Effects of intraperitoneal injection of reserpine on the levels of B_6 vitamers in mouse brain were examined. Injection of reserpine alone resulted in higher level of pyridoxal phosphate (PALP) and in lower level of pyridoxal (PAL), but did not influence on the levels of other vitamers. Intramuscular injection of pyridoxine (PIN) or pyridoxamine (PAM) after the injection of reserpine resulted in higher level of pyridoxine phosphate (PINP) or pyridoxamine phosphate (PAMP), respectively, but did not elevate to higher level of PALP than that of reserpine alone. Injection of PAL after the reserpine injection resulted rather in a decrease of PALP. Injection of PIN, PAL or PAM alone resulted in an unexpected decrease of PALP, though PIN and PAM elevated slightly the levels of PINP and PAMP, respectively.

Ornithine aminotransferase (EC 2.6.1.13) was purified from baker's yeast (Saccharomyces cerevisiae) with following steps : aceton powder, crude extract, nucleic. acid digestion by ribonuclease T_1,protamine sulfate treatment, heat treatment, ammonium sulfate fractionation and DEAE-Sephadex column chromatography. Finally the specific activity increased 10-fold times and about 7% of the enzyme activity was recovered. The enzyme was unstable and showed the optimum pH at 7.0 to 7.5. Km values determined kinetically were as follows ; for ornithine, 1.2〜2.5×10^<-3>M and for α-ketoglutarate, 6.7×10^<-3>M. Branched chain amino acids were weakly inhibitory and K_i value for L-valine was 4.3×10^<-3>M. These properties were compared with those of rat liver enzyme.

One and half hours after feeding of ^<57>Co-vitamin B_<12> to rats, small intestines were extirpated. Using Weiser's method, gradient isolation of the intestinal epithelial cells from villus to crypt areas were carried out and five different levels of the villus and crypt areas were obtained. The uptakes of ^<57>Co-vitamin B_<12> and the formations of Receptor-Intrinsic factor-^<57>Co-vitamin B_<12> complex were measured, indicating that the upper villus cells have a higher uptake of ^<57>Co-vitamin B_<12> and more increased formation of it than crypt cells do showing a gradient from crypt to villus. Alkaline phosphatase activity of these levels of epithelial cells also showed the same pattern. These results suggest that the receptor activity for vitamin B_<12> absorption in the intestinal epithelial cells indicate a gradient increase from crypt to villus areas.

Antivitamin B_6(anti-B_6) activity of α^4-norpyridoxine (4-NOP) was tested using microorganisms and rat. This compound was proved to be a potent vitamin B_6 antagonist in the growth of Saccharomyces carlsbergensis, Leuconostoc mesenteroides and Kloeckela apiculata, and the inhibitory activity in the test organisms was comparable to that of 4-deoxypyridoxine. The inhibition indexes for S. carlsbergensis were 125,77 and 550 against pyridoxal, pyridoxamine and pyridoxine (PIN), respectively. In contrast, the compound showed no anti-B_6 activity for vitamin B_6 deficient rat growth when orally administered at the ratio of 4-NOP to PIN, 1,000 to 1. The anti-B_6 activity of several other 4-substituted PIN analogs was also examined using S.carlsbergensis.

Thiobarbituric acid (TBA) value of rat liver homogenate was studied using sodium dodecylsulphate (SDS) as a tissue solubilizing agent and the following results were obtained. 1. TBA value of rat liver homogenate increased by SDS treatment. TBA positive substance was suggested to be localized in tissue membrane. 2. Vitamin E decreased TBA value in vivo and in vitro, suggesting that this TBA positive substance seemed to be a polyunsaturated fatty acid radical species. 3. Alloxan and carbon tetrachloride raised liver TBA value. 4. Vitamin E scavenged TBA positive radical species which led to two moles of malondialdehyde by degradation.
