The Proteins relating to the Physiological Function of Vitamin D_3

The action of vitamin D_3 was discussed from the role of the specific binding proteins (SBP). SBP for vitamin D_3 metabolites in rat intestinal mucosa was analyzed by Sephadex G-200 and sucrose density gradient ultracentrifugation. Sedimentation constant of the binding protein for 25-hydroxycholecalciferol and 1,25-dihydroxycholecalciferol was 6,3S and 5,3S respectively. Calcium binding protein (CaBP) was analyzed by Sephadex G-100,DEAE-cellulose and sucrose density gradient ultracentrifugation. Three sorts of CaBP in rat intestinal mucosa were indentified as those of molecular weights ; >100,000,24,000〜27,000 and 13,000. Sterol carrier protein was also studied. The binding of 7-dehydrocholesterol to this protein was inhibited by vitamin D_3. From these results, the protein will participate in the intracellular movement of vitamin D_3 metabolites and physiological function of vitamin D_3.

2', 3'-Di-o-nicotinoyl riboflavin-4'(5')-phosphate (DNRP) caused a pharmacological action at relatively large dose. (1) Acute toxity of DNRP was studied in mice and rats (see TABLE 1). (2) DNRP decreased spontaneous motor activity in mouse (200〜300mg/kg, i.p.), rat (200〜300mg/kg, i.p.), and dog (50〜100mg/kg, i.v.). (3) DNRP (50mg/kg, s.c.) prolonged the sleeping time of pentobarbital sodium in mouse (40mg/kg, i.p.), but did not show analgesic effect. (4) DNRP (0.15〜0.3%) showed the more effective local anesthetic action than procaine did. (5) DNRP (50mg/kg, s.c.) inhibited the passage of charcoal meal through intestine. (6) DNRP decreased the tonus of isolated rabbit intestine and inhibited the spontaneous movement of isolated rat uterus. (7) DNRP depressed (nonspecifically) the contractive response of guinea pig ileum and rat uterus induced by acetylcholine, histamine, serotonine (5HT), BaCl_2,angiotensin and bradykinin. (8) DNRP potentiated the contractive response of isolated guinea pig vas deferens induced by tyramine, ephedrine, norepinephrine, epinephrine and phenylephrine.

Involvement of Ca^<2+>ion in the formation of macromolecular complex of vitamin B_<12>, intrinsic factor and receptor prepared from rat ileum mucosa was investigated by using ^<45>Ca ion and ^<57>Co-B_<12>. Filtration of the complex through Biogel column revealed that the eluted position of ^<57>Co coincided with that of ^<45>Ca. By the treatment of this complex with EDTA, the position of ^<45>Ca in the eluent shifted to that of free ^<45>Ca ion. These results indicate that ^<45>Ca is bound to this complex and plays an important role in the formation of B_<12>-intrinsic factor-receptor complex.

The present study was undertaken to confirm the curative effect of O-benzoylthiamine disulfide (BTDS) on diabetic neuropathy. BTDS of 30 mg (A group), 75 mg (B group) or 150 mg (C group) as a daily dose, was administered by means of completely randamized double blind method to 116 diabetics with neuropathy for six weeks. Changes of the following symptoms and signs were checked every week, i.e. sensory disturbances, spontaneous pain, other neurological symptoms including visceral neuropathies, headache, general malasia, thirst sensation, knee jerk, ankle jerk and calf tenderness, and the effect was compared among the three groups. The result showed that the curative effect on sensory disturbances, pain and other neurological symptoms, assessed by the weeks needed for the improvement or by the percentage of effective cases, was significantly greater in the B and C groups than the A group. However, the effectiveness on the symptoms was not significantly different between the B group and the C group, except other neurological symptoms. This study indicates that BTDS is effective for diabetic neuropathy and that 75 mg of BTDS is necessary to get the sufficient effect and 30 mg is not an enough dose for it.
