JOURNAL FREE ACCESSGlycine N-methyltransferase (EC 2. 1. 1. 20) catalyzes the transfer of the methyl group of S-adenosyl-L-methionine (AdoMet) to glycine to form sarcosine and S-adenosyl-L-homocysteine. As opposed to monomeric structure of most AdoMet-dependent methyltransferases, glycine N-methyltransferase is a tetramer consisting of identical subunits. Glycine N-methyltransferase exhibits sigmoidal rate behavior with respect to AdoMet and hyperbolic kinetics with respect to glycine. The sigmoidal kinetics arises from the cooperative binding of AdoMet to the catalytic site residing on each subunit. In the three-dimensional structure, the four spherical subunits are arranged to a flat square shape with a large hole in the center. Each subunit has a long N-terminal segment protruded from the main body. The segments interact with each other in the central hole, and this interaction appears to cause cooperativity in binding AdoMet. The AdoMet-binding region folds into a typical α/β structure as other methyltransferases of known structure. However, glycine N-methyltransferase possesses an additional domain that surrounds AdoMet. The unique structure of the AdoMet-binding site suggests that folate coenzymes are bound at the same site. JOURNAL FREE ACCESSOn the basis of National Nutrition Survey in Osaka Prefecuture, an intake of vitamin E and polyunsaturated fatty acids were calculated according to the fat soluble nutrients from the Fat-Soluble-Nutrient Composition Table in Japan. 1. Vitamin E intake per capita per day was 8.3 mg alpha-tocopherol equivalent. 2. Intake of total fat, saturated fatty acid, mono-unsaturated fatty acid and poly-unsaturated fatty acid was 59.8 g, 17.0 g, 20.9 g and 15.1 g, respectively. 3. Ratio of vitamin E to poly-unsaturated fatty acid (E/PUFA) was 0.55, while it was 0.39, as PUFA was calculated as linolate on the bases of their peroxidizability.View full abstractDownload PDF (417K) -

JOURNAL FREE ACCESSStandards of reference range for all laboratory test values are mandatory. This study was designed to establish a reference range for blood vitamin B_1 levels, since the normal range has not been determined in the Japanese population. We found the Japan Committee for Vitamin Laboratory Standards, which was incorporated with the Vitamin Society of Japan and the Japanese Society of Nutrition and Food Science. We standardized whole blood vitamin B_1 levels using three HPLC techniques (post-column revesed-phase HPLC, pre-column reversed-phase HPLC, and pre-column GP-HPLC). The reference range was obtained in 54 volunteers administered a 1800 Kcal diet with 2 mg of vitamin B_1 (1.74 mg measured) daily to avoid marginal vitamin B_1 deficiency in the population. The range for each assay was 26〜47 ng/ml, 28〜51 ng/ml and 28〜56 ng/ml, respectively. Our data suggest that 26〜28 ng/ml is the lower limit of normal for whole blood vitamin B_1, but further studies in a larger population are needed to obtain more definitive results.View full abstractDownload PDF (333K) -
