The amount of pantothenic acid which appeared in the urine of the rat after the oral and intraperitoneal administration of D-pantothenol or calcium D-pantothenate were determined by means of microbiological assay with Lactobacillus arabinosus. The amount of pantothenic acid excreted in the urine within 24 hours after intraperitoneal injection of D-pantothenate to the rat was larger than that of D-pantothenol injection. However, the total amount of pantothenic acid excreted in the urine during a week after the administration was exactly the same in both cases. It suggests that the utilization of D-pantothenol in vivo continues longer than of D-pantothenic acid. On the contrary, the amount of pantothenic acid excreted in the urine within 24 hours after the oral administration of D-pantothenol was larger than that of D-pantothenate. This fact may show that the absorption rate of D-pantothenol from the intestine is more rapid than that of the D-pantothenate. Above results suggest that the utilization of D-pantothenol when used as a pharmaceutics in vivo is higher than that of calcium D-pantothenate.

The inhibitory activity of various proteins such as, pepsin, α-casein, whole casein, egg albumin, β-lactoglobulin, gelatin, human serum fibrinogen, upon the oxidation of ascorbic acid was examined by the rate of oxygen consumption with the aid of Warburg manometer. The sequence of them by copper ion is as follows : Pepsin<α-casein<whole casein>egg albumin>β-lactoglobulin. This effect of these proteins decreases when the prothins are denatured by heat. The highest inhibitory activity is observed when the protein whose isoelectric point is approximately 4.6 is used and this activity decreases when the protein whose isoelectric point is either higher or lower than 4.6 is used. The same effect of proteins was observed in the enzymatic oxidation of ascorbic acid by ascorbic acid oxidase.

Investigations were made on the relation between the thiamine content of the medium and the yield of bakers' yeast under and aerobic condition. It was found that the yield of yeast was decreased by the addition of thiamine to the synthetic medium. The removal of thiamine from molasses medium by the adsorption using the acid clay was effective for increasing the yeast crop, but the destruction of thiamine by sodium sulfite was ineffective The addition of KCN was also effective for the elevation of yeast crop.

It is well known that some of the vitamin B_6 enzymes need metal ions and thiol group with pyridoxal phosphate. Taking this in mind, the authors synthesized pyridoxal-DL-dihydro-α-lipoic acid mercaptal hydrochloride and pyridoxal phosphate-DL-dihydro-α-lipoic acid mercaptal from pyridoxal or pyridoxal phosphate and lipoic acid and investigated the physiological activity of them. They had both the vitamin B_6 and lipoic activities which were increased by the acid hydrolysis. The amount of vitamin B_6 and pyridoxal phosphate in the liver increased by an subcutaneous injection on vitamin B_6 deficient rats. These compounds showed some preventing effect on histological changes of liver cells due to CCL_4.

The author studied dehydroascorbic acid metabolism and effect of X-ray radiation on the metabolism by means of the method of quantitative determination of ascorbic acid utilizing strong alkaline colored reaction. Dehydroascorbic acid is hardly found in urine and various organs of normal rabbit and it is rapidly reduced or dissolved in vitro as well as in vivo. The reduction capacity decreased by a minimal amount of X-ray radiation. This decrease is not due to the alarm reaction, but some factor other than SH-radical has relation to it.

A Modification of Stokstad's medium for the determination of lipoic acid was carried out, with which growth of Stc. faecalis 10 Cl was more accelerated than with media reported previously. The method was also detailed which permitted the accurate turbidimetric determination of lipoic acid with Stc. faecalis 10 Cl after a 24 hous incubation period, and which had the assay range approximately 0.25 to 5.0 mμg of lipoic acid per ml. Reliability of this assay method was indicated by reproducibility of assay and by recoveries of lipoic acid added to rabbit urine and yeast extract. Growth stimulating activity of the several derivatives and analogues of lipoic acid were investigated. Methyl DL-dihydroliponate and methyl DL-6,8-diacetylthiooctanoate were found to have about 130 and 100 per cent, respec- tively, of the growth stimulating activity of lipoic acid. Smaller activities were also found for methyl DL-6-chloro-8-mercaptooctanoate and methylDL-6-hydroxy-8-mercaptooctanoate such as 16 and 1.2 per cent.

The growth of Kloeckera apiculata (thiamine-less) was inhibited by deoxythiamine, but not the growth of Saccharomyces carlsbergensis which does not require thiamine for its growth. The inhibition of deoxythiamine for Kl. apiculata was recovered competitively by thiamine, and its inhibition index was 100-150. This compound had no thiamine activity. The order of decreasing antithiamine potency was : methylthiothiamine≒pyrithiamine> deoxythiamine> chlorothiamine> thiamine sulfuric acid ester> oxythiamine.

The biological activities of thiamine and related compounds for a strain (70-23) of Escherichia coli mutant were investigated by estimating the growth response in the range of a nearly minimal concentration. Bioautographically (by paper disc method), the minimal amounts of thiamine and its phosphoric esters for the growth response corresponded to 0.2 mμg of each compounds while those of the disulfide and S-acyl compounds of thiamine were much higher, but both of these compounds gave the same activities in the degree of 10 mμg. Turbidimetrically, the activities of the latter compounds were more labile than the former under a condition of incubation. The response curve for time course of the disulfide and S-acyl compounds showed lower activities for the half maximal growth compared with thiamine, while the maximal growths were equal to thiamine except dibenzoyl thiamine. Therefore it is assumed that these disulfide and S-acyl compounds are utilized for growth of the microorganism after transformation to thiamine or its phosphoric esters.

Aspergillus oryzae No.10 (Higuchi) was cultivated for 30-72 hours at 30℃ in a synthetic medium by surface or shaking culture, and the lipoic acid content of the mycelium was measured by microbioassay method using Streptococcus faecalis 10Cl. No significant difference was observed between the lipoic acid contents of the mycelia obtained by the two different methods of culture. The maximum value, 1μg/g which was almost equal to the contents of yeast or liver was obtained in 72 hours cultivation.

It has been reported previously that the thiamine content of mash in the process of alcoholic fermentation was not enough as compared with that of pantotheic acid. In the present study, an attempt to promote the fermention rate by the addition of thiamine was made with a sufficient result that 10μg/g of thiamine added to mash caused 10 hours reduction of the fermentation time.
