A series of studies by the author and his co-workers on the chemical properties and behaviors of cobalamins are summarized together with the advances in elucidation of their biochemical functions. The informations and suggestions obtained are mainly concerned with the cleavage of Co-C bond of Coenzyme B_<12> analogs and the consequent formation of reduced forms of cobalamin as well as the behaviors of the reduced forms including the formation of Co-C bond. The results are discussed in view of their contribution to the understanding of coenzyme actions of cobalamins.

The availability of Florex XXS as a chromatographic adsorbent to the determination of vitamin D was reexamined. When samples containing vitamin A and D (the international unit of vitamin A was not more three times than that of vitamin D) were applied to the column of Florex XXS weakened by mixing with 7 % of water, vitamin D was satisfactorily separated from vitamin A alcohol and its degradation products. In order to confirm on the availability of Wilkie's correction factor (0.67) to the nonvitamin D substances, the benzene eluate obtained by applying vitamin A alcohol alone to the column was reacted with antimony trichloride - acetyl chloride reagent. The factor was found to be available when the amount of non-vitamin D substances was not so much, whereas it was unavailable when vitamin A alcohol and its degradation products were not sufficiently separated from vitamin D.

As for the nonenzymatic reaction of thiamine-S-sulfonate (B_1SSO_3H) and cysteine-S-sulfonate (CySSO_3H) with GSH, the apparent second order rate constants were estimated under physiological conditions. It was revealed that the mixed disulfide forming reaction (1) was the rate limitting step in the case of B_1SSO_3H. B_1SSO_3H(CySSO_3H) + GSH→B_1SSG^*(CySSG)^<**> +H_2SO_3 (1) B_1SSG(CySSG) + GSH→B_1SH(CySH) +GSSG (2) ^*thiamine-glutathione disulfide ^<**>cysteine-glutathione disulfide An enzyme catalizing these reactions (1,2) was demonstrated in the supernatant fraction of rat liver extracts. This GSH-dependent enzyme was supposed to be different from the previously reported GSH-B_1 derivatives transhydrogenase. Mechanisms of the conversion of B_1SSO_3H to thiamine and of CySSO_3H to cysteine were discussed.

The effect of daily administration of the vitamin B mixture on the functional recovery from the neuropathy induced by the chronic administration of acrylamide and on the regeneration of crushed sciatic nerve in the rat were investisated. The abnormal motor functions were observed by the administration of acrylamide and the vitamin B complex suppressed the lethality and restored the abnormal functions. The mixture of thiamine disulfide (TDS), PIN and aquocobalamin (OH-B_<12>)[3B]was more effective than each component. The further investigation of 3B was carried out. The decrease in the conduction velocity of the nerve resulting from the neuropathy induced by acrylamide was observed, but no decrease was shown in the rat received with 3B and the decreased velocity after the withdrawal of acrylamide was recovered by 3B. The effect of 3B on the electromyogram patterns of the crushed sciatic nerve in the rat was also investigated. 3B prompted the reinnervation of the crushed peripheral nerves. These results suggest that 3B has some favourable functions on the neural regeneration and the functional restoration from the neuropathy.

The effect of 2', 3'-di-o-nicotinoyl riboflavin-4'(5')-phosphate (DNRP) on the hypercholesterolemia in mice, rats, chicks and rabbits induces by feeding a cholesterol containing diet was studied. The following results were obtained. 1. The elevation of plasma cholesterol level in mice being fed a cholesterol containing diet was prevented in s.c. administration of 100 and 200 mg/kg/day of DNRP. 2. The elevation of plasma and liver cholesterol levels in rats was significantly prevented in s.c. administration of 60,120 and 120 mg/kg/day of DNRP, respectively. 3. The elevation of plasma cholesterol level in chicks was markedly accelerated at the first week, but not altered for the following 4 weeks by i.m. administration of 100 mg/kg/day of DNRP. The elevation of liver cholesterol level was also prevented in same dose of DNRP. 4. The elevation of plasma and liver cholesterol levels in rabbits was markedly reduced by s.c. or p.o. administration of 120 mg/kg/day of DNRP.
