The incorporation of L-dehydroascorbic acid (DAsA) into human erythrocytes in vitro was investigated. DAsA was added to the mixture of 10% of erythrocytes and 90% of isotonic phosphate buffer (pH 7.4) at 37℃. The incorporation was finished within one minute and some of the incorporated DAsA was reduced L-ascorbic acid (AsA) by GSH and other reducing substances in the erythrocytes. The concentration of erythrocyte DAsA was proportional to that of the extracellular DAsA (100〜77% of external concentration) and the production of AsA was dependent on reducing substances in erythrocytes, mainly on GSH. When a small quantity of DAsA (5mg/dl) was added, the reduction of DAsA was due to GSH alone, but when large quantities of DAsA (100 mg/dl) was added, the reduction of DAsA was affected by other reducing substances.

The incorporation of L-dehydroascorbic acid (DAsA) into the pretreated human erythrocytes with N-ethylmaleimide (NEM), irreversible inhibitor to GSH, was investigated. DAsA was added to the mixture of 10% of NEM-treated erythrocytes and 90% of isotonic phosphate buffer (pH 7.4) at 37℃. Since DAsA was incorporated into NEM-treated erythrocytes within one minute in the same manner as non-treated erythrocytes, the permeation of DAsA into erythrocytes through the erythrocyte membrane was independent of the reduction of DAsA to L-ascorbic acid. The concentration of erythrocyte DAsA was proportional to that of extracellular DAsA, and reached an equilibrium state at 87% of external DAsA concentration within one minute. After 60 minutes, however, 81% of intracellular DAsA and 65% of extracellular DAsA were degraded and the concentration of erythrocyte DAsA was 47% of the external DAsA.

Metabolic conversion of thiamine-S-sulfonate (B_1SSO_3H) to the thiol form (thiamine) in the rabbit and rat was studied in comparison with those of thiamine disulfide (TDS) and cysteine-S-sulfonate (CySSO_3H). Intravenously administered B_1SSO_3H gave lower blood level of thiamine than TDS did. Blood cystine levels after the administration of CySSO_3H and CySH were almost the same. In the rat, the amounts of unchanged B_1SSO_3H, TDS and CySSO_3H excreted in the urine were 36,13 and 0%, respectively, of the intravenous doses. The brain, heart, liver, kidney, small intestine and blood of the rat were found to metabolize B_1SSO_3H and CySSO_3H in vitro. The above results suggest that B_1SSO_3H, like in case of TDS and CySSO_3H, was converted to the thiol form (thiamine), and that the conversion rate was the lowest of the three.

Thiamine decomposition reaction, catalized by the bacterial thiaminase I in the system containing pyridoxine as a second substrate was investigated. The transfer of the pyrimidine group from thiamine to nitrogen atom in pyridoxine was shown by isolating the reaction product and by determining its chemical structure. On the basis of wave length shifts with pH of the ultraviolet light absorption peaks caused by an electron-donating substitution, it was concluded that the product ionized in the phenolic hydroxyl group to give the dipolar ionic form, N-(4-amino-2-methyl-5-pyrimidinyl)-methyl pyridoxinium-3-phenolate, in neutral or alkaline solutions but took the cationic form in acidic solution. Chlorine was not detected from crystals of the product. These results suggest that the crystalline product is a betain-like inner salt. Two kinds of crystal forms, plates and needles, were observed, and the melting point of crystals (plates) was 204.5〜206.0℃ (decomp.). The NMR and IR spectra of pyrimidinyl pyridoxinium-3-phenolate were also presented.

An autoanalytical method for determination by the thiochrome fluorometry of thiamine (B_1), thiamine pyrophosphate (TPP) and thiamine disulfide (TDS) in blood (whole blood and plasma) or urine, has been studied using autoanalyser (Technicon Co.), and the following results were obtained. 1. The autoanalysis was accomplished almost satisfactorily by a) measurement of thiochrome fluorescence in aqueous state, b) reduction of TDS into B_1 with cysteine, c) isolation of TPP with ion-exchange resin, d) dialysis of the thiamines on the flow diagram of the analyser, and e) elimination of the most part of fluorescent substance contaminated in urine with ion-exchange resin. 2. Comparing with Permutit method (the conventional method), this method gives measured values with a slightly additive bias for blood, and with a rather negative bias for urine. However, the biases are small and almost constant for samples loaded or not loaded with the thiamines. Accordingly, the method can be applied to determine accurately increased or decreased concentrations of the thiamines in blood or urine. 3. In conclusion, since the method gives fairly good approximation of data to the conventional method, and enables to treat rapidly a lot of samples, it is a valuable method for some purposes, such as tracing thiamine levels in blood and excreted thiamines in urine after the administration of thiamine drugs, some clinical tests, and so on.
