Susceptivility for PZA on H37Rv in vitro was investigated. For this purpose various kind of hydrogen liquid or solid nutrient medium were applied.
1) With solid media (KH
2PO
4 media) under pH 7.5-5.5 by 10
-1mg demonstrated complete resistance at 3, 000γ, under pH 5.0 to 50γ. However, by applying 10
-4mg indicated under pH 7.0 at 3, 000γ, under pH 6.5-5.5 to 1, 000γ, and under pH 5.0 to 10γ. This was decided at the number of 1/3 colonies against the contrast.
2) Liquid media (Kirchner media) - under pH 7.0-6.0 by 10
-1mg, growth of the bacillus were seen up to 3, 000γ.
However, under pH 5.5 growth was demonstrated up to 1, 000γ and under pH 5.0 inhibition of bacillary growth was at 10γ. When 10
-4mg of bacillus was inoculated the growth was variable dependent upon pH, pH 7.0 to 3, 000γ, pH 6.5 to 1, 000γ, pH 6.0 to 3, 000γ and under than pH 5.5 showed no growth.
This indicated that the inhibition of growth of tubercle bacillus was seen under the following conditions : the greater H37Rv quantity the higher concentration needed, and the more acidity the media inclined the lower concentration needed.
The INH was added to the above mentioned kinds of nutrient medium and the similar test was made.
1) Solid media with 0.01γ/cc INH - under pH 7.0-6.0 demonstrated complete resistance at 1, 000Wγ, under pH 5.5 to 50, and pH 5.0 to 10.
2) Liquid media demonstrated the similar phenomenon.
This indicated that a synergic relation exists between PZA and INH.
For the purpose to study variety of susceptivility for PZA, investigation was made on two different conditioned media: 3% K
2PO
4 media with pH ca. 6.2 resisting up to 3, 000γ and 6% KH
2PO
4 media with pH ca. 5.5 resisting up to 1, 000γ. And 57 patients who had been unsuccessfully treated by SM, PAS and INH were tested.
The results are as follow:
1) Before their chemotherapies, the case of emergence of PZA-resistant strain was none.
2) One month after their therapies the percentage of emergence was 33.3.
3) After 3 months 73.7% and after 6 months 89.2%.
4) Compare with the INH-sensible strain, the INH-resistant strain had higher percentage on the emergence of the PZA-resistant strain. And this difference was noticed particularly in the early stage of the therapy which was estimated at least less than 4 months.
5) Six months' PZA-INH therapy showed decrease of both the SM resistant and PAS resistant-strain.
