    The total and free pantothenic acid levels of the tissues were determined in mouse hepatoma MH-134,Sarcoma-180 (solid and ascites types) and normal mouse liver. Low values (12.5% of normal level) of total pantothenic acid were shown in hepatoma MH-134,while both types of Sarcoma-180 were almost same to the value of hepatoma MH-134. On the other hand, high percentages of free pantothenic acid were noted in hepatoma MH-134 and solid type of Sarcoma-180. It seemed that the reason for this high percentage was due to containing considerable amount of dead cells in their tissues. The values of coenzyme A in livers which prepared from the mice bearing Sarcoma-180 ascites tumor were found to be lower in comparison with that of the normal. The injection of the crude toxohormone, prepared from Sarcoma-180 by Nakahara's method, resulted in decline of coenzyme A level of the normal liver. These facts may suggest the action of toxohormone on pantothenic acid metabolism.

    In order to know the α-tocopherol requirement, young rats were fed with a basal diet containing 10% of hydrogenated methyllinoleate, supplemented with 0,50,100 or 500μg per day of α-tocopherol. The result of haemolysis test after 5 weeks feeding suggested that the α-tocopherol requirement of rat was 100 to 500μg per day. Similar experiments were done, using 0-20% of methyllinoleate instead of hydrogenated methyllinoleate in the former basal diet. By increasing the methyllinoleate in the diet, haemolysis became higher than those with saturated oil diet. On the other hand, in the diets supplemented with 80 μg of α-tocopherol per day, the lowering of liver tocopherol content and increasing of the conjugated double bond content of adipose tissue were recognized in either saturated or unsaturated fats diet.

    The equiliblium constants of pyridoxal- and pyridoxal phosphate-Schiff bases are determined by an optical method and the pH dependence of the Schiff bases is analysed. These results suggest that aromatic pyridoxal-Schiff bases are unstable compared with aliphatic pyridoxal-Schiff bases and the stability of pyridoxal phosphate-Schiff bases decreases at high pH value. These and the other factors affecting the stability of the Schiff bases are discuseed. These compound served as a key intermidiate in the synthesis of pyridoxal-5-phosphate.

    Intraperitoneal1y administered TTFD (thiamine tetrahydrofurfuryldisulfide) and TPD (thiamine propyldisulfide) showed a strong inhibitory effect on carrageenin induced rat paw edema, while the strong anti-inflammatory effect was not observed by oral administration of TTFD and TPD. On the other hand, BTMP (S-benzoylthiamine monophosphate), DCET (O, S-dicarbethoxythiamine) and thiamine hydrochloride hardly inhibited carrageenin edema even with intraperitoneal route. None of TTFD, BTMP, DCET and thiamine hydrochloride showed a significant suppressive effect on ultraviolet erythema on guinea pigs and croton oil granuloma pouch in rats.

    This experiment was done to study the effects of dietary essential amino acid deficiencies on the distribution of flavin compounds in rat liver. Weanling male Wistar strain rats were fed with experimental diets for 10 days. Amino acid composition of experimental diet was based on the Rama Rao's pattern and was equivalent to 10% protein level. Total riboflavin contents in liver decreased in all essential amino acid-deficiencies, especially in the methionine-deficiency. But total riboflavin per 100 mg nitrogen of liver decreased significantly only in the threonine-and tryptophan-deficient group. The ratio of FAD, FMN and riboflavin contents in liver of amino acid deficient rats showed statistically insignificant difference compared with control group except threonine-deficient group. In the threonine-deficient group, FAD decreased and FMN increased significantly.

    By the reaction of O, S-disubstituted thiosulfites with thiamine in thiol form, a series of asymmetric thiamine disulfide derivatives were obtained in satisfactory yield. Thiamine propyl-, butyl-, isobutyl-, octyl- and tetrahydrofurfuryldisulfide were synthesized by the new method of disulfide formation. Eight of O, S-disubstituted thiosulfites thus synthesized are new compounds.

    For the purpose to clarify the reason of a characteristic phenomenon in the growth-OMP curve(reported previously), inoculum size of Leuconostoc mesenteroides and incubation period for the growth of the bacteria in the presence of various concentration of OMP were studied. It was observed that heavier inoculum size showed the refractions at higher concentration of OMP. The double refractions were observed after incubation for more than 68 hours even if absence of D-alanine in the medium, while after 24 hours incubation with DL-alanine. When higher concentration of OMP added to the medium, the bacterial growth was inhibited more in the early stage of incubation. However, the growth was stimulated more in the later stage of incubation with higher concentration of OMP between those concentrations at the first and the second refractions. It was presumed from the above result that OMP might be transferred to some unknown substance which could substitute vitamin B_6 function in the condition, in which stimulation of the bacterial growth was occured.

    A new type of antithiamine compound, deoxythiamine(DOB_1), inhibits the growth of thiamine requiring microorganisms, such as Lactobacillus fermenti 36 or Kloeckera apiculata and competitively antagonizes thiamine (B_1) (Vitamins 22,200 ; 23,213,1961). In order to clarify the mechanism of action of this compound, the influence of DOB_1 on B_1 uptake by L. fermenti was observed using labeled B_1-^<35>S-HCl. Cells absorbed 73〜92% of B_1 under our experimental conditions. When DOB_1 was added to the incubation medium, the amount of absorbed B_1 decreased proportionally with the increase of DOB_1. The ratio of DOB_1 to B_1 to yield 50% of inhibition was 6 : l. Competitive inhibition was observed between DOB_1 and B_1. A comparison of the antagonistic activity by DOB_1 with several other antithiamine compounds was done and the order of their inhibition indices was as follows : pyrithiamine (2)>DOB_1 ≒ deoxymethylthiamine ≒ imidazolethiamine (6)> amprolium ≒ thiamine sulfuric acid ester (30). It was interesting to observe that the degree of inhibition of B_1 uptake by the antithiamine compounds was similar to their effects on cell growth.

    In the course of the studies on the metabolic pathway of TDS derivatives, absorption mechanism of TDS, BuTDS, and BTDS was investigated. Vitamins were administered in ligated rabbit intestine, and transferred thiamine derivatives in mesenteric venous blood were determined sparately with acylthiochrome fluorometry. In the venous blood collected immediately after administration of acyl TDS, main part of the total thiamine was found to be administered form. With the lapse of the time, the ratio of thiamine to total thiamine was gradually increased. These results showed that acyl TDS was absorbed from lumen with being administered form and metabolized to thiamine in gut wall and blood. Slightly different absorption mechanism of TDS was also discussed.

    Although TGD has long been assumed as the intermediate product in the reaction of TDS and GSH, its isolation has not been achieved. In this study, TGD and its O-benzyl derivative, BTGD, were successfully synthesized and isolated as the colorless crystals, mp. 179-181℃ (decomp.) and 172-174℃ (decomp.), by the reaction between TDSO (BTDSO) and GSH. The newly synthesized compounds were confirmed by IR-, UV-spectrum, PPC, PEP and elementary analysis. TGD was tested for prevention of thiamine deficiency in Uroloncha domestica and was confirmed to possess the same efficacy as that of thiamine. The significance of the compound as the intermediary metabolite of TDS derivatives, was also discussed on the basis of the equilibrium constant estimated in the exchange reaction between the compound and GSH.

    Uptake of thiamine hydrochloride-^3H (B_1-^3H) and O-benzoylthiamine disulfide-^3H in various organs of the mice with oral administration has been studied by microautoradiogram. The tissue obtained from the experimental animals were fixed in cold absolute alcohol at -2O℃. Paraffin sections were made and dipping method was employed for autoradiographic technique. Both B_1-^3H and BTDS-^3H were mostly absorbed from the upper half of the small intestine, though a small amount of BTDS-^3H was absorbed from the stomach. BTDS-^3H passed out the absorption epithelium more quickly than B_1-^3H. The liver, heart, kidneys, pancreas, adrenals, hypophysis, testes, muscle, thyroid gland, spleen and lungs, etc. revealed marked the uptake of both B_1-^3H and BTDS-^3H. But the uptake of BTDS-^3H in these organs was more marked than it of B_1-^3H. Distribution pictures of grains in general viscerae were described and demonstrated in 40 pictures. Throughout this observation, following findings were pointed out. (1) In the cells the grains were found in cytoplasm, nucleus and nucleolus. (2) Marked uptake of B_1-^3H and BTDS-^3H was found in β-cells of Langerhans islets of the pancreas, cortical cells, especially in zona fasciculata of the adrenals, spermatic cells in the testes, follicular epithelia in thyroid gland besides in liver cell, muscle cells of the heart, and urinary epithelium of the kidneys, etc.
