Tropolone makes the nncleal body on the groupe of Cycloheptatrienolone which is one type of insaturated cycloheptatriene compounds. Some of the derivatives from these, for ex-ample, -Colchicin in 1932, Hinokitiol in 1936-had been extracted from the botanical tissue since quite a long ago, and because of its peculiar characteristics the studies concern-ing the presence of the biological effect so well as the chemical and physical research of these compounds had been strongly demanded.
And then, the decision of the chemical construction of Tropolone and the artificialy build up of Tropolone and its derivatives became possible only in recent years (between 19-48 and 1950).
1, I have tried to make clear the biological effect of this new compound, and then, as the first step, I have made the reference investigation in order to obtain the whole picture of chemical and physical characters of this compound. That is as follows.
1) Tropolone compound is one kind of cycloheptatrienolone which is one type of in-saturated cycloheptatriene compounds. Tropolone is a nucleal body of these compounds.
2) However, Tropolone and its derivatives belong to the insaturated cyclotriene com-pound, they have quite a stable characteristics chemicaly together with the semiaromatic character. According to the condition of the reaction it is possible, that nucleus may ele-ctrified both electropositive and electronegative. Therefore semetimes it may react acidi-ty and other timos as alkaline, and even it is possible to create great difference in their reaction direction.
3) In order to be the flat heptatrien and the ring strain which is caused upon the con-struction of the Tropolone kern the flatness of the kern may be lost due to the cubic dis-turbance which is caused by the far greater cubic result in comparison with the Benzen Kern. And because of this it is assumed that this will bring a great change in the insaturat-ed electron system and will influence greatly to the chemical and physical characteristics. 4) Tropolone group have the strong mutual action toward some substances, because of this construction of having, such characteristics. Therefore, it is understood that some of the Tropolone group give the spectacular biological effect.
5) Therefore not only from the standpoint of pure chemical or pure physical science, Tropolone group in recent days are investigated from various points of view such as biolo-gy, biochemistry, or heredity especially for application of it forr chemical treatment against cancer and others. This is valuable of attention. 6) However, I can find the very few investigations about the biological effect of Tro-polone itself.
7) Due to some connection at the beginning of the research procedures of this sub-stance, Professor Katsura and Nozoe had been investigating mostly on the clinical applica-tion of Hinokitiol, which is Isopropyl-Tropolone to tuberculous diseases. The result of this was proved to be a little effective against tuberculous diseases.
2. I have made several experiments using this Tropolone which is composed according to professor Nozoe's method (mp 48°-°c, colorless crystal …… tetragonal system……) in order to clear the biological effect of Tropolone.
A. At first, I examined the biological effect of Tropolone will apply the Arndt-Schulz's Pule or not, and obtained the following results.
a) The growth of Mycobacterium tuberculosis Typus gallinaceus (Su-cho strain) is definite-ly prevented in the Sauton medium with addition 1/10, 000 diluted Tropolone according to the culture result of 24 to 240 hours
b) The growth of Typhoid, Solmonella paratyphoid A, Salmonella paratyphoid B, Ko-magome B II & Esche.richia coli is definitely prevented in the 1/10, 000 Tropolone dilution added Buillon medium according to 24-72 hours culture results.
c) The growth of Bacillus anthracis, 209 p and Heatley is definitely prevented in the 1/320, 000 Tro
