, were administered orally or intravenously to spontaneously hypertensive rats. In the case of oral administration, the hypotensive action of form I was milder than that of form II and tachycardia was not observed after administration of form I.

In this paper, we discuss the reason why plots of the reciprocal of ([sodium dodecyl sulfate]-cmc) against the reciprocal of the shift-length of the absorption maxima of solubilized alkylparabens show linearity, and what information can be derived from the straight line. Extrapolation of the double-reciprocal plot was utilized to obtain the maximum value of the shift-length, which can not be measured directly. The average of the distribution parameter values can be obtained from the slope of the plot.

The thermal cycloaddition reaction of 1-ethoxycarbonyl-1H-1, 3-diazepines (4a, b) with 2, 5-dimethyl-3, 4-diphenylcyclopentadienone (5) gave three kinds of 1 : 1 cycloadducts, exo [6+4]π (6), syn-endo [4+2]π (7), and anti-endo [4+2]π (8) cycloadducts, the structures of which were elucidated by spectral analyses and the following photochemical study. Irradiation of the [4+2]π cycloadducts 7 and 8 resulted in an intramolecular [2+2]π cycloaddition to afford the cage compounds 12 and 13, respectively, while the [6+4]π cycloadducts (6), upon irradiation, underwent a retro-reaction to give the bicyclic compounds (11), presumably via the parent 1, 3-diazepines (4).

3-Alkylamino-3-deoxy-D-allose and -D-glucose were synthesized from 1, 2 : 5, 6-di-O-isopropylidene-α-D-glucofuranose and investigated regarding their promotion or inhibition of plant growth using the Avena coleoptile straight growth test. The alkylamino monosaccharides with an 8 to 11 carbon alkyl chain promoted growth at a concentration of 500/3 ppm. The activity depends on the sample concentration : at a high concentration plant growth was inhibited. At a low concentration growth was promoted.

The known tumor promoter des-O-methylolivoretin C, and a new possible tumor promoter, des-N-methylteleocidin B-4, were isolated from Streptomyces mediocidicus and the structure of the latter metabolite was elucidated by chemical correlation with the known teleocidin B-4. The structures of teleocidin B-2 and a new metabolite, olivoretin E, obtained from Streptomyces mediocidicus and Streptoverticillium olivoreticuli respectively, were determined by means of single-crystal X-ray diffraction analysis.

Pleurisy was induced in rats by intrapleural injection of 10 uM phorbol myristate acetate (PMA) or 2 % carrageenin. Thirty minutes after PMA was administered following pretreatment with CV-3988, a specific antagonist of platelet activating factor (PFA), pleural fluid accumulation and its exudation rate were significantly suppressed. However the pleurisy induced by carrageenin was not affected by CV-3988. These results indicate that PAF may contribute to the exudation of PMA-induced pleurisy but not in carrageenin-induced pleurisy.
