Histological changes of neuropathy induced by chronic administration of acrylamide in the rat were investigated quantitatively under the light and electron microscope. The effect of the mixture of thiamine disulfide, pyridoxine hydrochloride and aquocobalamin on the neuropathy was also investigated. Following parameters were used : (1) distribution of transversal area of medullated-nerve fibers, (2) degree of transformation of medullated-nerve fibers, (3) ratio of transversal area of myelin to that of medullated-nerve fibers, (4) stromal area of sciatic nerve, (5) numbers of degenerated medullated-nerve fibers and Schwann's nuclei in the unit area, (6) RNA level in anterior horn cells of the spinal cord of the lumbar area and (7) histochemistry of motor end-plate. A beneficial effect of the mixture of vitamin B group was observed on the acrylamide-induced changes of the transversal area of medullated-nerve fibers and the number of Schwann's cells.

Prior to investigation of metabolism of ascorbic acid 2-sulfate (AsS) in guinea pig, methods to determine the amount of AsS in serum, urine, liver and other organs intubated with AsS have been studied by high speed liquid chromatography (HSLC) and column chromatography (CPC). AsS in organs was extracted with water by using homogenizer, and the resulting extracts were filtered with a filter (0.45μ) after centrifugation 100,000 x g, 30 minutes. 1) Method for HSLC : the operating conditions are following, instrument; Model JASCO HSLC 350,detector; UV photometer 254nm, column; JASCOPAC AV 02-500,mobilephase; 0.05M KH_2PO_4 : 0.5M KH_2PC_4=7 : 3 v/v, column pressure; 50kg/cm^2,flow rate; 1.0ml/min, sample volume; 2〜2Oμl and retention time; 10 minutes. 2) Method for CPC : chromatography of extracts 0.1〜2ml were performed with a column (0.6×2cm) of DEAE-Sephadex A-25(C1-type), column were washed with 50ml of water and 50ml of 0.1M KH_2PO_4 after absorbing, AsS eluted with 1M KH_2PO_4. The elution was followed by measurement of ultraviolet absorbancy at 254nm. The lower limit of detection were following, HSLC : serum 0.05pmol/m1,urine 0.2-=0.3μmol/m1,other organs 0.5μmol/g. CPC : serum 0.05μmol/m1,urine 0.2〜0.3μmol/m1,other organs 0.8μmol/g.

Metabolism of ascorbic acid 2-sulfate (AsS) was studied by use of young male of hartley guinea pigs on intubation of 0.5mmol AsS. The AsS levels of serum, liver, kidney in AsS-treated animals were 26μmol/100ml, 5.6 and 3.1μmol/g in 30min, 7μmol/100ml, 3.8 and 2.1μmol/g in 180 min, respectively. The amount of AsS excreated in urine in 24 hours after intubating was 48%, in 24〜48 hours 18%, in 48〜72 hours 6% of the dose administered in guinea pig. In the absorptive mechanism of intestine of AsS studied by the Wilson's method using inverted small intestinal loops of guinea pigs, the absorption of AsS was considered to be the same or more that of ascorbic acid, dehydroascorbic acid. When AsS was passed through the small intestinal wall, the hydrolysis reaction of AsS were not recognized.

Characteristics of two different vitamin D-dependent calcium binding proteins in rat intestinal mucosa were studied using polyacrylamide disc gel electrophoresis and Scatchard method. The smaller vitamin D-dependent protein, found predominantly in the duodenum and jejunum, showed electrophoretic mobility similar to that found in other mammalian species, and has one high affinity site with an apparent intrinsic affinity constant of 2.5×10^5M^<-1>. On the other hand, the larger vitamin D-dependent protein, found mainly in jejunum and ileum, showed electophoretic mobility similar to that found in chick, and has about 3.6 high affinity sites with an apparent intrinsic affinity constant of 1.2×10^6M^<-1> and 85 sites with an affinity about 2.2×10^4M^<-1>.
