    Serum levels of carotenoids, such as β-carotene, α-carotene, lycopene, cryptoxanthin, and zeaxanthin/lutein, and additionally the levels of retinol and α-tocopherol were investigated in the residents living in a southern area of Hokkaido. Serum β-carotene levels were lower for the residents with the history of hypertension and coronary heart disease, while the retinol and α-tocopherol levels were higher. Moreover, diastolic blood pressure was inversely associated with serum β-carotene levels by a multiple regression analysis. Serum levels of carotenes and xanthophylls were lower in males, while serum retinol levels were higher in males. In a age pair-matched analysis for the residents without habits of smoking and alcohol drinking, the levels of sex differences were 0.518 μmol/l (difference rate : 53.1%) for β-carotene, 0.052 μmol /l (40.0%) for α-carotene, 0.061 μmol/l (29.5%) for cryptoxanthin, and 0.271 μmol/l (8.7%) for retinol, respectively. In the multiple regression analyses, there was also a significant sex difference on serum carotenoid levels such as β-carotene having provitamin A acivity. It was suggested that a part of these sex differences might be caused by the certain physiological functions such as some sex-hormone related metabolism.

    To know the activity of niacin metabolism in persons of advanced age who lived in three different housing facilities (61-96 years ; n = 73), urine samples were collected and the urinary excretion of nicotinamide metabolites, N^1-methylnicotinamide (MNA), N^1-methyl-2-pyridone-5-carboxamide (2-Py), and N^1-methyl-4-pyridone-3-carboxamide (4-Py) was measured. As a result, the differences in these values between any of the three facilities, and between men and women were not observed when compared with each other in terms of μmol of creatinine. Furthermore, the urinary excretion of MNA, 2-Py, and 4-Py were almost the same between the persons of advanced age and women's students. The niacin equivalent intake in the persons of advanced age was around 28 mg per day, which was above the Recommended Dietary Intake. Therefore, the metabolic activity of niacin in person of advanced age was proved not to decrease when compared with young women.

    The effects of biotin deficiency on the concentration of "free biotin" (unbound and reversibly bound biotin) and "total biotin" (free and covalently bound biotin) and biotinidase activity in dams and their fetuses of mice were examined. Free biotin concentrations in the serum were significantly decreased at near term, after consumption of a biotin-deficient diet. The percents of free biotin to total biotin (free biotin rate) were 12% and 82% in the biotin-deficient and control groups, respectively. The liver free and total biotin concentrations in the biotin-deficient dams and their fetuses decreased significantly compared with the controls (50% in the dams and 60% in the fetuses). Free biotin rates were invariable in the biotin-deficient state, indicating continually the presence of the normal recycling of biotin. On the other hand, a large amount of biotin accumulated in the fetal skin and amniotic fluid in the control fetuses. No changes in the placenta biotin concentration were observed even in the biotin-deficient state. These findings suggest that biotin in these tissues plays a critical role in embryonic growth and differentiation. The liver biotinidase activity was low in the fetuses compared with their dams. However, there were no major differences in the serum or placenta between the biotin-deficient and control groups.

    Optimum conditions at lower concentration of NaOH for the thiochrome(Thc) formation from thiamin and 2- (1-hydroxyethyl) thiamin (HET) were studied. Regardless the concentration of K_3Fe(CN)_6 between 0.01% and 0.1%, 5% NaOH gave the highest yield of Thc from thiamin, and the Thc compounds from HET were increased with the decrease of NaOH concentration. In the 5% NaOH, the formation of Thc from each of thiamin and HET was highest and stable around the concentration of 0.025% K_3Fe(CN)_6. And in 21 foods, the values for thiamin and HET of the crude extract agreed well with the values of the conventional extract which is pretreated by a column chromatography. In Green tea and Jute mallow, the colored matters in the crude extract and the unwanted components which were detected after HET by HPLC were removed effectively by using a C_<18> Sep-pak cartridge.
