Isopentenyl diphosphate and dimethylallyl diphosphate are known as active isoprene units for the biosynthesis of isoprenoids, the most diverse family of natural compounds containing >55,000 identified members. Isomerization between the two essential compounds for isoprenoid biosynthesis is catalyzed by isopentenyl diphosphate isomerase, and two types of the enzyme with no sequential homology have been found. Although the reaction mechanism of type 1 enzyme has been studied in detail, that of type 2 isopentenyl diphosphate isomerase, which curiously requires FMN and NAD(P)H for activity, is still unclear. The importance of isopentenyl diphosphate isomerase in isoprenoid metabolism, the biological distribution of the two types of enzymes, and the-state-of-the-art studies on the reaction mechanism of type 2 enzyme were introduced in this review.

This study was conducted to elucidate the nutritional role of biotin. The excessive intake of biotin for 29 days did not induce any clinical signs such as dermatitis, alopecia and body weight change, in immature mice. In the group given excessive biotin, the relative weight (liver weight/body weight) was decreased and the percentage of free biotin was increased in the liver in the 1% biotin-supplemented group, although no blood biochemical and histological changes were found. Also there was no change of biotin concentration in the liver between groups given <0.5% biotin in the diet. However, serum concentration and urinary excretion of biotin was increased, indicating that the extra biotin in serum is easily eliminated from urine. From these findings it is suggested that the excessive intake of biotin may not induce severe nutritional influences in young mice.

Vitamin B_<12> (B_<12>) is a coenzyme of methionine synthase (MS) and methylmalonyl-CoA mutase. It is especially important for MS functions in the methylation cycle that requires a supply from methyl donor S-adenosylmethionine (SAM). In order to identify a novel B_<12>-dependent gene expression pathway via DNA methylation, we looked for genes that were regulated by B_<12>. B_<12> deficient rats were used in this study. We searched for genes that were up- or down-regulated during B_<12> deficiency, and found that the Otx2 gene was influenced by B_<12> status in brain. We then analyzed the DNA methylation status of the promoter region of the Otx2 gene and identified a hypermethylated area in B_<12> deficiency. This study suggests that B_<12> status may have an impact on specific gene expression through DNA methylation.

The number of people taking large amounts of vitamin supplements is increasing in Japan. Taking large amounts of water-soluble vitamins is not considered risky because excess water-soluble vitamins are quickly excreted in the urine. In the present study, we investigated the clearance of water-soluble vitamins in Japanese. In the first week, six young female subjects ate the diet. In the second week, they ate the same diet with 6 times the recommended water-soluble vitamin dose based on the Dietary Reference Intake (DRI) for Japanese. In the third week, they ate the same diet without the vitamin mixture. The 24-h urine samples were collected every week. All water-soluble vitamins except vitamin B_<12> in 24-h urine samples were measured. The urinary excretion of nicotinamide metabolites, biotin, and ascorbic acid in the third week was same as that in the first week. Urinary thiamin in the third week was 4.4 times higher than in the first week, riboflavin was 3.4 times, vitamin B_6 metabolite 4-pyridoxic acid 2.0 times, pantothenic acid 2.4 times, and folic acid 3.1 times.
