    It is well known that pantothenate stimulates the growth rate of yeast. Concerning with pantothenol, Snell discovered that the compound may response to the microorganism as a competitive inhibitor. Afterwards, Bird repeated the studies on the inhibitive properties of D-pantothenol in the microbioassy of pantothenate by Leuconostoc mesenteroides, and it was found that the microbial determination of pantothenol may be possible using the same strain. Recently, the present authors have attempted further investigation for the competitive ratio between both compounds, and it was realized that there is the competitive ratio of 200. Finally, it was demonstrated that the estimation of D-pantothenol can be successfully completed in the range of 0.5〜1.0 μg/2ml D-pantothenol.

    In previous paper, the authors reported the chemical synthesis of vitamin D_2-sulfate and its properties and confirmed that the vitamin D_2-sulfate isolated from rabbit urine, supplemented with vitamin D_2 was coincided with the synthetic one. Afterwards, further investigations have been continued in our laboratories in regard to the biogenesis of the compound. The abilities of vitamin D_2-sulfate biogenesis were examined with various organs of rat and it was found that vitamin D_2-sulfokinase is distributed in stomach, colon and small intestine, especially in liver. The crude enzymic solution was prepared from rat liver homogenate by the salting out with ammonium sulfate. The formation of vitamin D_2-sulfate by the enzymic solution from vitamin D_2 and K_2SO_4 was recognized by Al_2O_3-impregnated paper- and thin-layer chromatographies and it was confirmed by the use of K_2^<35>SO_4. The maximum biogenesis was recognized at the two hours incubation with 0.04 μmole per 0.1ml of vitamin D.

    Large dosage of dietary myoinositol seemed to be inhibitory on the growth of young rats (about 20 days after birth) ; it was found that the body weight increase of young animals fed on a large excess of myoinositol (1g/kg/day) was significantly lower than those fed on the standard diet containing myoinositol (10 mg/kg/day), and even lower than those fed on the inositol deficient diet. However, such an inhibitory action was not seen in the case of older rats (3-4 months after birth) fed on large amounts of myoinositol (5g/kg/day). Concerning these findings, the changes in the pattern of lipids in the liver were investigated, and it was found that the larger the dosage of dietary myoinositol, the larger the P/N ratio of hepatic lipids, although there was no significant changes in the relative amounts of the total lipids and cholesterol in the liver. The result suggested the possibility that the dietary myoinositol had some effects on phospholipid metabolism in the liver, in particular of the young rats. Relating to such changes in liver lipid patterns, discussions were made on the mechanism of the growth-depressing action of myoinositol when it was supplied to the young animals in a large excess amount.

    A simple colorimetric assay method of methylmethionine sulfonium derivatives (MMSD) in pharmaceutical preparation was established using potassium 1,2-naphthoquinone-4-sulfonate as a coloring reagent. MMSD was purified as phosphotungstate from its decomposition products and other impurities. The presence of borate as a component of buffer solution in reaction mixture had advantages for the lowering the blank value and for the improvement of the recovery rate.

    In order to obtain the information on the precursor of the pyrimidine moiety of thiamine, the effect of amino acids and some other compounds on the production of thiamine of thiamine requiring mutant of E. coli (thiazole-less, 26-43) was investigated. The thiamine synthesis per growth of the mutant in culture media reduced with glucose and nitrogen sources are presumed to depend on the synthesis of pyrimidine by addition of the test materials, since the growth could be satisfied by the thiazole supplemented. The production of thiamine in overnight cultivation without shaking was increased by addition of aspartate, glutamate, histidine or oxaloacetate, fumarate and malate, respectively. By incubation of the cell suspensions for 2 hours on a rotatory shaker, thiamine synthesis was stimulated by addition of aspartate while other amino acids gave no significant stimulation. Maximal production was achieved with the combination of aspartate aad alanine. Further addition of pyruvate or fumarate was also effective. Increase of glucose in the medium depressed the thiamine synthesis by the cells. The resting cell expriments gave no significant effect by addition of such stimulators.

    The thiamine production from hydroxymethylpyrimidine (OMP) in the medium containing excess of thiazole by a mutant strain (thiazole-less) of Escherichia coli was proportional to a small amount of OMP (1-10 mμ moles) for a short time incubation. Therefore, the effects of the components of nucleic acid and their precursors on the thiamine production and conversion of these test materials to OMP were investigated using thiazole-less strain of E.coli. No other compounds than 2-methy-4-aminopyrimidine derivatives strictly similar to the structure of OMP gave significant effect on the thiamine synthesis while orotate and uridine had a little effect. Further, the conditions of OMP determination by baker's yeast suspensions containing glucose and thiazole was obtained and the conversion of purine derivatives to OMP base on the OMP determination method was tested by cell free extracts of E.coli. No any positive result was obtained.

    The incorporation of radioactive carbon from DL-aspartate-2-^<14>C and Na-formate-^<14>C into the pyrimidine moiety of thiamine was investigated by isolation of thiamine produced by a mutant strain (thiazole-less, 26-43) of Escherichia coli growing in culture media supplemented with thiazole. IRC-50 column chromatography, conversion of thiamine to thiochrome, repeated paper chromatography and actigraphy were devised for the separation and isolation of thiamine in the tracer studies. The incorporation of radioactive carbon from the both ^<14>C-compounds was demonstrated by detection of radioactivity corresponding to the fluorescent spot of thiochrome on paperchromatogram. From the comparison of specific radioactivities of thiochrome isolated and those of ^<14>C-compounds added, aspartate-2-^<14>C is incorporated to a better extent than formate-^<14>C.

    In previous papers, the authors studied on the absorption of thiamine propyldisulfide. In this paper, comparison was made between thiamine tetrahydrofurfuryldisulfide and thiamine propyldisulfide labeled with ^<35>S in thiazole ring on the mode of intestinal absorption, the tissue intake, and the urinary metabolites after the oral administration to rat. No difference was found between them in the amount of ^<35>S in urine and various organs and also in the rate of absorption from ligated intestinal canals. As the metabolites of ^<35>S in urine, five main substances, tentatively named as F_1-F_5 besides thiamine were recognized by DEAE-Sephadex fractionation. Among them, F_3-F_5 were considered similar to X-substances, which we have previously reported as the urinary metabolites of thiamine and thiamine propyldisulfide.

    The fate of tetrahydrofurfurylsulfide-^<35>S group of thiamine tetrahydrofurfuryldisulfide-^<35>S in rat was investigated. The side chain of the thiamine derivative was considered to be separated from thiamine molecule by reducing substances in blood after the absorption from the intestinal canals, and then excreted in urine as its metabolized products. Six metabolites were found in the rat urine by paper chromatography and paper electrophoresis. One of them was sulfate-^<35>S (about 30% of admintstered ^<35>S) and the others (about 70%) were soluble in ethanol or butanol but insoluble in benzene. It is considered that part of the metabolites combined with albumin in blood.

    When riboflavin-enriched noodle was treated by mixing alcoholic solution of riboflavin tetrabutylate during the preparation, 60% of the vitamin remained after the boiling. The value was far higher than that of the noodle enriched by ordinary riboflavin. The value was further elevated up to 80% by containging riboflavin tetrabutylate in the inner part of the noodle.
