Aurothioglucose (oil soluble) was mainly tested to the cases who did not show good effectiveness by American method in hyposensitization therapy, and the results obtained were as follows: 1) Among 43 patients treated with Aurothioglucose, the number of cases showed excellent result was 16, good 15, non-effective 5, unknown 7 and the total number of effective cases was31 (86.1%). 2) The effectiveness of the treatment was observed especially in the cases of patients who have not been treated with steroid therapy before, younger age, atopic type and mild cases. But the effectiveness was not so clear in patients who have been treated with steroid therapy for a long period, older age, mixed or infectious type and severe cases. 3) In most of the cases showed good results, the effectiveness was observed within 3 months after treatment, while in most of the non-effective cases the effectiveness could not be observed in the cases treated over 5 months. 4)In the cases with good results the effectiveness was observed with the administration of drugs within 400 mg as the total dosis. However, there are a few cases with good results who have the effectiveness was observed after the administration of over 500 mg. 5) Not a correlation was observed between the effectiveness and gold concentration in serum. 6) After treatment with Romosol, rebound was seen in 2-3 months, however, it was getting better with treatment. 7) As side effect exanthem with itching, albuminuria itching of skin and stomatitis were observed. Side effects were usually observed in young women, and the occurance of side effect and effectiveness of the drug were usually observed at the same period. The cases who did not show side effect were usually resistant to the administration of large amount of the drug.

Anti-membrane factor was already reported as new factor detected in sera of patients with systemic lupus erythematosus. The eluate of solubilized erythrocyte membrane, using sepharose 4B column, contained two peaks with O.D. 280 nm absorption and phenol-sulfric acid method. The specific antigenicity against the anti-membrane factor was detected in the first peak. Sephadex G-150 column was used in the present paper for further analysis of antigenic glycoproteins in the solubilized erythrocyte membrane. Six and five peaks were demonstrated with O.D. 280 nm absorption, and Lowry-Folin's and phenol-sulfric acid methods, respectively. SDS-polyacrylamide gel electrophoresis detected the molecular weight in each fraction, been ranged from 230000 to 15000. Positive antigenicity, using the micro-Ouchterlony method, against anti-membrane factor was found identically in all fractions except the peak 6 of O.D. 280 nm absorption. From these data, the antigen activity of the glycoprotein in the solubilized erythrocyte membrane was found to localize in the smaller portion of glycoprotein chains which had the molecular weight approximately 15000 and contained glucose and protein with the ratio of 0.39:1.0.

Serological studies including immune electrophoresis, agar double diffusion technic and passive hemagglutination method were performed on sera in 34 rabbits immunized with the so-called focal antigens. The antigens were prepared from the subcutaneous Arthus type lesions induced in rabbits which had been immunized with hen egg-white. The lesions were aseptically excised and separated into central necrotic (N) and peripheral wall (W) parts, which are referred to as the focal antigens (N and W) in this series of experiment. They were homogenized and repeatedly injected again either autologously or homologously to rabbits previously immunized with hen egg-white. Following results were obtained. 1) Antibodies to the focal N-substance, which were identified as antibodies against main antigenic component of the egg-white, were detectable in rabbits sensitized with N- or W-substance from the eary stage, especially to the later period (over 120 experimental days) with high titer in rabbits sensitized with N-substance. 2) Antibody to the W-substance was observed in 10 of 34 rabbits in over 80 experimental days with agar double diffusion technic and passive hemagglutination method. This antibody which was not absorbed with the egg-white or rabbit whole serum, cross-reacted with emulsion of autologous tissues such as the kidney, liver and heart. 3) In addition to the main component of egg-white, the focal antigens contained some components of rabbit whole serum. These serum components were present in much more amount in the W-substance than in the N-substance. 4) Immune electrophoretical analysis of urine disclosed appearance of IgG, native and denatured, and transferrin in increased amount in cases with amyloid nephropathy and glomerulonephritis with amyloid deposits.

Intractable asthma is defined as a type of bronchial asthma which is resistant to conventional therapies and, consequently, steroid-dependent for keeping patients' daily lives active. This study was aimed at investigating whether or not and how hyperresponsiveness of the bronchi played a role to make asthma intractable among patients with such type of asthma. The responsiveness of the bronchi was expressed as the threshold value to acetylcholine, i.e., the minimal amount of acetylcholine to induce bronchoconstriction. Seventy patients with asthma were divided into 4 groups, i) mild ii) seasonal iii) perennial and iv) intractable. The geometric means of the threshold values were 1135, 1143, 483 and 245μg/ml, respectively. The difference of mean values between group ii and iv is statistically significant at 1% level. These results show that the responsiveness of the bronchi is increased among patients with intractable asthma as compared with those with less severe asthma. The distribution of the threshold value in each group overlapped widely. So that, hyperresponsiveness of the bronchi does not seem to be only one factor to make asthma intractable. Inhalation of 5% orciprenaline raised the threshold value by 4 times in intractable group and by 10 times in other groups. This defference is not statistcally significant. Forty mg of prednisolone hemisuccinate i.v. or 3 days' oral administration of prednisolone, 10 mg per day, brought slight but statsitcally significant increase of the threshold value among patients with asthma. Corticosteroids seem to suppress the hyperresponsiveness of the bornchi in asthma.

An exact mechanism of gold therapy on bronchial asthma is still unknown. In order to elucidate this mechanism, the adrenocortical function was studied in asthmatics treated with gold salt. Gold sodium thiomalate was administered intramuscularly into 42 patients once or twice a week, and each patient was given 1850 mg of it in total. The adrenocortical function was examined by using rapid ACTH test, i.e., the plasma cortisol levels were measured before and 60 minutes after the administration of 250μg of ACTH intravenously. The serum gold concentration was measured at the time of rapid ACTH test simultaneously. The results were as follows: 1) The initial adrenocortical function was decreased in severe, chronic or long term suffered asthmatics. 2) In 21 out of 33 patients whose initial adrenocortocal function showed lower than normal, the function were improved at the end of the treatment. 3) The result of the treatment, from the view-point of clinical manifestation was effective in 31 cases, however the improvement of the adrenocortical function was not necessarily accord with the clinical effectiveness. 4) The average concentration of gold in serum, which had increased in parallel with dases of the administration, was 633μg/100ml at the end of treatment. However the serum gold cocentration was not related to the clinical effectiveness or the improvement of the adrenocortical function. From the results above-mentioned, it is considered that activated adrenocortical function by gold salt plays one role on the effectiveness of gold therapy on bronchial asthma.

In order to elucidate a mechanism of gold therapy on bronchial asthma, on the basis of antigen-antibody reaction, the IgE level in serum was measured by using redioimmunosorbent test in 29 asthmatics treated with gold salt. The results were as follows: 1) The average value of the serum IgE levels was heigher in asthmatics than in normal subjects and especially heigher in the atopic or paroxysmal asthmatics. 2) In 23 out of 29 patients, the serum IgE levels were decreased at the end of the treatment. 3) The decrease of serum IgE level was not necessarily accord with the clinical effectiveness, however it was nearly accord with the time of manifestation of effectiveness. 4) The decrease of serum IgE level was not related to the concentration of gold in serum or adrenocortical function. From the results above-mentioned, it is considered that suppression of antigen-antibody reaction by gold salt plays one role on the effectiveness of gold therapy on bronchial asthma.
