Polyethylene glycol precipitation-complement consumption test (PEG-CC) of Harkiss was simplified and applied to measurement of circulating immune complexes (CIC) in the sera of the patients. The concentration of more than 5 μg per ml of aggregated IgG can be detected by PEG-CC and about sensitivity, PEG-CC is equal to the radioimmunoassays which have been commonly used. Clinical studies showed that the normal range was less than 20 percent of PEG-CC and that high percentage of PEG-CC was found in SLE (mean is 37.0%), rheumatoid arthritis (43.9%), and hepatitis (63.9%), Relatively high percentage of PEG-CC was found in diabetes mellitus (29.9%), acuteglomerulonephritis (27.5%) and malignancies. PEG-CC was utilized for differential diagnosis of the diffuse lung diseases. Large quantity of CIC was found in the systemic immune diseases such as SLE, periarteritis nodosa and so on. Idiopathic pneumonitis, extrinsic allergic alveolitis and sarcoidosis with uveitis showed relatively high percentage.

Eighty eight asthmatic children and 24 healthy control children participated in six ski camps held since 1973 were subjected to pulmonary function test, and serum FFA, cortisol and DβH measurement before and after skiing. Frequency of asthmatic attack on skiing was 11%, 27% and 62% in the mild, moderate and severe group respectively. Recovery from an attack with no medication was observed in asthmatic children in the mild group. However, most asthmatic children in the severe group needed extra bronchodilator. Marked increasing of FEV_1.0 and PFR was observed after two hours of skiing. The serum FFA, cortisol and DβH after skiing increased significally except for the severe group. We conclude that the asthmatics can play ski safely when adequate advice and help are given by the doctors.

A clinical survey on patients with mosquito allergy was performed through questionnaire. The survey was cooperated with 120 dermatologists mainly and 120 pediatricians in the Kanto area. The results were as follows: 1) The patients with mosquito allergy were reported 87 cases of both sexes in total, in which 66% of them was under 10 years of age. 2) The patients who have at least a case with mosquito allergy in their family histories were 7 cases out of 87 patients and atopic diseases were considered to be partly related with mosquito allergy through their family histories. 3) The symptoms due to mosquito bite were mainly limited on skin such as redness and swelling (94.3%), itching (74.7%), bulla(46.9%), pain, ulcer, scar, etc. 4) The locality of biting by mosquito was seen mainly on their extremities and it took place in the evening and in or near their houses. 5) Laboratory findings were revealed abnormalities only in liver function tests and blood examinations in which the decrease of neutrophil and the increase of lymphocyte in differencial counts of while blood cells and the increase of immunoglobulins, especially IgE in several patients were recognized. 6) Some patients reacted to the skin test by antigens from mite or mosquito. 7) They were considered as insect bite or mosquito allergy and most of them recovered within a week with symptomatic therapies. 8) Almost all the patients did not react to the bite by other insects except mosquitoes.

We found a phenomenon that T-cells bearing receptors for the Fc portion of IgG (TG-cells) show cytotoxicity against cultured lymphoid cell lines bearing Fc (IgG) receptors and designated as immune complex dependent T-cell mediated cytotoxicity (IDTC). TG-cells are also known to work as effectors ofs pontaneous cytotoxicity (SC). However, these two phenomena seemed to be distinct from each other, since IDTC worked even at low effector/target (E/T) ratio while higher E/T ratio was required for SC and IDTC activity was lost by the treatment of TG-cells with mitomycin C (MMC) while SC activity was resistant to MMC. Various cell lines were studied for their sensitivity to SC by ^51Cr relerase, inhibition of DNA synthesis and inhibition of cell growth. Some were lysed but some other were only inhibited of DNA synthesis or cell grouwth and theremining showed resistance. These observation indicase that there is the difference of sensitivity to SC among cell lines. Cord TG-cells usually showed IDTC but only few cases had cytolytic activity by SC. Some showed SC only when they were assayed by inhibition of DNA synthesis or cell growth.

Changes of serum IgE levels and IgE antibody titers were studied in 40 bronchial asthmatic patients in an asthma home. Radioimmunosorbent test (RIST) or radioallergosorbent test (RAST) were employed. All patients were house-dust sensitive, and antibody titers against house dust measured by RAST were above score 2. Thirty patients were on hyposensitization therapy for house dust and ten were not. The results were as follows. 1. The dose-response curves of RAST using paper disc-coupled antigen and sera from D. pteronyssinus-sensitized patients and reference serum were linear and paralleled with each other when % total count (radioactivity of samples × 100 divided bu total radioactivity added) and IgE antibody titers were plotted on a log-log scale. However, the dose-response curve between paper disc-coupled antigen and sera sensitized by house dust was not linear and formed a plateau in a high antibody titer region. The deficiency of house dust antigens which were coupled to paper disc was suggested. 2. Serum IgE antibody titer against mold was higher in summer than in spring. Serum IgE antibody titer against D. farinae was higher in winter than in spring. Serum total IgE levels were higher in summer than in spring. These results suggested seasonal changes in IgE antibody titers. 3. Serum IgE antibody titer against house dust in the hyposensitization group showed no significant tendency of decreasing IgE antibody titer. 4. Serum total IgE levels in the hyposensitization group showed no obvious tendency of decreasing total IgE levels. 5. The RAST score in patients hyposensitized by house dust showed few changes, but the concentration of allergents required to produce a positive reaction increased in about half of the patients. This might suggest a reduced skin reactivity after hyposensitization therapy.

In 139 workers exposed to diatomaceous earth dust and 146 normal subjects, we examined the levels of serum protease inhibitors such as α_1antitrypsin, α_1antichymotrypsin, antithrombin III, inter-α-trypsin inhibitor, α_2macroglobulin and CLs inhibitor using single radial immunodiffusion methods. 1. The levels of antithrombin III in normal subjects (146 cases) and those of α_1antichymotrypsin and α_2macroglobulin in normal female ones (73 cases) significally correlated with aging (p<0.05). The values of antithrombin III, inter-α-trypsin inhibitor and α_2macroglobulin tended to be higher in female controls than male, whereas the inverse trend was seen in the case of α_1antichymotrypsin. 2. The levels of α_1antitrypsin, antithrombin III, inter-α-trypsin inhibitor and α_2 macroglobulin were significantly lower in workers exposed to diatomaceous earth dust than in normal subjects, while levels of Cls inhibitor were significantly higher in such workers than in the normal subjects. 3. Partial regression analysis of the values of inter-α-trypsin inhibitor and α_2macroglobulin revealed a significantly negative correlation with years exposed to diatomaceous earth dust in male non-smokers, but not in the case of male smokers. On our results and those reported in allied literature we discuss the important role of protease inhibitor in workers exposed to diatomaceous earth dust.
