A new derivative of pyridoxine was formed in the incubation mixture containing uridine 5'-diphosphoglucuronyltransferase from rabbit liver, uridine 5'-diphosphoglucuronic acid and pyridoxine. This derivative was isolated and assumed to be pyridoxine β-glucuronic acid-like substance from data, viz., mobilities on paper chromatography and paper electrophoresis, ultraviolet absorption spectra at acidic, neutral and alkaline pH, and products by hydrolysis with β-glucuronidase from Helix pomatia.

The antioxidant effects of riboflavin on the lipid peroxidation and respiratory control of mitochondria were studied. Rats were fed on regular diet, 3 mg% riboflavin free-form (free-B_2)-supplemented diet, and 3 mg% riboflavin tetrabutyrate (B_2-But)-supplemented diet respectively. The mitochondria was prepared from rat liver after 8 weeks feeding. Ultraviolet irradiation upon the mitochondrial suspension resulted in formation of lipid peroxide and uncoupling of respiratory control. The mitochondria suspension prepared from rats fed on riboflavin-supplemented diet, prevented the above mitochondrial damage. The addition of riboflavin to the mitochondrial suspension following ultraviolet irradiation could restore the lipid peroxide content and the uncoupling damage of respiratory control. The anti-oxidant effect of B_2-But was higher than that of free-B_2 in the in vivo and in vitro administration study mentioned above.

Myocardial cells obtained from fetal mouse heart beat spontaneously and rhythmically in monolayer cultures. Asymmetric disulfide derivatives of thiamine, such as thiamine propyl disulfide (TPD)(5 to 50μM) or thiamine tetrahydrofurfuryl disulfide (TTFD)(5 to 50μM), and a SH reagent, p-chloromercuriphenyl sulfonic acid (PCMPS)(50μW), stopped the spontaneous beating of cultured myocardial cells. The stopping induced by PCMPS was reversed within 19min on addition of SH protective reagent, dithiothreitol. On the other hand, the stopping induced by TPD or TTFD was not reversed within 3h on addition of SH protective reagents, such as dithiothreitol, glutathione (reduced form) or L-cysteine, but the normal beating was recovered within 20h. When TPD or TTFD was added in presence of SH protective reagents on cultured myocardial cells which had been preincubated with the SH protective reagents, the addition of TPD or TTFD did not stop the spontaneous beating.

Escherichia coli mutant strains, 64-21 and 42-11,both requiring either high concentrations of hydroxymethylpyrimidine or low concentrations of thiamine, have been produced by further mutation of a mutant strain, 26-43,requiring the thiazole moiety of thiamine. The concentration of hydroxymethylpyrimidine required for a half-maximal growth of the respective organisms was approximately 10,000 and 1,000 times higher than that of thiamine required for the same extent of growth. Experimental results showed that phosphomethylpyrimidine kinase activity was deficient in these mutant cells and that they could not take up hydroxymethylpyrimidine into the cells in a significant amount when it was supplied at the concentrations less than 0.01mM. Another mutant, 70-23,lacking in phosphomethylpyrimidine kinase activity, could not also take up hydroxymethylpyrimidine. Some role of phosphomethylpyrimidine kinase in the uptake of hydroxymethylpyrimidine in Escherichia coli was discussed.
