We compared 24-hour plasma insulin profiles from summed free insulin aspart and free human insulin concentrations in subjects with Japanese type 2 diabetes administered thrice-daily BIAsp 70 (BIAsp 70 TID) or twice-daily BIAsp 30 (BIAsp 30 BID) for 4 weeks to healthy Japanese nontreated physiological insulin profiles. We also characterized 24-hour plasma glucose profiles.
The plasma insulin concentration reached Cmax after a relatively rapid increase after a BIAsp 70 dose at each meal (tmax: 1 hr after each meal) . The post-BIAsp 70 TID concentration exceeded post-BIAsp 30 BID, with a notable difference after lunch because BIAsp 30 was not injected after lunch. The plasma insulin profile was closer to that of healthy volunteers after BIAsp 70 TID than after BIAsp 30 BID.
Mean plasma glucose and postprandial plasma glucose increases at breakfast and dinner after BIAsp 70 TID resembled those after BIAsp 30 BID. Postprandial plasma glucose at lunch was lower than that premeal after BIAsp 70 TID. This increased after BIAsp 30 BID because BIAsp 30 was not injected after lunch.
