The hepatic uptake of O, S-dicarbethoxythiamine (DCET) was studied by rat liver perfusion technique of Brauer in comparison with thiamine-HCl. The function of the excised liver during perfusion was evaluated from the speed of bile formation. It was found that the liver takes up free thiamine and DCET in the perfusate and it is more efficient with the latter. While the liver takes up free thiamine or DECT, it also liberates bound form of thiamine into the perfusate and the increase of bound thiamine is more pronounced with DECT. Part of DCET in the perfusate passes the liver without being taken up, and this derivative of thiamine may prove to be effective in increasing tissue thiamine levels after oral administration. Under inadequate conditions or when the liver cell functions are failing, no change of thiamine metabolism or increase of free thiamine in the perfusate would occur.

Investigations were made on the clinical application of S-carbalkoxythiamine (CAT) to the 37 cases, picked up from the 405 out-patient cases of our clinic, by measuring the pyruvic acid level in blood. Dicarbethoxythiamine (DCET) being most absorbable among CAT group, was administered per os on the said cases and observations on the clinical appearance, the liver function and the pyruvic acid level in blood were made. As a result, a close relationship between the administration of DCET and the pyruvic acid level in blood was observed.

The enzymatic synthesis of thiamine by Staphylococcus aureus FDA 209-P was studied. The active form of pyrimidine moiety was diphosphate of hydroxymethylpyrimidine. Both thiazole moiety and its phosphorylated derivative were active for the synthesis of thiamine by the acetone-dried powder of the microrganism or by the crude enzyme preparation extracted from the powder. Thiazole phosphate was the most active form of thiazole for the enzyme concentrate, purified by the treatment with ammonium sulfate and chromatography on DEAE-cellulose. The activity of free thiazole for the partially purified enzyme was negligible. The reaction products from pyrophosphate of hydroxymethylpyrimidine and thiazole phosphate were examined by paper chromatography and bioautography and proved to be di- and monophosphate of thiamine. The activity of the enzyme was accelerated by the addition of Mn^<2+> or Mg^<2+>. The enzyme was comparatively thermostable.

A preparation of apotryptophanase for the determination of pyridoxal-5-phosphate was studied. By using p-dimethylaminocinnamaldehyde as the color reagent, the indole produced by pyridoxal phosphate and apotryptophanase from tryptophan was found to be measurable with the sensitivity three times higher than the usual method. Escherichia coli K-12 strain was cultured in a solid medium composed of agar, peptone and bouillon, and the dry cells were obtained in good yield. Enzymatic properties of the cells were quite similar to those prepared by the ordinary liquid culture, and the activity was found to be constant at 5℃ for fifty days.

Among 205 strains of yeast cultured by the Takamine Laboratory, Sacch. rouxii 4028 and Candida albicans 4888 were found to be most available for the preparation of pyridoxal-5-phosphate. 0.2 mcg of pyridoxal-5-phosphate was formed in 1ml of the medium by the strains. The maximum amounts of pyridoxal-5-phosphate were found at about three days after inoculation in the cells, and at about four days in the medium. Therefore, it has been assumed that pyridoxal-5-phosphate may accumlate for the logarithmic phase in the cells and excrete into the broth in the stationary phase.

In conformity with Wirtschafter's method, lipoic acid was given intraperitoneally to male rats of Wistar strain, weighing about 2O0g. The animals were divided into 5 groups. Of these, 4 groups of 5 animals received 0.5,1.0,10 and 20 mg/kg of lipoic acid, respectively, and the remaining 1 group of 20 animals received 65 mg/kg. S-GOT, S-GPT, and total liver lipid were determined at 24 and 48 hour intervals. The histological examination, using hematoxylin eosin and sudan III stainings, was done in all animals. As controls, the animals were divided into 3 groups, i. e. 1) non-treated group, 2) saline administered group, and 3) NaHCO_3 solution administered group (NaHCO_3 was used as a solvent of lipoic acid). No significant difference was seen in S-GOT, S-GPT and liver total lipid between the lipoic acid administered groups and the controls. Histological study also revealed no remarkable change in the liver of rats given lipoic acid. None of the 20 rats receiving 65 mg/kg of lipoic acid died within 120 hours when they were sacrificed. These data were entirely different from those of Wirtschafter, who found lipoic acid was toxic. The cause of this discrepancy is still unknown.

On the determination of synthesizing activity of vitamin A ester, the several conditions were studied. The activity was recognized in the liver and pancreas homogenate of the rat, which were incubated at 38℃ for 30 or 60 mininutes with the substrate mixture containing vitamin A alcohol and palmitic acid in ethanol. The reactions were carried out adequately in the enzyme concentration containing 10 mg of pancreas tissue or 100 mg of liver tissue for 30μg of vitamin A alcohol in the reaction mixture. The ethanol in substrate solution have shown inhibitory effect at 0.5% of the concetrations for liver tissue and at 8.0% for pancreas tissue, The esterase reaction in the both tissues on vitamin A was found at pH 7.0 to be available for esterification, while at pH 8.5 for hydrolysis.

Vitamin A contents of 73 kinds of bread, used for the school lunch program in Osaka City and 59 kinds of flour, used for their raw materials were determined in the term of 1962 and 1963. The results showed that there were some of bread contained only a slight degree of vitamin A owing to the insufficiency of vitamin A in the flour. It was revealed that vitamin A added to the flour was considerably lost during storage, especially at high temperature and the presence of benzoyl peroxide, while viamin A added to the shortening oil was kept well. Thus the vitamin A enriching agent should be added to shortening oil, but not to the flour.

It was found that three new flavin compounds as FMN' or flavin cyclic 2', 5'-mononucleotide, flavinylglucoside mononucleotide of Rhizopus oryzae and flavin cyclic diphosphate of Aspergillus oryzae had no growth-promoting activity for Lactobacillus casei ATCC No. 7469,at the concentration ranged up to 4×10^<-8>M, but the significant growth responses of the acid hydrolysates of them were observed.

The presumptive test for the thiamine decomposing bacteria on nutrient agar plate is difficult. If there is a differential medium, it would be very advisable to identify the thiamine decomposing bacteria on presumptive test. In this point, the studies on colonies of the thiamine decomposing bacteria and other enteric bacteria on the media which contain soluble starch and pH indicator were made. In the soluble starch plate, Bacillus thiaminolyticus yields yellow colonies, while the other bacteria, such as Shigella flexneri, Escherichia coli 119,Salmonella paratyphiA, Aerobacter aerogenes, Bacillus subtilis, Proteus OX 19,etc. have not.
