    The in vitro effect of D-arabo-ascorbate-iron(II) complex ([DaA-Fe^<2+>]) on bacteria was investigated using eight bacterial species. [DaA-Fe^<2+>] was bactericidal toward all species examined. The bacteria were sensitive to [DaA-Fe^<2+>] in this order from greatest to least: Bacillus subtilis, Lactobacillus casei, Salmonella typhimurium, Escherichia coli, Staphylococcus aureus, Proteus vulgaris, Micrococcus flavus and Morganella morganii. Mechanism of the bactericidal action of [DaA-Fe^<2+>] was investigated with E. coli, B. subtilis, L. casei and S. aureus. Bubbling of nitrogen gas through the reaction mixture inhibited the action. Addition to the reaction mixture of reducing agents, chelating agents, radical scavengers, catalase and superoxide dismutase inhibited the action. Transition metal ions enhanced the action. The findings indicate that free radical mechanism is involved in the action and reactive oxygen species are responsible for the bactericidal action. In [DaA-Fe^<2+>]-treated bacterial cells, adsorption of T-phages was inhibited, suggesting the alteration of cell surface by [DaA-Fe^<2+>].

    The effects of oral administration of L-ascorbic acid (AsA) and dehydro-L-ascorbic acid (DAsA) on ODS rat development were examined. AsA or DAsA was administered orally at 2mg/0.5mL ion-exchanged water/day. No difference was observed in body weight changes during the observation period between the AsA and DAsA groups. The animals were anatomized on Day 21, and various component analyses were performed. The total vitamin C levels in the brain and liver and serum alkaline phosphatase activity were significantly lower in the DAsA group than in the AsA group (p<0.01). The malondialdehyde concentration, which is an index of serum lipid peroxide level, was significantly higher in the DAsA group than in the AsA group (p<0.05). The hydroxyproline level, which is an index of the collagen content, was measured in the sternum (xiphoid process) and auricle, but no difference was noted between the two groups. No difference was noted in the systolic blood pressure or heart rate on the day before autopsy between the two groups.

    Glutamate dehydrogenases from thermophiles and hyperthermophiles have been reported to be expressed as inactive forms in a mesophile Escherichia coli and differ in their form from the native enzymes. We have recently found that the recombinant Pyrobaculum islandicum glutamate dehydrogenase (Pis-GDH) expressed in E. coli hardly exhibits the activity, and that the heat treatment of the inactive enzyme at 90℃ or the addition of 5M urea to the inactive enzyme solution dramatically activated to the activity level comparable to that of the native enzyme. Our small-angle X-ray scattering measurements showed that the size of inactive molecule markedly decreases and the arrangement of quaternary structure changes accompanying the activation without change in the molecular weight of the enzyme. This finding indicates that the subunit rearrangement - i.e., a change in the quaternary structure of the recombinant hexameric Pis-GDH - is essential for creation of the active enzyme.
