Over the past 10 years, it has been growing to support the concept that ischemic stress produces the generation of active oxygen species and free radicals in the tissue. The ESR spin trapping technique was applied to heart mitochondria obtained from dog heart affected by ischemia, in order to detect the generation of active oxygen from the respiring mitochondria. ESR spectrum of active oxygen spin adduct was enhanced in ischemic mitochondria. The hydroxyl radicals were found to generate from mitochondria, by way of superoxide anion produced by electrons released from the electron transport system. The heart ischemia resulted in a decrease of membrane fluidity, with concomitant dysfunction of mitochondria and ATP depletion. The hydroxyl radicals were discussed to be the direct active oxygen species involved in the cellular damages. The antioxidant vitamins ; tocopherol, riboflavin, CoQ_<10>, were found to have the protective effect against the ischemic damages of heart mitochondria.

A method for simultaneous determination of retinol and all 4 tocopherols was developed using HPLC equipped with a reversed-phase column. Using this method, β-, γ-tocopherol and retinol, which had been difficult to separate by HPLC equipped with a reversed-phase column, can be separately determined. HPLC conditions were as follows ; column : YMC-PACK A 302 S-5 120A ODS, detector ; fluorophotometer (excitation wave length 340 nm and emission wave length 460 nm for retinol, excitation wave length 298 nm and emission wave length 325 nm for tocopherols), mobile phase : 65% isopropanol, sample solvent : 99.5% methanol, temperature : 30℃. Retinol and tocopherols in umbilical cord plasma, adult plasma, and liver of rats were determined using this method. This method can simultaneously determine retinol and each tocopherol homologue in biological samples.

Retinol and tocopherols in umbilical cord plasma, maternal plasma placenta, and vernix caseosa were determined by a modified method of HPLC. Retinol and all 4 tocopherols were detected in maternal plasma. Retinol and α- and γ-tocopherol were detected in cord plasma, and placenta. α- and γ-tocopherol were detected in vernix caseosa. The ratio of γ-tocopherol to α-tocopherol was higher in cord plasma than in maternal plasma. The retinol concentration in maternal plasma was positively correlated with the ratio of tocopherol to total lipids i cord plasma. The tocopherol concentrations in placenta were high in the cases with high tocopherol concentration in maternal plasma. α-tocopherol concentration in vernix caseosa was about 6 times of that in cord plasma. It is implied that vernix caseosa could be a source of α-tocopherol for fetus.

Bioavailability of β-carotene in a carotenoid preparation derived from Dunaliella bardawil was investigated in human adult volunteers, who were all healthy non-smokers. Sixty mg of β-carotene, in which all-trans form and 9-cis form were contained half-and half in this preparation, was administered once a day for 3 months. All-trans β-carotene levels in plasma, mononuclear cells, and platelets increased by 4-, 2-, and 3-fold the base-line after 2 weeks, respectively, which those in RBCs did not change during the experimental periods. 9-cis β-Carotene slightly increased in the biological samples after the administration. Neither side effects nor toxicity were documented in all the volunteers on the basis of the findings of the questionnaire and the clinical screening test.
