Adult female rats were used to study the feeding technique to produce vitamin K deficiency syndrome and its influence upon the general condition, sexual cycle and pregnancy of the animal. In some of the rats fed on vitamin K-free diet, equipped with the coprophagy preventing tail cup and in all of the animals fed on the diet with 0.05% of sulfaquinoxaline, it was observed hypothrombinemia and hemorrhage of some degree. The sexual cycle and the potency for conception of the animal were tolerably maintained even in a vitamin K deficient condition, but the growth was impeded under the deficiency and the pregnant animal was more susceptible than non-pregnant one to the deficiency. About in a half of the pregnant animals fed on vitamin K-free diet, pregnancy could be kept on to the term by simultaneous administration of estrogen and progesterone, although the fetal development was seriously arrested. The administration of progesterone alone could not maintain the pregnancy to the term.

The serum concentrations of pantothenic acid in healthy subjects and patients with moderate or advanced pulmonary tuberculosis were measured by bioassay, and the determinations of serum ascorbic acid, serum cholesterol and urinary 17-hydroxycorticosteroids were carried out simultaneously. The pantothenic acid concentration was 179.0±50.0 mμg/ml in the moderate tuberculous patients and 183.9±35.8 mμg/ml in advanced cases. Neither of those concentrations was significantly lower compared with the level in the healthy subjects which was 216.3±43.3 mμg/ml. The concentations of serum ascorbic acid and cholesterol, and urinary 17-hydroxycorticosteroids in the patients were lower than those in the healthy subjects, respectively. No correlation of pantothenic acid to ascorbic acid, cholesterol or urinary 17-hydroxycorticosteroids was found.

Electroretinogram (ERG) and microscopic structure of retina were investigated in both cases of riboflavin deficient and recovered rats. In the liver of riboflavin deficient rat, the content of total riboflavin became about 40 per cent of nomal rat by six weeks feeding and succinic acid dehydrogenase activities were less in every layers of retina than those of normal ones. A marked decrease of succinic acid dehydrogenase activity was found in both layers of bacillary and outerlimiting membranes. The b wave of ERG showed about 35 per cent decrease and ratio of b and a wave became below than one half in the riboflavin deficient rat in comparison with that of normal one, and the difference of two groups disappeared after the administration of riboflavin to riboflavin deficient group accompanied by normalization of total riboflavin content of liver.

In this communication, it was described the influence of the amount of dietary protein or choline chloride upon the delay of the growth of young rats and the rise of P/N ratio of their hepatic lipid fraction, provoked by the successive dosage of the excess myoinositol. Observed results indicated that the elevation of casein contents in the diet from 22% to 30% was able not only to suppress the rise of P/N ratio of the liver lipids, but also to recover the growth suppressing effect of the excess dietary myoinositol. Elevation of choline dosage also suppressed the rise of P/N ratio. From these facts, discussions were made on the nutritional role of myoinositol.

The metabolic activity of inosine on the incorporation of riboflavin into rat liver xanthine oxidase system was studied. On oral administration of inosine in rats, statistical significance was scarcely observed in the growth promoting effect, but it was found that the incorporation of riboflavin into rat liver is markedly accelerated. Further, the increases of FAD and FMN in liver tissue were always demonstrated.

The CoQ content in the liver was examined after the administeration of various CoQ compounds to male albino rats. Of five CoQ compounds, CoQ_2,CoQ_6,CoQ_7,CoQ_9 and CoQ_<10>, only CoQ_7,CoQ_9 and CoQ_<10> raised the CoQ content after giving it intramuscularly in surfactant solution two times daily beforehand. After cell fractionation these increments of CoQ content were found mainly in the other fraction except mitochondria, but CoQ_9 and CoQ_7 raised it in mitochondria, too. In the rats with acute CCl_4 poisoning, the administration of CoQ_9 increased the CoQ content in the liver and its mitochondria more than that of CoQ_7 did. The paperchromatography of CoQ compounds in liver mitochondria showed that a corresponding CoQ increased respectively after the administration of various CoQ compounds.

Some experimental conditions were described on the reaction between thiamine and aldehydes affording hydroxyethylthiamine (HET) or its homologs. It was found that, as discribed by Miller and his coworkers, the best yield was attained when thiamine chloride hydrochloride was treated with 1.8-2.0 equivalents of alkali, and reacted with 20-40 equimolecular amount of aldehydes at 50-60℃ for about 3 hours. Several diacylated derivatives of HET and its homologs were synthesised by the reaction with acyl chlorides in acidic conditions.

Investigation was made on the synthesis and the physicochemical properties of thiochrome (Thc), thiochrome monophosphate (Thc-MP) and thiochrome diphosphate (Thc-DP). Rf values of Thc, Thc-MP and Thc-DP are 0.37,0.16 and 0.05,repectively, when a developing solvent consisting of 1M formate buffer, isopropanol and water (15 : 65 : 20) is used. Ultraviolet absorption and fluorescence spectra of their aqueous solution were measured at various pHs. It was found that absorption and fluorescence maxima of these compounds shifted to longer wave lengths with decreasing pH. In alkaline solution, absorption and fluorescence maxima of these compounds locate 370 and 450 mμ, respectively. On the other hand, fluorescence intensity is influenced with the phosphate-groups and the ratio of fluorescence intensity, Thc : Thc-MP : Thc-DP, is 10 : 8 : 9 in alkaline solution (pH9-12). The concentration of thiamine, thiamine monophosphate and thiamine diphosphate was determined by the well-known method without dephospholization. A reasonable result is obtained when bromine cyanide is used as an oxidizing agent, whereas an unreasonable result is obtained when potassium ferricyanide is used because of the quenching effect of potassium ferricyanide for the fluorescence of thiochrome derivatives.

Polished barley and polished rice were enriched with the saturated alcoholic solution of riboflavin tetrabutylate and the amount of riboflavin tetrabutylate in them was determined after they were washed with water for various periods of time. After washing for 5,10,15 and 20 minutes, the amount of riboflavin tetrabutylate which remained in the barley was 78.4,73.0,70.3 and 68.9 %, respectively, while, that in the rice was 39.3,30.4,28.6 and 25.0%. The difference of data between barley and rice was clearly remarkable. Accordingly, the enrichment of polished barley by riboflavin tetrabutytate may be performed easily, but that of polished rice must be further investigated.

Some biological and pharmacological properties of O-butyroylthiamine disulfide (BuTDS) were studied. BuTDS showed thiamine activity approximately equivalent to that of thiamine hydrochloride in thiamine deficient rats. The thiamine levels and the cocarboxylase activities of blood in dogs and in rabbits at intraveneous or intramuscular administration rose remarkably and its higher level was maintained for a long time. BuTDS produced moderate falls of blood pressure and depressed slightly the respiration of rabbit and myocardial activity of isolated heart of frog. The tension and the peristalisis of isolated intestines of rabbit decreased by BuTDS, while those of intact intestines increased. Acute toxicity of BuTDS in mice was much lower than that of thiamine hydrochloride.
