Method of separation and detection of several antioxidants in vitamin A oil or in vegetable oil by thin layer chromatography was investigated. Butylhydroxyanisol (BHA), dibutylhydroxytoluene (BHT), isoamylgallate (IAC) and nordihydroguaiaretic acid (NDGA) in oils were extracted with an equal part of methanol. BHA and BHT in the extract were successfully separated with one-dimensional chromatography using silicagel as chromatographic plate and chloroform, methanol or petroleum ether as the developing solvent. In the case of separation of IAG and NDGA, the two-dimensional chromatography, using polyamide powder as chromatographic plate and methanol and chloroform as solvents, were applied. After separation, 20% phosphomolybdic acid solution in ethanol and ammonia vapour were used as colour developing reagents. Micro-quantities of antioxidants in vitamin A oil or in vegetable oil could be successfully separated and identified, by the above procedures.

Two g of L-tryptophan was administered per os to patients of portal hyperension, who were classified to 2nd and 3rd groups by Imanaga, and tryptophan, kynurenine, kynureninic acid, xanthurenic acid and N-methylpyridone carboxamide in 24 hours urine were determined. It was revealed that the excretion of these tryptophan metabolites of the 2nd group was similar to that of normal, while in the 3rd group, the increased excretions of xanthurenic acid and N-methylpyridone carboxamide were observed. Therefore, the convertion rate of typrophan is supposed to be higher in the patents of 3rd group. The excretion of tryptophan metabolites was not changed before and after the portocaval end-to-side anastomosis, except just after the operation.

When riboflavin tetrabutyrate dissolved in oil or fat was preserved for 6 months in dark place, 93-87% of it remained unchanged. In case riboflavin tetrabutyrate was dissolved in highly oxidized oil or fat, its decomposition became remarkable. Therefore, to enrich oil or fat by the addition of riboflavin tetrabutyrate, it is recommended to adopt freshly prepared oil or fat.

In the colorimetric reaction of vitamin D with antimony trichloride reagent containing acetyl chloride, chemical isomerization of vitamin D by acetyl chloride and a role of acetyl chloride were studied. Vitamin D_2 in ethylene dichloride was able to be easily isomerized to isotachysterol by acetyl chloride at room temperature. When antimory trichloride reagent without acetyl chloride was added to isotachysterol, no color was produced. Therefore, it may be concluded that the isomerization of vitamin D_2 by acetyl chloride has no relation with this colorimetric reaction.

In the studies in vivo, rats were injected via femoral vein or intramuscularly with aqueous dispersion of vitamin A alcohol or acetate with Tween 80. At various time intervals the serum, liver and kindney were analysed for vitamin A alcohol and its ester. After the intravenous injection of vitamin A alcohol, the ester and alcohol in the serum were recognized, while in the liver the elevation of ester was pronounced but that of alcohol was insignificant. On the other hand, the increases of ester and alcohol were slight in the kidney. The similar results were obtained with the administrations of vitamin A acetate. After the intramuscular injection of vitamin A alcohol or acetate the elevations of alcohol and ester in the serum were recognized. However, after the administration of vitamin A alcohol, the alcohol in the serum was increased more significantly than the ester, while the elevation of ester was more predominant after the administratious of vitamin A acetate. In the liver the elevation of ester was predominant and no increase of alcohol was showm, after the administration of vitamin A alcohol or its acetate. In the kidney, a slight elevation of ester was recognized, but alcohol was not increased.

In the synthesis of O-benzoylthiamine disulfide (BTDS) by the benzoylation of thiamine disulfide, crystals showing 2 spots (Rf 0.57 and 0.75 on paper chromatography using acetic acid・n-butanol・water=1 : 4 : 5 system) were often isolated and they had a lower melting point and a higher growth-stimulating activity on L.fermensi than the authentic sample of Rf 0.75. The Rf 0.57 and 0.75 substances were separated fom the crystals by the means of similar chromatography and their activities on L.fermenti in the broth containing cysteine were compared with that of thiamine. The Rf 0.57 substance was found to have 1/2,the crude BTDS 1/40 and the authentic BTDS about 1/70 activity of thiamine, while the Rf 0.75 substance was proved to have no growth-stimulating activity. As the Rf 0.57 substance was reduced to thiamine and O-benzoylthiamine and the Rf 0.75 substance was reduced to O-benzoylthiamine by the incubation with cysteine or Na_2S_2O_3,it was suggested that the Rf 0.57 substance was monobezoate of thiamine disulfide and the Rf 0.75 substance having no microbiological activity was the purest sample of BTDS free from the Rt 0.57 substance

Histopathological studies of experimental pantothenic acid deficiency using rats have been performed. In cardiovascular system, moderate degenerative changes of muscle cells are found in the heart, though no marked lesion is encountered in both arterial and venous vessels. Slight or moderate atrophic and degenerative changes are demonstrated in mucosa of stomach and intestinal tracts. Marked centrolobular fatty degeneration and necrosis are observed in the liver. Marked arrest of spermatogenesis and atrophic changes are encountered in the testes. Hypophysis reveals moderate degenerative changes of glandular cells of anterior lobe. Marked scattered disruptions of follicles are found in thyroid. Adrenals exhibit bleeding and break-down of the cortex. In nervous system, both nerve cells and fibers are damaged. Degenerative changes are demonstrable in motor, sensory and vegetative neurons. Lowering of choline esterase activity and destruction of end-plate are also demonstrable.

It has been tested that dl-pantothenol prevents the development of deficiency of pantothenic acid in rats which were fed on its deficient diet. It has been confirmed that the development of the lesion has been interrupted by administration of 100μg of dl-pantothenol per day.

The effects of four kinds of inorganic salts at 10^<-4>, 10^<-3> and 10^<-2>M on the new flavin monophosphate producing activity of Rhizopus oryzae were investigated. Both of 10^<-4> and 10^<-3> M of MnSO_4,ZnSO_4 added in reaction mixture with the dry mycelium showed stimulating effects, but CdSO_4 inhibited the activity at higher concentration than 10^<-4>M. Each of CdSO_4,ZnSO_4 and MgSO_4 added in culture medium at 10^<-4> and 10^<-3> M inereased the activity of the mycelium, respectively, and at the same time these salts showed remarkable influences on the mycelial weight and organic acids production. These effects were reduced by the coaddition of riboflavin, generally.

Only 10^<-4> M of NiSO_4,added in culture medium, showed the stimulation. Fe_2 (SO_4)_3 inhibited the activity at any procedure, even if under the co-addition of riboflavin.

When the salts were added in the reaction mixture, NH_4VO_3 showed stimulative effects, on the other hand, BaCl_2 and K_2CrO_4 showed inhibitory effects at any concentration. The addition of 10_<-4> M NH_4VO_3,10_<-4> or 10_<-3> M BaCl_2,and 10_<-4> M K_2CrO_4 in the culture medium, respectively, stimulated the activity of mycelium. The co-addition of riboflavin reduced the inhibitory effect of 10_<-2> M NH_4 VO_3 or 10_<-3> M K_2CrO_4.
