Activity of papain was inhibited by incubating with CySH, GSH or thiol form of thiamine in the presence of a catalytic amount of aquocobalamin (OH-B_<12>) or cyanocobinamide (Factor B). On the contrary, the inhibition of papain by OH-B_<12> in the presence of the disulfides of these thiols was scarecely observed. The inhibition was completely suppressed by the addition of catalase. The loss of thiol groups in the inhibited papain corresponded to the decrease of the enzyme activity. The inhibited papain was completely reactivated by treatment with CySH. When the papain inhibited by the reaction with OH-B_<12> and thiol-thiamine was treated by Na_2S_2O_3,thiamine was released from the enzyme. From these results it was concluded that OH-B_<12> and Factor B catalyze the formation of mixed disulfides between small molecular thiols and papain.

Cyanocobinamide (Factor B) and aquocobalamin (OH-B_<12>) inhibited yeast alcohol dehydrogenase (YADH). This inhibition was remarkably increased by the addition of GSH. GSSG, however, did not affect the inhibition of YADH with OH-B_<12> or Factor B. The inhibition with Factor B and GSH was suppressed by the addition of catalase. Completely inhibited YADH, obtained by the gel filtration of a reaction mixture, was found to lose about 7 SH groups (mol/YADH mol) and to release the GSH bound to the enzyme by CySH reduction. The inhibited YADH was partially reactivated by CySH. From these results it was concluded that OH-B_<12> and Factor B catalyze the formation of mixed disulfides between small molecular thiols and YADH.

The distribution and metabolism of thiamine disulfide (TDS) in rat organs were compared with those of vitamin B_1 and O-n-butyryl TDS (BuTDS) by the use of ^<35>S-labeled compounds. In the present study we developed a column chromatographic method which enables to discriminate conveniently between the intact and the metabolites of these compounds. Total B_1 contents in various organs were higher than those of plasma or blood corpuscle 4 or 8 hours after the administration of TDS or BuTDS, and the characteristic distribution of TDS was similar to that of BuTDS but differed from that of B_1. All the three drugs were found in the liver with the highest concentration. This fact suggests that the liver probably plays an important role as a storage organ for these drugs.

The effect of 1α-hydroxyvitamin D_3 (1α-OH-D_3) on the intestinal Ca transport and serum Ca level were studied in vitamin D deficient anephric rats. Intestinal Ca transport was stimulated by a dose of 312 pmol of 1α-OH-D_3 in anephric rats as well as sham operated rats, whereas serum Ca level was not changed by the dose. Increasing the amounts of 1α-OH-D_3 to 6,250 pmol, the effect on serum Ca level was observed. These results suggest that 1α-OH-D_3 is less effective on the mobilization of Ca from bone but is effective on intestinal Ca transport. From these points, the some possibility of the presence of unidentified active vitamin D_3 metabolites on bone was discussed.
