1) The method of horizontal starch grain zone electrophoresis was employed for the studies on ALD isozyme in the normal kidney, the bladder, the prostata as well as the respective organ tumors, the normal liver and the iliopsoas muscle.
2) The ALD isozymes, ALD I and II were confirmed for the first time in the normal prostata, the bladder, prostatic tumors (cancer and adenoma). Especially, in the normal prostata and prostatic cancer, ALD II which was failed to demonstate with veronal buffer but borate buffer afforded the excellent electrophoresis. Protein patterns with a lower albumin peak and A/G ratio were observed in a group of kidney, liver and iliopsoas, while that with a higher albumin peak and A/G ratio, in a group of bladder and prostata.
3) Kinetics of ALD isozyme were determined as Optimum pH being 8.5-9.0, Km 0.64-0.81×10
-3M, and relatively thermostable, which were observed with little difference among isozymes.
The effectors employed included Na, K, Ca, Mg, Mn, Zn, Cu, Co, Fe
++, Fe
+++, cysteine-HCl, glutathione of reduced type, ascorbic acid, 2-mercaptoethanol, EDTA, diethylstilbesterol, PCMB, hexestrol, testosterone propionate, among which only diethylstilbestrol was able to demonstrate the differences in the isozymes with ALD II more strongly inhibited.
