Thirty ppm or 3 ppm of mercuric chloride of protein bound form were mixed with commercial pellet diet and given to rats ad libitum for 4 months. Fifteen mg/kg or 1.5 mg/kg of methylcobalamin (CH_3-B_<12>) was orally administered daily throughout this period. Total mercury and methyl mercury levels in tissues, especially total mercury levels, increased in parallel with the mercury content in food and with the duration of feeding, but these increases were not related to the administration of CH_3-B_<12>. Furthermore, general toxicological findings were not deteriorated by CH_3-B_<12> but the toxic symptoms induced by inorganic mercury were found rather improved by the administration of CH_3-B_<12>. It is concluded from these results that biological methylation of inorganic mercury in vivo is not accelerated by oral administration of CH_3-B_<12>.

The effect of various vitamin D_3 analogues on chick embryonic duodenal alkaline phosphatase (AP) was observed using organ culture techniques. 1) AP was maximally induced by 1,25- (OH)_2-D_3,when 20-day old embryonic duodenum was cultured in the presence of 62.5 nM of 1,25- (OH)_2-D_3,1α-OH-⊿^<24>-D_3,1α, 24 [R], 25-(OH)_3-D_3,1α, 24 [S], 25-(OH)_3-D_3,1α, 24 [R]-(OH)_2-D_3,or 1α, 24 [S]-(OH)_2-D_3. 2) In the two different stereoisomers of 24-hydroxylated vitamin D_3,the Risomer showed higher activity on the AP induction than the corresponding S isomer. 3) No effect of vitamin D_3 analogues on sucrase activity was observed. 4) The inhibitory effect of cycloheximide on the induction of AP activity by 1,25- (OH)_2-D_3 was larger than that of actinomycin D.

To investigate the effects of α-linolenate on reproduction, pregnant rats were orally administered with 100 mg/rat/day of linoleate, α-linolenate or oleate during the first 14 days of gestation. Dams ingested linoleate showed a normal increment of body weight during pregnancy and their newborn rats were normal in the body weight, the histology of skin and function of transepidermal water barrier. On the other hand, the dams ingested α-linolenate and their newborn rats showed almost same properties to those ingested linoleate except for the function of transepidermal water barrier. The function of transepidermal water barrier of rats ingested α-linolenate were impaired. This disorder of barrier function may explain the reason why α-linolenate could not maintain normal reproduction.

The effects of vitamin E deficiency on the pituitary-gonadal function in the rats were examined by light and electron microscopy of the pituitary and testis and by measuring levels of pituitary-and circulating-luteinizing (LH) and follicular stimulating hormones (FSH) by radioimmunoassay, at 60 and 90 days after feeding of vitamin E deficient diet. In the vitamin E deficient rats, the FSH- and LH- secretory cells in the anterior pituitary gland appeared accelerated secretory function indicated by enlargement of cells, development of Golgi apparatus and cytoplasmic vacuoles and accumulation of secretory granules. However, the testis showed remarkable degenerating changes, such as atrophy of seminiferous tubules, appearance of polynuclear giant cells and sloughed germ cells in the tubular lumen and disruption of spermatogenesis. Serum LH and FSH levels were rised in the vitamin E deficient rats after 2 and 3 months and pituitary contents of LH and FSH were also increased in vitamin E deficient rats. These results indicate that the gonadal function was impaired by vitamin E deficiency, neverthless the pituitary function to secret gonadotrophin was accelerated. It is, therefore, suggested that the vitamin E deficiency gives a suppressive effect directly to the gonadal function and that the acceleration of pituitary function is due to impaired gonadal function.

To clarify the metabolic relationships between thiamin (B_1) and calcium especially in nervous systems of the rats, the following experiment was carried out. Male Wistar rats were divided at first into two groups and two synthetic diets (A, calcium deficient and B, normal diet) were given adlib. After 3 weeks on these dietary regimens, each group was divided again into two groups and four synthetic diets (A-1,calcium deficient-low B_1,B-1,calcium sufficient-low B_1,A-2,calcium deficient-B_1 sufficient, B-2,calcium-B_1 sufficient) were given for 5 weeks. Neurological symptoms occurred in the rat fed calcium deficient-low B_1 diet (A-1) and sometimes these rats died. When B_1 was administered to these rats, they recovered quickly. The growth, the body temperature, the blood pressure and the heart beat decreased in calcium deficient-low B_1 rats (A-1). B_1 levels in nervous systems and heart decreased in rats fed a calcium deficient-low B_1 (A-1) diet as compared with in calcium sufficient-low B_1 rats (B-1). Calcium levels in femur decreased in calcium deficient rats (A-1,2) and calcium level in telencephalon decreased only in rats fed a calcium deficient-low B_1 (A-1) diet. Magnesium levels in femur decreased in calcium deficient rats (A-1,2) and increased in blood in calcium deficient rats (A-1,2). These results suggested that calcium plays an important role in B_1 metabolism in nervous systems.
