    Administration of isoniazid to mice induced a remarkable inhibition of liver cytosolic aspartate aminotransferase. Experimental findings indicated that the inhibitor was not isoniazid itself but probably its metabolite. DL-Hydrazinosuccinate, a model compound of the metabolite, was synthesized since the metabolite was not successfully isolated. The compound was found to be even more inhibitory. The inhibition by the compound proceeded time-dependently and was not readily released. D- and L-Hydrazinosuccinate were synthesized, respectively, for further analyses of the inhibition mechanisms. D-Hydrazinosuccinate was found to behave as a slow-binding inhibitor via a single-step reaction mechanism and gave Ki of approx. 3 nM. L-Hydrazinosuccinate was a more powerful inhibitor with Ki of approx. 0.2 nM, and was found to be a slow, tight-binding inhibitor and interact with the enzyme through consecutive reversible steps. Administration of L-hydrazinosuccinate to mice produced in vivo a potent and long lasting inhibition of cytosolic aspartate aminotransferase in the liver and kidney in a relatively specific manner. The administration of this compound also caused a remarkable accumulation of citrulline in the liver and plasma, which was explained as a result of both specific and non-specific effects of the inhibitor.

    The biosynthetic pathways of riboflavin were shortly reviewed. The involvement of 4-carbon compound essential for formation of the o-xylene ring in riboflavin molecule was studied. Stimulation of riboflavin synthesis with ethanol in Ashbya gossypii indicated a clearcut precursor-product relationship between maximum accumulation of riboflavin and diacetyl plus acetoin during the resting cell incubation. The tracer experiments with [U-^<14>C]-glucose and -diacetyl in Eremothecium ashbyii provided evidence that the 4-carbon unit is not supplied with the ribityl fragment of 4-ribitylamino-5-amino-2,6-dihydroxypyrimidine. but with diacetyl. Furthermore, the experiments with a N-189-10A strain of the several riboflavin deficient mutants of yeast (Saccharomyces cerevisiae) which lacks GTP cyclohydrolase showed a specified relationship of diacetyl to riboflavin formation. These results indicate, thus, a strong possibility that diacetyl is the 4-carbon compound essential for formation of riboflavin although it remains to be solved whether its involvement in riboflavin formation is enzymatic or non-enzymatic.

    Fatty acids in muscles in Pigs of Yorkshire and Touyuen were examined. A significant difference was observed in PUFA such as linoleic and arachidonic acids between both breeds. Touyuen pigs had a higher PUFA content (53.3%) including 5.7% linoleic and 1.6% arachidonic, as compared with that of Yorkshire ones in whom PUFA, linoleic and arachidonic were 44.8%, 2.0% and 0.8% respectively. In muscles of the hybrid pigs between both breeds, the values of PUFA, linoleic and arachidonic were in between. Thus, muscular fatty acid constituents seem to be hereditarily in part controlled. Rabbit muscles has a higher content of linoleic than pig ones. Marbled beef had less linoleic and arachidonic than ordinary one.

    Determination of urinary NiANH_2 and MNA was investigated. A fluorometric-HPLC method was developed for determining NiANH_2 in urine. In 18 single urine samples of male, 1.22-49.2ppm of MNA and 0-0.7ppm of NiANH_2 were detected. Further, the daily variation of urinary NiANH_2 and MNA in 3 subjects was examined. Almost the same excretion patterns were obtained from 3 subjects, and the highest concentration of MNA and also NiANH_2 Were detected in the urine collected in the morning (8:00, 8:20 and 9:30). A good correlation was observed in the concentrations of MNA and NiANH_2 in all the urine tested.

    2-Methyl-4-amino-5-hydroxymethylpyrimidine(OMP) was as effective as thiamin for the growth of Euglena gracilis Z, but 4-methyl-5-β-hydroxyethylthiazole (Th) was not. Quinolinothiamin, heteropyrithiamin and pyrithiamin also sup-ported the growth. However oxythiamin, amprolium, pyrimidinylthiophenol and pyrimidimylcysteine were not effective. Simultaneous addition of 1μM quinolinothiamin and 50μM Th was effective for accumulation of thiamin in the cells. The results indicate that pyrimidine moiety is supplied by the cleavage of thiamin analogues for normal growth of thiamin-deficient Euglena cells.
