Action of vitamin E as a chain-breaking antioxidant has been studied and summarized from kinetic and mechanistic points of view. The rate and mechanism of scavenging of lipid peroxyl radicals were studied in homogeneous solution by measuring oxygen uptake, products formation, chemiluminescence and free radicals with electron spin resonance. The oxidation of phospholipid liposomal membranes and its inhibition by tocopherols were studied aiming at elucidating the action of tocopherols in the membranes. It was found that tocopherols suppress the oxidation of phosphatidylcholine liposomes by scavenging both aqueous radicals attacking from outside of the membranes and lipophilic radicals within the membranes. It was suggested that laterally tocopherols move fairly rapidly but that it gets less efficient for tocopherols to scavenge radicals as they go deeper into the interior of the membrane. The phytyl side chain had a profound effect in the retainment of tocopherols in the membranes but it makes tocopherols difficult to move between the membranes. 2, 2, 5, 7, 8-Pentamethyl-6-chromanol could penetrate and transfer between the liposomal membranes freely. Tocopherols also suppressed the oxidative hemolysis of erythrocytes induced by free radicals. It was found that tocopherol within the erythrocyte membranes was the most effective in suppressing hemolysis induced by aqueous radicals by scavenging chain-carrying radicals and breaking the chain propagation.

Bacteriorhodopsin is an only protein present in the purple membrane of a highly halophillic bacterium. The light energy which was absorbed in the retinal linked to the protein through the protonated Schiff base is used for proton ejection from the membrane and the subsequent uptake of proton from the opposite side of the membrane. In the present paper, the mechanism for the proton uptake was studied in terms of the changes in the electrostatic interaction among various protein residues and the Schiff base in the light-induced reaction process. The stabilization of the L' intermediate, whose decay is correlated with proton uptake, was governed by the pK_a=9 residue assigned to aspartic acid residue. The specific cationic residues, lysine and arginines, influenced the pK_a values of the aspartic acids and made the proton-donating system to the Schiff base active. A large UV spectral change associated with the formation of L' intermediate was revealed to be due to the unstacking of tryptophan residues from the retinal.

The bioavailability of vitamin B_1 in liquid preparations marketed for internal use as the so-called "Drink" was investigated in six healthy volunteers after oral administration by a cross over trial. The vitamin B_1 concentration in the blood after intake of the drink containing thiamin tetrahydrofurfuryl disulfide (TTFD) rose markedly and more rapidly than did after taking drinks containing thiamin nitrate. Vitamin B_1 excretion in the urine after taking the drink containing 5mg of TTFD was more than twice as much as that after taking a drink containing 5 or 10mg of thiamin nitrate. The bioavailability was compared between the drink and sugar-coated tablets of TTFD. The peak vitamin B_1 concentration was obtained 30 minutes after taking the TTFD drink and 1.5 hours after taking the sugar-coated tablet of TTFD. The peak value was the same for both preparations. The amount of vitamin B_1 excreted in the urine for 0-3 hours or 0-6 hours after taking the drink was significantly larger than that for the sugar-coated tablet.

Levels of total NiA-NH_2, NAD (NAD^++NADH), NADP (NADP^++NADPH), N^1-methylnicotinamide (MNA), N^1-methyl-2-pyridone-5-carboxamide (2-Pyr) and N^1-methyl-4-pyridone-3-carboxamide (4-Pyr) in whole blood were measured using women students. As the results, total NiA-NH_2 level was around 60nmol/ml whole blood, NAD level around 30nmol/ml, NADP level 10nmol/ml. When the value of [total NiA-NH_2-(NAD+NADP)] is presumed to be free NiA-NH_2, its level would be around 20nmol/ml whole blood. The levels of catabolic metabolite from NiA-NH_2 such as MNA, 2-Pyr and 4-Pyr in whole blood were trace or below the limit of detection.
