Three methods, i, e. (I) using gas of both cyanogen bromide and ammonia, (II) spraying with cyanogen bromide and 5% sodium hydroxide and (III) spraying with 0.25% potassium ferricyanide in 5% sodium hydroxide solution were compared in the sensitibity at the detection of thiamine or its propyl disulfide on the paper partition chromatography. It was confirmed that method (I) was sensitive as much as method (III), while the method (II) was less sensitive than the others.

Thiamine disulfide monobenzoate (TDSMB) (II), a new derivative of thiamine, was prepared by the benzoylation of TDS (I) with equimolar benzoylchloride or sodium benzoylthiosulfate, followed by silicagel column chromatography. By recrystallizing from a mixture of benzene-ethanol, (II) was obtained as colorless scales consisting of C_<31>H_<38>N_8O_5S_2・1/2C_6H_6・H_2O and decomposed at 147-149°. The structure of (II) was confirmed by UV, IR, NMR spectra, elemental analysis and determination of benzoic acid. By treating (II) with alumina in CHCl_3,the partial disproportionation to (I) and BTDS (III) was observed by thin layer chromatography.

^<14>CH_3-Methylmethionine sulfonium iodide was administered on mice orally or intravenously, and its distribution in body was detected by autoradiographical technique. In the mucosa of digestive tract and liver, high density of radioactivity was detected. In the gastric and intestinal mucosa, the radioactivity was highest 4 hours after the administration. A small radioactivity was found in kidney and adrenal gland. These results agreed with the fact that methylmethionine sulfonium halide was capable to be the methyl-donor to both methylhistamine in gastric mucosa and lecithin in liver.

Uptake of pyridoxine by ascites-sarcoma cells in vitro has been investigated. Pyridoxine was uptaken against concentration gradient at low initial concentrations of pyridoxine and accumulated in the cells as phosphorylated forms of B_6. The uptake of pyridoxine by ascites-cells was greatly enhanced in the presence of glucose but it was diminished by the addition of either 2,4-dinitrophenol or sodium fluoride with glucose. 4-Deoxypyridoxine also inhibited the uptake of pyridoxine in the cells. Incubation of ascites-cells with glucose and labelled pyridoxine resulted in accumulation of labelled pyridoxine in the cells. Similarly, the accumulation was decreased by the addition of each 2,4-dinitrophenol or sodium fluoride. Furthermore, in the presence of vitamin B_6 phosphates and 4-deoxypyridoxine, labelled pyridoxine uptake was inhibited. These results support the conclusion that pyridoxine was uptaken in ascites-sarcoma cells by an energy-associated process.

Uptake of pyridoxal and pyridoxal phosphate by ascites-sarcoma cells in vitro, has been investigated. Pyridoxal was quickly uptaken against concentration gradient and accumulated in the cells as pyridoxal phosphate. The uptake of pyridoxal was markedly diminished either in the absence of glucose or by decrease of the temperature. Pyridoxal uptake was also diminished in the presence of 2,4-dinitrophenol and sodium fluoride. In contrast, the uptaken amount of pyridoxal phosphate in the cells was smaller than that of pyridoxal, but increased proportionately with the external pyridoxal phosphate concentration. The uptake of pyridoxal phosphate was also influenced with the addition of glucose and metabolic inhibitors and temperature of incubation. Vitamin B_6 analogues, especially 4-deoxypyridoxine inhibited the uptake of pyridoxal and its phosphate by the cells.

It was clarified in the previous report that pyridoxal and pyridoxal phosphate were uptaken by the ascites-sarcoma cells and these uptake processes depended upon temperature, properties of incubation medium, and energy supply. In order to clarify the relationship between the uptake of these vitamins and intracellular metabolism, intracellular lactate, phosphate, and ATP were determined under the conditions of uptake of these vitamins. Intracellular lactate and ATP corresponded almost in parallel with the uptaken amount of the vitamins. Pyridoxal phosphokinase activity was found in the soluble fraction of homogenate of the cells but not in the membrane fraction. The enzyme activity was inhibited moderately by 4-deoxypyridoxine.

The preventive effects of tocopherol-homologues on the toxicity of oxidized oil to carp have been investigated. Carp fingering were fed with artificial diets containing oxidized ethyl ester of fatty acids prepared from pollack liver oil with or without tocopherol-homologues (see TABLE 1). Muscular dystrophy was observed in the group receiving oxidized oil without tocopherol in 60 days and the ratio or contraction amounted to 40% at the end of experiments. Such symptom, however, was effectively prevented by the addition of d-or dl-α-tocopherol whereas d-non-α- tocopherol was less effective. In the histological observation, especially of intestinal tract, the group receiving non-α-tocopherol showed excellent results on preventing intestinal tract injury. The concentration of total tocopherol of the group 3 was remarkably lower than the others, whereas the concentration of sterol of the group 6 was clearly lower than the others. From these observations, it is suggested that tocopherol-homologues may have different effects on the toxicity of oxidized oil to a living body.

Thiamine (I) and chloroethylthiamine (II) were separately estimated on thin layer chromatography by oxidatation of the compounds to their thiochrome forms. By this method, the radioactivities of ^<35>S-thiamine in chicken blood after oral administration of ^<35>S-chloroethylthiamine was counted, and it was observed that about 3% of (II) orally administered to chicken, were converted to (I).
