Kynureninase activity was investigated in the livers biopsied from the patients with B6 dependent xanthurenic aciduria. The activity was found to be markedly reduced but it was restored to a considerable extent by the addition of' an excess of pyridoxal phosphate to the assay system. This suggests that the basic defect in B_6 dependency is an inability of apoenzyme to combine with the coenzyme. Glycine cleavage system was investigated in the livers and brains autopsied from patients with nonketotic hyperglycinemia. The overall activity of the glycine cleavage system was found to be extremely low in both the liver and brain. A-nalyses of four protein components involved in glycine cleavage system revealed that nonketotic hyperglycinemia is caused by a specific defect in either pyridoxal phosphate-dependent protein or tetrahydrofolate-requiring protein. A duble-stranded RNA phage, φ6, has been found to be inactivated by ascorbic acid (ASA), and the reaction mechanism of inactivation of the phage by ASA has been studied. The addition of hydrogen peroxide or cupric ion into the reaction mixture enhanced the inactivation. In contrast, removal of oxygen from the reaction mixture, and the addition of reducing agents, chelating agents or general radical scavengers into the reaction mixture prevented the inactivation. Dehydroascorbic acid or hydrogen peroxide had no direct effect on the phage. The results indicate that the free radical intermediates produced during the course of the autoxidation of ASA are responsible for the inactivation of the phage; free radical mechanisms are involved in the inactivation reaction. The experiments using specific radical scavengers indicate that ・O_2, ・ OH and 1^O_2 are involved in the inactivation reaction, and that ・OH would be mainly responsible for the inactivating effect on the phage. The effect of vitamin D3 and dietary calciurn on the cadmium-induced morpho-10gical changes in rat intestinal epithelial cells were examined by a transmission electron microscopy. Observation with transmission electron microscopy showed significant changes in cadmium-exposed rats, especially those raised on a vitamin D deflcient diet. Swelling and destruction of mitochondria, sparseness and exfoliation of microvilli, disappearance of terminal web were observed. Rough surfaced endoplasmic reticulum and Golgi apparcrtus were enlarged. Furthermore, many vacuoles were observed in epithelial cells.
