The intracellular proportion of the pyridoxal 5'-phosphate (PLP) form of aspartate aminotransferase (AspAT) to the total enzyme in E. coli B cells and rabbit erythrocytes was determined by a newly devised method which based on selective inactivation of PLP form of AspAT by sodium borohydride added extracellularly. A large portion (80-90% and 75%) of the intracellular AspAT was in PLP form in E. coli cells and the erythrocytes, respectively. The saturation levels generally used to describe in vivo the proportion of the apo and holo vitamin B-6-dependent enzymes did not reflect the intracellular amount of PLP (holo) form of AspAT. The interaction between PLP form of AspAT and vitamin B-6 antagonists were studied in situ. The concentration of vitamin B-6 compounds in the erythrocytes and effects of the antagonists on the concentration of vitamin B-6 compounds were also determined after the erythrocytes were incubated with free vitamin B-6 compounds.

Changes in the contents of thiobarbituric acid reactive substances (TBARS), reduced glutathione (GSH) and flavin in the liver, mitochondrial ADP/O ratio and cytosolic enzyme activities, by the administration of 1.2 mmol/kg of trichloroethylene (TCE) or trichloroacetric acid (TCA) to mice were examined, and these changes were suppressed by the administration of flavin adenine dinucleotide (FAD). Then, the significant correlation between TBARS and ADP/O ratio, TBARS and flavin content, flavin content and ADP/O ratio, and TBARS and GSH content were observed. From these results, the liver injury by probably due to the lipid peroxidation induced by TCE or TCA is considered to be suppressed by the simultaneous administration of FAD.

Effects of folic acid on rat liver regeneration were evaluated by the activities of thymidylate synthase and thymidine kinase, which are rate-limiting enzymes of DNA synthesis in liver regeneration, and the contents of DNA, RNA and protein, and liver weight. By injection of folic acid immediately after partial (70%) hepatectomy, thymidine kinase activity was inhibited in the 24 h-regenerating liver, whereas thymidylate synthase activity increased in the 48 and 72 h-regenerating liver, The DNA and RNA contents and liver weight were also reduced in the 72 h-regenerating liver of folate-injected rats. These results suggest the stimulatory effect of folic acid on the induction of thymidylate synthase and the inhibitory effect on rat liver regeneration.
