The experiments were designed to study the effect of some watersoluble vitamin deficiencies in Wistar strain rats upon the anserine and carnosine contents in gastrocnemius muscle. Male rats (body weight 70〜80g) were divided into 5 groups and fed on each vitamin deficient and control diet for 4 weeks. The anserine, carnosine, protein, DNA, RNA, free N^π-methylhistidine, free histidine, free β-alanine and creatinine contents in the muscle were measured. Body weight and food intake were also measured. By the vitamin B_1 deficiency, an increase of anserine and a remarkable increase of carnosine were observed. By the vitamin B_2 or vitamin B_6 deficiency, no effect on anserine content and a remarkable decrease of carnosine were recognized. And free β-alanine decreased. By the pantothenic acid deficiency, no effects on anserine and carnosine were observed. The free histidine and N^π-methylhistidine levels were not affected by the above vitamin deficiencies.

An overshoot phenomenon in ^3H-pyridoxine (^3H -PIN) uptake was observed with cells of a vitamin B_6 auxotroph (KG980) of Escherichia coli K12,as well as its wild strain. The overshoot was clearly detected with higher ^3H-PIN concentrations, e.g., 10μM in the medium and was most remarkable with the cells harvested in log phase of growth. This phenomenon appeared specific for PIN since little or no overshoot was observed with ^3H-pyridoxal or ^3H-pyridoxamine. ^3H-Pyridoxal phosphate and ^3H-pyridoxamine phosphate were the two major forms of intracellular ^3H-compounds at the time of maximal accumulation and were also the major forms of ^3H-compounds released subsequently into medium. The results have indicated that the phosphate forms of vitamin B_6 can cross E. coli cell membranes from inside to outside.

The relationship between amino acid metabolism and lipid contents in the liver of rats fed a vitamin B_6-deficient diet was studied. A high level of lipid in the liver of male rats fed a 70% casein-vitamin B_6-deficient diet was confirmed in the present paper. Female rats, on the other hand, failed to develop the fatty liver under a similar condition of high-protein-vitamin B_6-deficient diet. The degree of lipid accumulation in the liver of rats under various conditions of vitamin B_6-deficiency was found to be correlated with the degree of abnormal accumulation of free citrulline, serine and threonine in the liver.

^3H-Pyridoxal 5'-phosphate (^3H-PALP) was produced by addition of ^3H-pyridoxine (^3H-PIN) to growing cultures of Escherichia coli and was readily isolated by chromatography on columns of aminoethyl-cellulose and aminoethyl-Sephadex. The preparation obtained appeared practically pure from its several properties including the coenzyme activity and the behavior on ion exchange chromatography. The yield of ^3H-PALP with respect to the tritium label of added ^3H-PIN was around 50% under the conditions employed. Specific radioactivity of the product was about 90% of that of ^3H-PIN when a vitamin B_6 auxotroph (KG980) of E.coli K12 was used and was about 70% when a wild strain, E.coli K12 was used.

In an attempt to develop a sensitive and highly reproducible method for the determination of individual tocopherols in red blood cells (RBC), high-speed liquid chromatography (HSLC) was investigated. RBC suspension was saponified after addition of tocol as an internal standard, and extracted with n-hexane. The extract was subjected to HSLC using JASCOPACK WC-03-500 column, and isopropylether・n-hexane mixture as mobile phase. A combination of saponification technique and HSLC made it possible to separate individual tocopherols and the fluorometric detection prevented the influence of interfering substances. This method gave almost 100% recovery of tocopherols, therefore it was not necessary to be corrected based on the recovery of the added radioactive tocopherols. Using this method, the tocopherols levels of plasma and RBC were determined for some human subjects, mice, rabbits, guinea pigs and rats, and for rats feeding with vitamin E-supplemented or vitamin E-deficient diet. Tocopherol ratio of RBC to plasma was different in species, but relatively constant within a same species.
