In order to establish pyridoxine as a precursor of the pyrimidine moiety of thiamin in Saccharomyces cerevisiae, labeled pyridoxines, [^<15>N]-, [2'-^<13>C]- and [6-^<13>C]pyridoxines were chemically synthesized with alanine, formic acid and ethyl maleate via oxazole derivative. [^<15>N]-, [2'-^<13>C]- and [6-^<13>C]Pyridoxines were labeled with [^<15>N]-, [3-^<13>C]alanine and [^<13>C]formic acid, respectively. [5'-^2H_2]Pyridoxine was synthesized by the reduction of isopropylidene pyridoxic acid methyl ester with LiAID_4 followed by hydrolysis.

The plasma concentration-time profile and urinary excretion of vitamin C after the oral administration of a single dose (1000 or 2000 mg) was examined using six healthy males (25-29 years) in a two-period crossover trial. The mean T_<max> (h) was 3 for both 1000 mg and 2000 mg. The mean C_<max> (mg/100 ml) was 1.13 for 1000 mg and 1.55 for 2000 mg (based on concentration above the initial value). The mean AUC (mg.g/100 ml, 0-24 h) was 11.16 for 1000 mg and 17.12 for 2000 mg. The plasma concentration-time profile could be simulated by a one-compartment model with first-order absorption. Ka (h^<-1>) were 0.31-0.33 and Ke (h^<-1>) were 0.31-0.33. V/F (liter) was 35 for 1000 mg and 49 for 2000 mg. The mean amount (mg, above the initial value) excreted in the 24-h urine was 269 for 1000 mg and 362 for 2000 mg. The percentage of the dose excreted was 27% for 1000 mg and 18% for 2000 mg.

The changes of hepatic lysosomal cathepsins, alanine aminotransferase (AAT) activities and serum amino acids were studied on rat fed on high and low protein diets with or without B_6. The group fed on high protein diet without B_6 caused the marked suppression of growth and also in the pathological bad lie of hair with dermatosis, while these pathological syndromes were not observed in rats fed on low protein diet without B_6. The activities of cathepsins B, L, H and J in rats fed on high protein diet these cathepsin activities were not affected by B_6 deficiency. Therefore, the biosynthesis and also degradation of these cathepsins are not influenced by B_6 deficiency. Some amino acid levels in serum were increased by high protein diet intake and B_6 deficiency. The levels of glycine were significantly decreased in rat fed on high protein diet. The levels of aspartate and glycine were markedly elevated in the cases of B_6 deficiency and those of alanine and hydroxyproline were decreased in the cases of B_6 deficiency.
