A stereoselective synthesis of retinoid isomers, which are signal molecules of retinoid proteins such as rhodopsin, retinochrome, retinoic acid receptor, retinoid x receptor, and so forth, was devel oped from the β-ionone-tricarbonyliron complex 1. Treatment of the complex 1, prepared from the reaction of β-ionone and dodecacarbonyltriiron, with the lithium salt of acetonitrile afforded the nitrile 2, on the contrary, the reaction of the complex 1 with the lithium enolate of ethyl acetate and subsequent dehydration gave the ethyl 9Z-β- ionylideneacetate 9 predominantly. These compounds (2 and 9) were converted to the corresponding all-E- and 9Z- retinoic acids (7 and 16) via β-ionylideneacetaldehydes (3 and 12), respectively. The Peterson reaction of the aidehyde complex 3 with ethyl trimethylsilylacetate provided the 11Z-isomer 18 preferentially accompanied by the 11E-isomer 17. The ester 18 was transformed into the 11Z- retinal 30 in good yield without isomerization of double bonds. In order to investigate the interaction between the 9-substituents of retinal and apoprotein of rhodop sin, various 9-substituted 11Z-retinals 42 were synthesized by applying our developed method. The binding experiments of retinal analogs 42 with bovine opsin gave the new artificial pigments except for phenyl, (1-naphthyl)methyl, and tertiary butyl substituents in the alanogs 42, and the conforma tional analysis of chromophore in these pigments was also discussed.

It has been hypothesised that oxidative damage plays an important role in the pathogenesis of Alzheimer disease. Antioxidant substances, such as vitamins E (E, α-tocopherol) and C, have been shown to modify oxidative processes in laboratory experiments and animal trials. Epidemiologic studies have suggested that intake of antioxidant nutrients may be associated with a reduced risk of develop ing Alzheimer disease. A recent clinical trial indicated that E supplementation or/and selegiline sig nificantly slows the progression of Alzheimer disease. Since E was demonstrated to have far fewer adverse effects, it has become a noteworthy nutrient or drug for Alzheimer disease. In this review, we focus on the relation between Alzheimer disease and E, and summarise the presumed mechanisms of E pharmacological action (mainly, anti-oxidant function) and recent epide miologic and clinical studies of E against Alzheimer disease.

When mice were fed on extreme vitamin B_6 deficient diets, the ovalbumin-dependent antibody productions (IgE, IgG1)were significantly suppressed. In this case, the alanine aminotransferase activity and the aspartate aminotransferase activity in the liver were also significantly decreased. In the case of pyridoxine excess (6 mg% = about ten times of standard amount) with high protein (70% casein) diet, the ovalbumin-dependent antibody productions (IgE, IgG1) were also considerably sup pressed. These responses were much weaker in a normal casein (20% casein) diet. The administra tion of high doses of pyridoxine (6 mg%) resulted in the suppression of hepatic cathepsin B activity. Therefore, ovalbumin-dependent antibody productions (IgE, IgG1) were suppressed by the pyridox ine excess diet, especially in the case of a high protein diet.

Cobalamins in human serum were separated using HPLC gradient system after extraction with N- ethylmaleimide and citrate-phosphate buffer system. Each cobalamin analogue in the elute was con verted to cyanocobalamin (CN-Cbl) by the addition of KCN, and the cobalamin contents were deter mined by a microbiological assay with Lactobacillus delbrueckii subsp. lactis ATCC 7830. This method was applied to determination of serum cobalamin analogues in healthy volunteers. The ratios of each cobalamin analogue in the serum of the male volunteers were methylcobalamin (CH3-Cbl): 66.5 ± 12.3%, hydroxocobalamin (OH-Cbl): 17.1 ± 9.4% and adenosylcobalamin (Ado Cbl) : 14.1 ± 5.8%. The ratios for female volunteers were CH_3-Cbl: 72.4 ± 9.3%, OH-Cbl: 12.3 ± 8.6% and AdoCbl: 9.8 ± 5.8%.

In recent years, improvement of the food supply, changing eating habits, and so on cause the increase in obesity in Japan. On the other hand, average BMI of young women is around 20, which is judged as thin. Our groups are periodically carrying out a survey about their eating habits from 1972, especially niacin sources and intakes. In this paper, we report the result being carried out in 1998. Subjects are36 female college students (21-22 years old, 158 cm, 52 kg, 20.7 BMI). The subjects consumed self-selected foods for one day and a sample blood was taken from an arm vein at the next morning and the sample blood was used for measuring NAD and NADP. Energy intake was below the Recommended Daily Allowance (RDA) 1800 kcal, nevertheless, "niacin equivalent (NE) intake/ 1000 kcal" was 16.1 mg, which is three-times higher compared with the RDA, 6.3 mg/1000 kcal. However, energy intake dropped from 2155 kcal in 1972 to 1448 kcal/day in 1998, and NE intake also dropped from 31.3 mg/day in 1972 to 23.4 mg/day in 1998. A significant relationship between energy and NE intakes was observed. A main source of supply of NE was fish and meat low in fat. These were similar results obtained during recent 30 years. This is because Japanese eating habit that like low-fat and high-protein foods such as fish and low-fat-meats, do not fundamentally change. The blood NAD and NADP levels (nmol/ml of whole blood) were 26.6 and 12.5, respectively.
