Liver is the most important site for choline metabolism and possesses all enzymes relating to three different metabolic pathways. In order to find the metabolic change of hepatic choline-flux concerning sex differences, senility and administration of hepatotoxine, two enzyme activities, one is choline dehydrogenase (EC 1.1.99.1), the initiating enzyme for oxidation pathway, and the other is choline kinase (EC 2.7.1.32) for Kennedy pathway, were measured with rat liver. A close correlation between choline dehydrogenase activity and body weight as well as liver weight was obtained on the normal feeding rats aging from 25 day-old to 12 month-old, totaling 132, independent of sex. Choline dehydrogenase activity of senile rat was significantly depressed. Upon ethionine feeding, choline dehydrogenase activity was considerably depressed. On the other hand, choline kinase activity was activated twice, presumably, for the metabolic compensation.

Thiaminase II (Thiamin hydrolase) of Bacillus aneurinolyticus was purified to homogeneity. The enzyme activity was spectrophotometrically determined by measuring the rate of decrease in absorbance at 318 nm. The optimum pH and temperature were 9.4 and 65℃, respectively. Apparent activation energy was calculated to be 16.1 kcal/M from Arrhenius plots, and the Km value was 8.4 μM. Some sulfhydryl reagents enhanced the enzyme activity by 180-300%, while EDTA had no effect. Zn^lt:2+> among metal ions tested strongly inhibited the enzyme activity.

The conversion of N^1-methylnicotinamide (MNA) and trigonelline (TRG) to niacin and their toxicity in rats were investigated. The conversion of MNA into nicotinamide in rats did not seem to occur since the niacin effect was not observed when MNA was administered with tryptophan-imbalanced-niacin-free diet to weanling rats. When a large amount of MNA or TRG was administered to weanling rats, no growth retardation was observed. All of the TRG taken was excreted unchanged into urine. On the other hand, 10% of MNA taken was metabolized to N^1-methyl-2-pyridone-5-carboxamide and 70% of MNA was excreted unchanged. Even when a large amount of TRG or MNA was administered to rats, urinary excretion of niacin did not increase. From these results, it is concluded that the conversion of MNA and TRG into niacin does not occur and MNA and TRG are inert and very safe compounds in rats.

Vitamin B_1 content of canned fish, pork, beef and vegetable was determined by three methods using various solvents and oxidation reagents. Vitamin B_1 activity of vitamin B_1-like substance existing in canned mackerel was investigated using Lactobacillus fermenti. 1) The method using K_3Fe(CN)_6-oxidation and benzene : n-butanol extraction seems to give true vitamin B_1 values of canned foods examined but does not by K_3Fe(CN)_6-oxidation and isobutanol extraction. 2) Vitamin B_1-like substances existing in canned mackerel exhibited no vitamin B_1 activity in L. fermenti.
