    In search of new compounds in simaroubaceous plants, we have been investigating naturally occurring substances [alkaloids, quassinoids, limonoids and picrotoxane-type terpenoids] on the following subjects : I. chemical studies on the constituents of Japanese simaroubaseous plants (I-1 and 2), II. chemical studies on the constituents of Indonesian and Chinese simaroubaseous plants (II-1-9), III. biological activities of the isolated natural compounds mainly and the chemical modification analogs (III-1-9). This article reviews the results obtained in our laboratory since 1976 on the subjects of I, II and III.

    This article accounts for the development of immunochemical studies on the antigenic polysaccharide, mannan, a major antigen of pathogenic yeast in man, genus Candida, in order to determine the chemical structures dominating the serological specificities of the parent cells as follows. 1. The serological classification system of 7 medically important Candida species by detecting 10 antigenic factors, 1, 4, 5, 6, 8, 9, 11, 13, 13b, and 34 the corresponding antisera, established by Tsuchiya and his coworkers is documented. 2. The structural studies of Candida mannans until early 1980s, which did not include any evidence for the presence of β-1, 2-linked Man unit, the common constituent of antigenic factors, 5, and 6, are also reviewed. 3. The process of structural identification of antigenic factor 5 residing in the mannans of C. albicans, C. tropicalis, and C. stellatoidea, in the phosphatebound form of β-1, 2 linked mannooligosaccharides, is summarized. 4. The results of structural identification of antigenic factors 4 and 6 in the mannans of the acid-stable domains of C. albicans are summarized as follows : In order to isolate oligosaccharides containing β-1, 2 and α-1, 6 linkages, a modified acetolysis method under mild conditions was established. By means of this procedure, oligosaccharides corresponding to antigenic factors 4 and 6 were successfully isolated, and their structures determined, subsequently. 5. Furthermore, effects of the alteration of cultivation conditions, carbon source, pH and temperature, on the chemical structure of mannans, especially of decrease and/or loss of densities of antigenic factors, 4, 5 and 6, are documented, because of the significance of these findings as basic concepts for in situ assay of Candida cells by antibody-staining technique in patients'foci.

    The methanol extract and water extract from the stem of Berchemia racemosa (Rhamnaceae) showed protective effects on liver injuries induced by carbon tetrachloride (CCl4) and α-naphthylisothiocyanate (ANIT) in rats. The acetone extract of B. racemosa protected the liver injury induced by CCl4. One of the protective substances was identified as carpusin. Some fractions showed significant protective effects against the liver injury and cholestasis induced by ANIT.

    It was found that (±) ethyl 2-[2-(3-hydroxypropyl)-4-oxocyclohexylidene]-propionate (1), (±) ethyl 2-[2-(3-hydroxypropyl)-4, 5-dimethoxyphenyl] propionate (2) and (±) 3-[2-(3-hydroxypropyl)-4, 5-dimethoxyphenyl]-2-butanone (3) had excellent antiulcer activities. In order to study structure-activity relationships, (±) 2-[(3-hydroxypropyl) phenyl] cyclopentanone derivatives (4, 5) and (±) 2-[(3-hydroxypropyl) phenyl] cyclopentanol derivatives (6, 7) were synthesized and tested for antiulcer activities. As a result, (±) 2-[2-(3-hydroxypropyl)-4-methoxy-5-(2-piperidinoethoxy) phenyl] cyclopentanone (5k) exhibited potent antiulcer activities.

    It was found that (±) ethyl 2-[2-(3-hydroxypropyl)-4-oxocyclohexylidene]-propionate (1), (±) ethyl 2-[2-(3-hydroxypropyl)-4, 5-dimethoxyphenyl] propionate (2), (±) 3-[2-(3-hydroxypropyl)-4, 5-dimethoxyphenyl]-2-butanone (3) and (±) 2-[2-(3-hyroxypropyl)-4-methoxy-5-(2-piperidinoethoxy) phenyl] cyclopentanone (4) had potent antiulcer activities. In order to study structure-activity relationships, (±) 3-[(3-hydroxypropyl) phenoxy]-2-butanone derivatives (5, 6) were synthesized and tested for antiulcer activities. Among them, (±) 3-[2-(3-hydroxypropyl)-4-methoxy-5-[3-(4-methylpiperidino) propoxy] phenoxy]-2-butanone·3/2 oxalate (6k) was selected as a preferred antiulcer agent.

    To obtain necessary characterizations of crystals, such as an extinction angle, an elongation, refractive indices and a birefringence, several polarizing photomicrographs taken under definite conditions by the immersion method using a kit of immersion oils prepared at intervals of 0.005 between nD 1.470-1.730 were found to be useful enough. A standard procedure to take the least sheets of necessary photomicrographs using the known key refractive indices was proposed, and 8 kinds of crystalline drugs of which key refractive indices had been measured definitely (group A) except the last one showing only one key refractive index (group B) were tested together. It was also found that habit parameters as well as habit coefficients useful to obtain a particle size distribution were easily measured from orthographic projections produced from colored photomicrographs and the above obtained birefringences.

    The direct expression of the pig testicular 3α/β (20β)-hydroxysteroid dehydrogenase (HSD) cDNA using a baculovirus expression system was investigated. A minimum essential cDNA containing the coding region of 3α/β (20β)-HSD was amplified by polymerase chain reaction (PCR) to extend the DNA linker including the Bam HI restriction site in the both ends of the cDNA. As the template, 3α/β (20β)-HSD cDNA, pBS 52, which was subcloned to Bluescript II was used. The PCR fragment was ligated to Bam HI-cut transfer plasmid (pBlueBac III). Recombinant transfer plasmid (pBlueBac-20β) constructed was cotransfected into Spodoptera frugiperda Sf-9 cells with the baculovirus. After transfection, the recombinant virus was detected by the plaque assay with color selection. The expression of 3α/β (20β)-HSD cDNA was detected by Western blotting and enzyme assay. The expressed protein showed the same molecular weight and immunochemical cross-reaction to the native enzyme. Furthermore, it had 3-keto reductase activity of 3α/β (20β)-HSD for 5α-dihydrotestosterone and 20-keto reductase activity for 17α-hydroxyprogesterone in the presence of NADPH.
