A method for partial synthesis of a cobalamin analogue containing 2-amino-2-methylpropanol (S102) in place of 1-amino-2-propanol, one of the strongest vitamin B_<12> antagonists, was investigated using cobyric acid and αribazolee phosphate, which had been obtained by hydrolyzing CN-B_<12> with HCl according to Bonnet's method as starting materials. The intermediates, cyc. α-ribazole phosphate and phosphate-diester of 2-amino-2-methylpropanol and α-ribazole, as well as the final product, S102, were identified by paper ionophoresis and UV・visible absorption spectra in comparison with those of the authentic preparations given by Prof. Friedrich.

In the high-performance liquid chromatographic (HPLC) determination of vitamins A, E and ubiquinone (UQ) in the biological materials, the microparticle columns were used in place of the porous layer beads columns which had been used in our previous reports, and then higher theoretical plate numbers, better resolution factors and less coefficient of variation on the microparticle columns were obtained. This HPLC method could be easily applied to the determination of vitamins A and E, but not to UQ in serum, in which β-carotene like substances interfered the UQ peaks. Furthermore, the simple HPLC method was established for the simultaneous determination of vitamins A and E using NH_2 microparticle column in combination with on-column concentration.

The influence of vitamin E on platelet count of rats associated with decompression was studied. In the first study, when rats stayed at 6 absolute atmosphere (ATA) for 2 hours and were decompressed in 15 minutes, platelet count fell from 56.13±4.14×10^4/mm^3 to 39.06±12.98×10^3/mm^3 after decompression. In the second study, rats were divided into 3 groups: group A rats were fed vitamin E-deficient diets (d-α-tocopherol 5mg/kg), group B rats were fed the control diets (dl-α-tocopheryl acetate 20mg/kg), and group C rats were the vitamin E diets (dl-α-tocopheryl nicotinate 234mg/kg). These groups were fed for 3 months. Platelet count fell from 64.84±3.70×10^4/mm^3 to 56.66±5.03×10^4/mm^3 after the decompression in group A. Platelet count fell from 56.28±3.12×10^4/mm^3 to 45.62±13.84×10^4/mm^3 in group B. But there was no significant changes of platelet in group C. In the third study, survival rate after decompression was studied. The highest survival rate was observed in group C (50.0%) and the lowest survival rate was in group A (16.7%). These resutls suggest that platelets changes after decompression are inhibited by vitamin E.

1) A vitamin B_<12>-producing bacterium, Protaminobacter ruber formed a considerable amount of B_<12>-dependent apo-methionine synthetase when grown on the cobalt-deficient medium containing 1,2-propanediol as carbon source and supplemented with sodium succinate and serine. Since more apo-enzyme was synthesized at early phases of the bacterial growth, the enzyme for affinity chromatography was extracted from such early phase cells. 2) With the affinity chromatography using a B_<12>-Sepharose, methionine synthetase from P. ruber was successfully purified in good yield and in high purity (specific activity: ca. 30 nmol/mg/min). 3) The behaviors in the affinity chromatography seem to support the view that the methionine synthetase from P. ruber catalyzed N^5-CH_3-THF: homocysteine transmethylation and CH_3-B_<12>: homocysteine transmethylation.
